Nursing Care Particularities in Children Admitted to Hospital with Autism
SCHÖNBAUEROVÁ, Andrea
The topic of the thesis is the Nursing Care Particularities in Children Admitted to Hospital with Autism. In the theoretical part is found the description of autism causes, clinical manifestation, diagnostics, basic therapeutic measures and nursing care in such children. The thesis has set three goals: Goal 1- finding out problems concerning the hospital admittance of an autistic child. Goal 2- determining the specific features of nursing care in children with autism. Goal 3- creating an informative brochure that would help nurses to gain basic information necessary to provide expert nursing care to autistic children. To gain these goals, a method of qualitative research and the technique of standardized interviews were used. In the frame of the research, two research samples were used. The first one consisted of nurses who work in children´s psychiatric wards and children´s neurologic wards. These nurses were chosen deliberately as they care about autistic children regularly and have a lot of experience in this branch. Altogether ten interviews were conducted and were recorded on the record keeping sheet and consequently processed. The other research sample were mothers of children in whom autism was diagnosed and who experienced hospital admittance either as a diagnostic stay or in connection with acute treatment. Also here ten interviews were conducted. After the completion of qualitative research answers to the set research questions were gained: Research question 1 finds out what are the specific features of nursing care in children with autism. On the basis of the research, it was concluded that nursing care in autistic children has some specific features such as communication, observing rituals, the necessity of a parent´s company, as well as the psychical burden both of children and staff. Research question 2 focused on finding what problems are connected with the hospital admittance of an autistic child. On the basis of the conducted interviews, it was concluded that the biggest problems concerning hospital admittance apply to children´s aggressivity, difficult adaptability to environment changes and problems that occur after the experienced admittance. Further on, it was concluded that also children´s individuality is a serious problem that has to be taken into consideration during nursing care. Research question 3 found out how nurses assess parent´s presence during the admittance of autistic children. All nurses perceive the parent´s presence highly positively. They see the most benefits in passing important information to nurses, accompanying the child during the whole period of admittance, helping with the adaptation to a new environment, providing the common daily care including spending free time and giving the child the feeling of safety and assurance. Research question 4 deals with nurses´ feelings when nursing autistic children. The provided information is individual. The research has found that nurses feel well, do not see problems and the work is routine for them. Negative feelings are influenced by circumstances concerning mood, tiredness or demanding communication with parents. Further, it was concluded that nurses feel insecurity in the period when they start working with autistic children or during the first contact with the child. With regard to high disorder prevalence in the population, it is essential for nurses to know basic information concerning autism and adopt the basis of nursing care in children with this disability type. In this connection, an information material was created that has to help nurses to acquire basic information necessary to provide expert care to autistic children.

Possibilities of flow cytometry in analysis of cellular response to genotoxic stress
Havelek, Radim ; Řezáčová, Martina (advisor) ; Bílková, Zuzana (referee) ; Štětina, Rudolf (referee)
Possibilities of flow cytometry in analysis of cellular response to genotoxic stress Flow cytometry is a modern tool for interrogating the immunophenotype and functional characteristics of cells. It is a technique of quantitative single cell analysis that works by sensing optical properties of cells in a flow stream with laser beam. Analysis and differentiation of the cells is based on size, granularity, and whether the cell is carrying fluorescent molecules in the form of either conjugated antibodies or dyes. The cells may be stained with fluorescent dyes, incubated with fluorogenic substrate or labelled with fluorochrome-linked antibodies specific for molecules either on the surface or in the intracellular components of the cell. Flow cytometers are multiparameter, recording several measurements on each cell. Therefore, it is possible to identify a homogeneous subpopulation within a heterogeneous population of cells. When cells are exposed to genotoxic agent, they respond quantitatively and qualitatively according to the absorbed dose and the cell type. To cope with the resulting damage to cellular DNA, the temporary cell-cycle checkpoints and DNA damage response mechanisms are activated to allow more time for effective repair. However, if these mechanisms fail or the damage is irreparable, then...

Bártů, Václava ; Sechser, Tomáš (advisor) ; Homolka, Jiří (referee) ; Marešová, Vilma (referee)
Background. Respiratory tract infections belong among the most frequent inflammatory affections. They are mostly cured as outpatients. Cases with complicated or serious course have to be hospitalized. The aim of the projeet was to evaluate the procedures of physicians of pulmonary department in the diagnosis and therapy of respiratory infections including community acquired pneumonia of inpatients. The drug prescription and its adherence to the positive list were evaluated. Clinical effectivity of therapy was evaluated according to the acquired data and cost effectivity was followed up. Methods. The projeet included 200 patients who were examined and treated according recommended standard guidelines as "usually". All diagnostic and therapeutic activity depends only on the decision of the physician without any intervention. As the patient was accepted and received antibiotics, so immediately he entered the study. The projeet protocol consisted of usual questions on which physician had to response during his deciding diagnostic and therapeutic action. The projeet had non-intervention character. The results of all decisions and examinations were recorded so to the documents of patient as to the protocol of the projeet and at the same moment to the PATS in the order not to be changed. Conclusions. Data analysis...

The Exploitation of Functional Capacity of Robotic Milking Machines in System of Free Moving Animals.
REICHOVÁ, Sandra
The aim of the thesis was to assess objectively the exploitation of the functional capacity of milking machines in the system of free moving animals. There were data coming from seven agricultural companies analysed in the thesis. The data collection took place from January to November 2009. We were provided with the preliminary data by the individual farms taking part in a programme called T4C. The information on problematic dairy cows comes directly from their breeders. The average production of the dairy cows was the first assessed criterion. The highest production (28.79kg) was achieved by little private agricultural companies. The lowest production (25.22kg) was ascertained in middle-sized companies. As far as the breed of dairy cows is concerned, the Holstein dairy cows gave the highest possible amount of milk (40.43; 30.16 a 27.01 kg). The CRV Fleckvieh cattle dairy cows gave the lowest possible amount of milk (24.83; 21.04 a 22.74 kg). The assessment of the number of milking by means of the robotic milking system represents the next criterion. Little private agricultural companies showed the highest frequency of milking (140.88). Whereas the big agricultural companies showed the lowest frequency of milking (119.28). Mostly the Holstein dairy cows were milked by the robotic milking machines (130.34). On the other hand, the CRV Fleckvieh cattle dairy cows were milked least by the robotic milking machines by contrast (107.94). The middle-sized agricultural companies achieved the highest number of milking per dairy cow, per day (2.47). The little private agricultural companies proved the lowest number of milking per dairy cow (2.34). The mixed herds of the Holsteins and CRV Fleckviehs proved the rate of 2.47. The Holstein dairy cows showed the milking frequency 2.45 per day while the CRV Fleckvieh dairy cows 2.32. The dairy cows from the big agricultural companies went to be milked by the robotic milking machine most frequently from the point of view of willingness, what follows is that these dairy cows showed the highest number of refusals per one milking (2.19). The lowest values of this criterion were shown in little private agricultural companies (1.10). With reference to the breed, the values of this indicator proved the lowest number of refusals (1.85) whereas the CRV Fleckviehs showed the highest number of refusals (2.25). The exploitation of the time capacity has been proven as the most effective one - 78.61% in little private agricultural companies. Whereas the lowest time exploitation has been proven in big agricultural companies - 68.11%. As far as the cow breed is concerned, it was ascertained that the Holstein dairy cows were milked longest -73.21%. On the other hand the CRV Fleckviehs were milked shortest - 63.17%. The highest amount of the dairy cows that needed to by accompanied to the robotic milking machine was recorded in big agricultural companies - 20.1%. Remarkably lower number of problematic cows was ascertained in middle-sized agricultural companies - 9.7%. The number of problematic cows in little private agricultural companies proved to be similar to the number in middle-sized agricultural companies 9.3%. It was ascertained that the most problematic dairy cows came from the mixed herds - 18.7%. The CRV Fleckviehs were the least problematic - 8.8%.
