Photosynthetic electron transport in purple bacteria: An in vivo spectroscopic study

Electron transport in purple bacteria was studied using combination of absorption spectroscopy and induced bacteriochlorophyll fluorescence in whole cells in vivo. Focus is placed on relations between fluorescence yield, the state of the electron transport chain and the membrane potential. A laboratory-built absorption spectrophotometer-fluorimeter is described.
Effect of selected exogenous and endogenous factors on transformation efficiency

The main goal of this bachalor thesis was to explore the possibility to use an ultrasound sonication as a way to increase the transformation efficiency of flax. Another part of this thesis is devoted to the searching for the seed sterilization method and finding an optimal concentration of kanamycin antibiotic useful for the selection of transformed plants.
Possibilities of the green fluorescent protein signal gene utilization for flax transformants identification and selection

An aim of the thesis was to verify possibilites of in vitro identification and selection of transformed flax tissues and plants based on the fluorescence assessment of objects transformed with a construct bearing the gfp gene coding for green fluorescent protein (GFP). Effect of plant genotype, explant type and modifications of the transformation protocol on fluorescence was evaluated too, as well as the possibility of use for selection of transgenic individuals.
On-line quantification of cyanobacteria in a raw water

A new fluorometer AOM (Algae Online Monitor) for the quantification of phytoplankton in raw water was tested on different localities. The AOM was serviceable in the variable field conditons. Because of low limit of detection and due the work in continuous mode with remote GSM data transfer to laboratory, the device is suitable for early warning system.

This contribution is devoted to quantum dots usage for cell imaging in fluorescence microscopy. Fluorescence excitation by evanescence wave (caused by total internal reflection) is compared with conventional epifluorescence microscopy.

A series of modified nucleosides bearing the fluorescent substituted biaryl labels attached to nucleobase was prepared and their luminescent properties were evaluated. The modified nucleosides show strong fluorescence at ca. 400 nm when excited at 310 nm in water depending on substitution pattern.

The fluorescence kinetic microscopy (FKM) provides new dimension in application of chlorophyll fluorescence techniques. It combines classical fluorescence microscopy with 2D kinetic detection systems. FKM allows to examine photosynthesis of individual algal cells or on the cellular level of macroscopic structures. FKM is well suited to study heterogeneity in photosynthetic response among individual cells in algal cultures or in natural mixed populations. Special microscopic chambers allow to maintain live cells under optimal conditions for several hours or days and to follow evolution of photosynthesis in individual cells during the course of their growth cycle.
