Awareness of cutaneous melanoma prevention among students of the University of South Bohemia in České Budějovice

The cutaneous melanoma is a very aggressive cancer disease which could be effectively prevented, even though the incidence of this disease is still high. For prevention, it is important to know about its possibilities and methods and to follow them. The aim of this work is to reveal the awareness of students of the University of South Bohemia in České Budějovice about the melanoma and its prevention. Moreover, this work focuses on the differences of awareness among students of Faculty of Health and Social Sciences and students of Faculty of Education. To fulfil the hypothesis and aims, I made a nonstandard electronic survey. All questions and offered answers are based on the theoretical part of this thesis where a few current pieces of research and their results are mentioned as well. The quantitative research was later evaluated by statistic methods, specifically one-sample and two-sample t-test. According to the quantitative research, there is conclusion that students are aware of the basic prevention of melanoma of the skin. The total average number is 21,7 from 30, which means that 72,3% of students were successful. At the same time, this thesis has confirmed that there is a statistic difference between the two compared units. The students of Faculty of Health and Social Sciences are much informed than the students of Faculty of Education that were successful in 70,6% in contrast to the 74% of the success of students of Faculty of Health and Social Sciences. Based on the data it is possible to conclude that the awareness about this issue is sufficient, however, the further education is needed especially in the case of the students of Faculty of Health and Social Sciences, for whom this topic is much closer.
Imunoterapie melanomu a pankreatického adenokarcinomu na myším modelu

The aim of this thesis is focused on possibilities of cancer immunotherapy which was studied in mouse B16-F10 melanoma model and in mouse Panc02 pancreatic adenocarcinoma model. We compared the effect of various compounds stimulating both innate and adaptive immunity using in vivo and in vitro experiments. The study also monitors incidence of metastasis.

This study examines the role of innate and adaptive immunity in the immunotherapy based on the combination of the ligands stimulating phagocytosis anchored in the tumour cells membrane and the mixture of TLR agonists. This immunotherapy is primarily focused on the innate immunity activation and induces strong inflammatory infiltration, which neutrophils and NK cells are part of. Therefore, the next aim of this study was to evaluate the anti-tumour activity of neutrophils and NK cells. For examination malignant melanoma and pancreatic adenocarcinoma mouse tumour models were used.
Relationship between the coat colour and melanoma occurrence in horses

The literary review is divided into two main parts, the first part is dealing with the coloring of horses and their genetic base and the second part is dealing with melanoma formations of horses and their division. In the chapter about several color types of horses there is a description of forming basic color types which is done by three locuses: COLOR, EXTENSION and AGOUTI. Locus COLOR (CREAM) controls whether or not to create pigment. EXTENSION locus controls about the type of pigment and AGOUTI locus controls the distribution of pigment. The next subsection gives a breakdown of the white color in horses. Greying proces is related with locus GREY where there is a loss of pigment and the coat itself. This process and mutation is commonly connected with dermal melanoma. Non-pigmented spots called vtiligos are connected with the greying process, which is more intense with increasing age. Vtiligios are commonly found in grey horses, at the same locations as dermal melanoma. The places are perianal area under the tail, around the vulva, lips, eyes and around the penis. Melanoma formations are divided according to whether they occur in Grey horses or other colored horses and whether they have a benign or malignant potential. A further breakdown of melanomas at grey horses is whether it is a separate, smaller segments or large, associated lesions, when this condition is called dermal melanomatosis. At the end of the thesis there is a description of the treatment options for melanoma. The most effective appears to be a surgical removal of smaller formations. There were studies which have examined the anticancer action of the drug cimetidine but the results are not yet sufficiently conclusive. As preventing the occurrence of melanoma in grey horses it is appropriate to implement the necessary measures and individuals who are concerned should not be included in the next breeding.
The Effects of the UV Radiation on the Human Body

Work maps tailored health students from the harmful effects of UV radiation and traces their knowledge about the effects of UV radiation on the body. The cohort consisted of students and combined form of Health and Social Studies University of South Bohemia in Ceske Budejovice. The aim of this work is to identify students' knowledge of ZSF JU. The effects of UV radiation on the human body and mapping the extent of protection of health of students of the ZSF JU from the harmful effects of UV radiation. The theoretical part describes the general characteristics of UV radiation and its effects on the human body. Work is more geared to actual problems of skin tumors, mainly the issues already mentioned malignant melanoma, which is currently the worldwide especially among the white population serious health problem. In this work he received a lot of attention because among all tumors occupies an important place mainly due to the increase in the number of cases in recent years, due to the very difficult to estimate prognosis and often very rapid pace. The theoretical part deals with the issues of prevention and alone and prevention programs that are reticent part of health care , and that can detect the disease at a stage when it is still treatable. Has a special position then the international program of the World Health Organization WHO Intersun that assesses and quantifies health risks and develop appropriate responses by issuing various directives, recommendations and dissemination of information, while providing practical advice and information to government health and environmental issues in UV radiation. Practical work logic device monitoring students' knowledge about the positive and negative effects of UV radiation on the body, while also monitoring their attitudes toward tanning , protection from sunlight and behavior in case of suspicious skin changes.
Electron microscopic analysis of processes in progress during cancer immunotherapy based on use of TLR agonists and the ligands of phagocytic receptors

The ligands of phagocytic receptors anchored in the membrane of tumour cells in combination with TLR agonists cause strong neutrophils infiltration within the tumour. These neutrophils are able to direct tumour cell lysis. Ultrastructure analysis of tumour with aim to determine mechanisms of tumour cell damage by neutrophils was performed with transmission electron microscope.

In the first part of the theses, I studied therapeutical effect of intratumoral application of various types of bacteria on melanoma B16-F10 bearing mice alone or in combination with another immunostimulatory compounds. Tumour size, metastasis and survival were monitored. The second part was focused on study of cytotoxic effect of neutrophils on melanoma cells and the role of neutrophil extracellular traps formation.

The aim of this thesis was to study the mechanisms of innate imunity involved in the degradation of tumor cells on which the ligands of phagocytic receptors were installed. For this purpose both in vivo experiments using immunodeficient mice, and in vitro experiments based on monitoring the levels of inflammatory cytokines produced in the tumor tissue and on measuring the level of myeloperoxidase released during neutrophil degranulation were performed.
