Floristicko-ekologická studie sinic a řas v různých typech stojatých vod na severním Plzeňsku

The algological research was realized on 10 various water bodies in the northern Pilsen region during the year 2020. They were mainly post-mining sites or artificial ponds, where plankton samples were collected. Environmental characteristics (pH, conductivity, water transparency, temperature, amount of nutrients) were measured. Cyanobacteria and algae found in samples were identified to the possible lowest taxonomic level, also relative abundance levels were stated. The relationship between environment and the species composition was studied. A total of 77 phytoplankton species were recorded.
Sezónní dynamika bičíkatých řas v různých typech rybníků

The research was conducted in the surroundings of Toužim town. Samples of plankton and metaphyton were collected from 10 fishponds over the year 2020, samples were taken in spring, summer, autumn, and winter. Various environmental variables were measured such as temperature, conductivity, pH, transparency, and nutrient content. The specimens were identified and relative abundance was assessed and later used for counting environmental indexes. The main focus was on flagellated algae and their seasonal changes in composition among other alga were compared. All the environmental data together with species data were statistically analyzed.

Distribution and ecology of colourless euglenoids was studied on more than four hundred localities in each region of the Czech Republic. In this study, a checklist of observed taxa, description of chosen taxa and several ecological statistics were made and discussed.

The methodology was created under the National programme on conservation of microbial genetic resources of economical imortance. Its preparation was planned in the beginning of a current period of solving the National programme (2018-2022). The methodology describe procedures for the evaluation of specialization of individual collections from the point of view variability of genetic resources held on the collections. The goal of the methodology is to improve the work of microbial collections for the sustainable conservation of microbial genetic resources.

Natural compounds potentially influencing neurodegenerative diseases

Mňuková D.: Natural compounds potentially influencing neurodegenerative diseases. Diploma thesis, Charles University in Prag, Faculty of Pharmacy in Hradec Králové, Department os Pharmacognosy and Pharmaceutical botany, Hradec Králové 2022, 88 s. This research diploma thesis draws mainly from foreign professional literature. It provides an overview of important natural substances that potentially influence neurodegenerative diseases. The first part describes the moct common neurodegenerative diseases, their characteristics, epidemiology, pathogenesis and clinical picture. These diseases include Alzheimer's disease, Parkinson's disease, Lewy body dementia, multisystem atrophy, Pick's disease and other tauopathies, Huntington's disease, spinocerebellar ataxia and amyotrophic latelar sclerosis. The second part of diploma thesis presents natural substances that have been studied in preclinical and clinical studies in the last two decades for their potential to positively affect neurodegenerative diseases. The last part discusses the contribution of these natural substances fot therapy and prevention of neurodegenerative diseases. Algal and Cyanobacterial Adaptations to Low Temperature and Desiccation

Algae and cyanobacteria, due of their evolutionary antiquity, are widely distributed primary producers that can withstand extreme environmental stresses. Low temperature, freezing and melting, and desiccation and rewetting, are common stresses prevalent mainly in polar regions and in winter seasons of temperate areas. In terms of physiological effects, these types of stresses share similar effects or are closely related to one another. Low temperatures and desiccation exert a variety of stresses that need to be negated or lessened by adaptations. Specifically, adaptations to chill, freeze, and desiccation stresses will be discussed, as well as strategies that allow for stress avoidance or resistant morphological adaptations. In this thesis, characteristics, functions and mechanisms of these adaptations and stresses are reviewed, as well as potential biotechnological uses of said adaptations. Raman spectrometry of pigments of cyanobacteria, algae and lichens in the astrobiology context

This work deals with the possibility of Raman spectroscopical identification of selected biomarkers of extremophile species. It focuses mainly on selected cyanobacteria, algae and lichens with an emphasis on the ability to detect carotenoids. These pigments exhibit three characteristic bands of Raman spectra which represent stretching vibrations C=C; C-C and bending vibration C-CH3 in molecules of carotenoids. Raman spectra were measured not only by laboratory microspectrometers (λ - 514 nm and 532 nm), but also by portable and handheld spectrometers (λ - 532 nm, 785 nm and 700 - 1100 nm). In the case of cyanobacteria, the spectroscopical analysis was performed also on the fractions obtained by high performance liquid chromatography (HPLC). This work critically evaluates the possibilities of Raman spectroscopy to identify the carotenoids of cyanobacteria, algae and lichens. Besides the signal of carotenoids, interpretation of other bands in the Raman spectra corresponding to the presence of other biomarkers is given here for selected samples. Sexual reproduction of ochrophyte algae

Sexual reproduction is one of the specific features of eukaryotes. Almost all the knowledge that we have about the sexual process is based on studies conducted on animals and plants. In fact, we know only a tiny fraction of what protist organisms could reveal about distribution and other aspects of the sexual process. For the vast majority of them we miss any evidence that the sexual process actually occurs. However, it is conceivable that the putative protist asexuality is only a human artifact. Likewise, for most ochrophyte algae, which are the main subject of this thesis, the sexual process is unknown. It has been observed only in individuals belonging to six of the sixteen hitherto described classes of Ochrophyta. In some cases, an enviromnental factor that stimulates the formation of gametes was also found. The gradual clarification of the phylogeny that we can expect in the years coming, along with knowledge of biology of ochrophyte algae, can provide more information about sexuality in this group.
Vertical migration of benthic cyanobacteria and algae

Vertical migration has been observed in different taxonomic groups of benthic microalgae and cyanobacteria in both marine and freshwater habitats. It seems to be an advantageous survival strategy of motile photoautotrophic organisms. In general, algae inhabiting tidal environments migrate up to the sediment surface during daytime emersion and then migrate below the surface prior to high tide or dusk. Freshwater microalgae stay on the surface during daytime and migrate below the surface before dusk. The migratory rhythm can be controlled or influenced by light, waves, subsurface nutrient supplies and reducing conditions in the sediment. Sometimes, the rhythm persists even in constant conditions and it seems to be controlled by biological clock system.
Addition of probiotics and algae extracts to baby foods

This bachelor thesis deals with the selection of a suitable combination of probiotic culture with algae extracts as an addition to the baby formula and the application of the optimal composition of baby formula. Arame, wakame, hijiki and kombu seaweeds were selected for these purposes. The theoretical part is focused on the proper nutrition of children, their deficiencies and possible diseases. Subsequently, it deals with the characterization of probiotic cultures and their importance in the child's nutrition. Last but not least, the theoretical part characterizes the individual seaweeds, their nutritional composition and also their importance in the child's nutrition. In the experimental part, samples of selected algae were first analyzed in terms of saccharides, insoluble fiber content, fructooligosaccharides and -glucans or pigment content. A sample of inulin, a commonly used prebiotic in probiotic products, was used as a reference component. An important analysis of this experimental part was to determine the effect of algal extracts on the viability and growth of probiotic bacteria. Subsequently, a model digestion of a mixture of probiotics and algal extracts was performed, and the condition of the probiotic cells was evaluated during digestion and after digestion by measuring turbidity and flow cytometry. Finally, not only the optimal combination of algae extract and probiotic culture was proposed, but also the possibility of improving the long-term viability of probiotic cells in combination with algal extracts.
