Vlastimil Vyskočil, Ph.D. Title: New Electrochemical Methods for Determination of Nitro and Oxo Derivatives of Fluorene Year: 2010 Abstract of the RNDr. Thesis This Thesis represents a contribution to the search for new electroanalytical (polarographic and voltammetric) for sensitive determination of genotoxic environmental pollutants - nitro and oxo derivatives of fluorene, namely of 2-nitrofluorene, 2,7-dinitrofluorene, 9-fluorenone, 2-nitro-9- fluorenone, and 2,7-dinitro-9-fluorenone. An experimental work was focused on assessment of new approaches in the detection of the studied compounds. Interactions of calf thymus double-stranded DNA with 2-nitrofluorene and 2,7-dinitrofluorene at the newly developed voltammetric biosensor (DNA modified screen printed carbon paste electrode; DNA/SPCPE) were also investigated and subtle DNA damage under conditions of direct DNA-analyte interaction at room temperature and damage to DNA bases under condition of electrogeneration of short-lived radicals of nitrofluorenes at human body temperature were found. Health economics: What heals us and what kills us

This study deals with the health production function. It analyzes the impact of health care, socioeconomic, lifestyle and environmental factors on the mortality and life expectancy of the population of the Czech Republic. The analysis is made by linear regressions with time series data for the period from 1993 to 2011. Health care is measured by health care expenditures or by non-monetary indicators, the number of doctors and the consumption of pharmaceuticals. The results show that higher health care expenditures increase the mortality and reduce life expectancy. On the other hand higher number of doctors and higher consumption of pharmaceuticals improve the health status of the population. It may indicate inefficiency and corruption in health sector. Important factors that positively influence health are wealth, education and fruit and vegetable consumption, smoking affects health negatively. The results suggest that health care policy should focus not only on effective allocation of health care expenditures but also on lifestyle and socioeconomic status of the population. The limits of this work are short time series which don't allow the use of the lagged explanatory variables.

Corporate identity of a tractor manufacturer

This diploma thesis deals with corporate identity of a tractor manufacturer. For this purpose fictive company called Haart was created. The endeavour of this corporate identity is to make an image of a reliable company which specialises in the production of high-quality agricultural machines. This is accomplished by an orange-grey colour scheme where orange stands for living (agricultural production) and grey refers to lifeless (tractor is a machine). The final corporate identity is presented next to company stationery, merchandise, orientation graphics and web page also on the tractor designed within the pre-diploma project. For advertising graphics needs typeface Haart was created. Part of this diploma project is also the creation of a design manual. It describes all elements of Haart‘s corporate identity.

Phenotypic traits and pairing success of the Grey Partridge \kur{(Perdix perdix)}

The master thesis works measures of phenotypic traits and unique data of pairing of wild living grey partridges. The secondary sexual traits and theirs variability is described. Especially the relationship between the ornamental traits and the pairing success was researched. The results proved the greater pairing success of males with the larger melanin breast patch and the greater quality of the mate in females with the larger breast patch. The strategy of male floaters, which are realatively well ornamented is discussed.

Effects of olfaction and visual stimuli on mate choice decisions in Grey Partridges

This theses focuses on olfactory and visual components of mate choice in grey partridge. The aim is to evaluate the importance of melanin-based ornametnation and olfaction in mate choice and to explain the role of sexual selection in the evolution of secondary ornamentation in this socially monogamous galliform bird. To do that several experiments were conducted to (1) assess the ability of individuals to recognize conspecific odour and (2) to estimate the significance of melanin based feather ornamentation in male-male and male-female interactions. In some experiments I manipulated the expression (size) of feather maleanin- based ornamentation while in others I used birds forced to pair randomly to find out how the size of ornament and similarity in ornament expression between males and females (assortative pairing) affects reproductive success and investments (egg hatchability and number of eggs laid).

Condition dependence of secondary ornamentation and mate choice in Grey Partridges

This study focuses on relationships between individual condition, measures of ornament expression and mate choice in the Grey Partridge (Perdix perdix). It is based on the correlative data as well as results of manipulative experiments. I found positive correlation between the size of melanin- and carotenoid-based ornaments in partridges. The size and the redness of the carotenoid skin patch, and the size of the melanin-based feather ornamentation were associated with several measures of immunity and overall body condition (complement activity, ratio of immature erythrocytes, heterophil/leucocyte ratio, response to novel antigen). Furthermore, data from natural populations indicate that the expression of carotenoid-based ornamentation is associated with sexual attractiveness (measured as the ability to obtain a social mate) in partridges. Data from captive individuals indicate that carotenoid supplementation improves the redness of carotenoid-based ornament, while the stimulation of the immune system under increasing concentrations of supplemented carotenoids has no measurable effect on condition and physiological state. Development of Grey Partridge (Perdix perdix) microsatellite markers

The aim of this work was the construction of set of microsatellites for the Grey Partridge (Perdix perdix). I succeeded in isolation of seven new specific microsatellites. Another microsatellite markers were passed from close relative galliform species (Galliformes). On the whole I gained 18 microsatellite loci which were used for study of population charakteristics of three partridge populations from Stodůlky, Písecko and Milešín. The analysis showed conspicuous structuredness of these populations and surprisingly low values (less then 100) of their effective population sizes. Seven of the microsatellites were chosen for analysis of paternity. Indeed I found the evidence of extrapair paternity in this putatively monogamous bird. Finally I conclude, on the basis of sequenation of mitochondrial control region that our populations belong to the western linage of mitochondrial DNA.

Mechanical properties of soils of the Kolný sand pit

This thesis originated as a part of the PhD research carried out by Mgr. Radek Suchomel. To calibrate a hypoplastic model for granular materials a large number of laboratory tests was needed and a part of the testing constitutes the present thesis. The hypoplastic model by von Wolffersdorff (1996) has eight materials parameters. The critical state fiction angle φc', which was obtained directly by the measurement of the angle of repose, is one of the eight parameters. Other parameters are determined form the results of experiments and empirically. Aim of this thesis was to carry out field and laboratory tests, to determine selected soil properties and to interpret them with regard to the variability of the soil in the selected face of the sand pit. The material for investigation comes from the south part of the Cretaceous Třeboň Basin in the South Bohemia from the Kolný sand pit. The pit is located in the upper part of the Klikov layers. The fluvial layers are characterised by rhythmical variation of gravely sands, sands and sands with dark grey clayey inclusion. Samples were taken in the face of the sand pit with the dimensions of 9 x 36 m. Utilizing of selected agri-environment measures in the South Bohemian and Pilsen Region

Theoretical part of the diploma thesis (review) is focused on historical development of the Common Agricultural Policy (reforms from the years 1999 and 2003 are emphasized) and on the comparison of the agri-environment schemes (AES) of the Czech Republic, Slovak Republic, and Austria. The aim of the review was to summarize the processes, which led to the recent form of AES. In practical part of the thesis measures, which support maintaining and increasing of populations of ground game and birdlife, i.e. measures Catch Crops, Rich Seed Habitat (Biobelt), and Bird Area on Grasslands were evaluated. Data from Paying Agency SZIF were used. The study covers both, the (shortened) programming period 2004-2006 and the programming period 2007 {--} 2013 (years 2004 {--} 2008). Thesis is focused on region NUTS II Southeast. The following indicators were analysed: interest of the farmers in the selected AES measures, area on which the measures were carried out and amount of the financial support spent to support individual measures. Finally, steps to optimize the use of financial resources are suggested. Catch Crops are used as a measure on arable land. The highest interest of the farmers in Catch Crops was observed in the year 2004, when the largest number of the farmers entered this measure. The interest was decreasing in the following years (especially in the years 2007 and 2008, when the endowment was lowered). Catch Crops cover 6 {--} 8 % of the arable land of the region and the area has been increasing in time. Biobelt is only sporadically used measure (probably because of the underestimation of the costs), but interest in it (albeit low) has been increasing during the whole period. Measure Bird Area on Grasslands is restricted to nesting areas of Corncrake (Crex crex) and Waders (Charadrii). In NUTS III South Bohemian Region, the measure was applied on approximately 27 % of the area where it potentially can be used during the whole observed period. In NUTS III Plzeň Region, the area increased from 26% in the years 2004 {--} 2006 to 60 % in the years 2007 and 2008.
