Evaluation of transdermal permeability of caffeine using methylparaben as marker.

The theoretical part comprises of following topics: skin barrier, properties of methylparaben and caffeine, brief introduction to transdermal absorption, description of the diffusion cells and methods for processing the results. The results, obtained by permeation of methylparaben and caffeine from various donor media (TRIS buffer, TRIS buffer with propylenglycol, isopropylmyristate and isopropylmyristate with paraffin oil) through full-thickness pig skin, are presented in the following part of this thesis. Measured values of fluxes of caffeine JKF were as follows: from TRIS buffer: 35,2 µg/cmš.hod ±13,1, RSD 37,3%, from TRIS buffer with PG: 40,1 µg/cmš.hod ±28,6, RSD 71,5%; from IPM: 2424,6 µg/cmš.hod ±23,7, RSD 96,6%; from IPM and PO: 23,2 µg/cmš.hod ±24,8, RSD 106,8%. In the final part of my thesis, the individual results are summarised and processed, and the conclusion, that methylparaben is not a suitable marker for assessment of caffeine transdermal permeation from the media used, is drawn from them. The Measurement of Isolated Cardiac Cells Conctraction and Their Viability

The master´s thesis deals with research of viability and contraction measurement of cardiomyocytes. The work is divided into two main areas – theoretical and practical part. Theoretical part is aimed at electrophysiology of cardiomyocytes. Practical part includes detection of contractibility based on eccentricity in program Matlab. For research of viability, were used several media for example DMEM and MPRI.

Influence of pH and ionic strength on barrier abilities of humic substances

The main aim of this bachelor thesis is the study of humic acids and their barrier properties. The reactivity of humic acids was studied by simple laboratory diffusion techniques in plastic diffusion cells. This thesis is mainly focused on the through-diffusion processes in hydrogel media. Hydrogel media were based on the thermoreversible polysaccharide (agarose) with or without addition of humic acids. The rate of reactivity of humic acids was compared by the interactions with organic cationic dye – methylene blue. The influence of ionic strength and pH was tested as well.

SMV-2015-21: Optical frequency reference for visible spectral range

The content of this contractual research project for the institute of biophysical chemistry was a research and development of an optical frequency reference for the stabilization of a laser for spectroscopy in the visible spectral range. Spectroscopy measurements with a stabilized laser improve significantly the precision of the measurement thanks to direct traceability of the optical frequency to specific transitions in a suitable absorbing media that are listed in the recommendations of the international metrology commission. The reference is represented by a medium of ultrapure vapour of molecular iodine filed into a cell. The optical design of the cell includes windows with antireflexion coatings and a design with multipass arrangement.

SMV-2015-22: Samples of absorbing media for spectroscopy measurements

The content of this contractual research project for the company dealing with development of technology for enrichment of nuclear fission fuel was the design of samples of absorbing media for referencing of lasers for spectroscopy. The samples are absorbing cells with optical quality filled at various saturation temperatures. Thus the various pressure of saturated vapour was achieved.

SMV-2015-24: Optical frequency references for laser spectroscopy

The contractual project was oriented to development and research of novel types of absorption cells for laser spectroscopy applications – these cells are intended for frequency stabilization of ultra-stable laser standards and investigation of spectral parameters of unknown liquids and gaseous environments. Novel methods for preparation of antireflection coatings on optical windows were investigated due to strict request for minimalization of optical losses and backreflections of interacting laser light at optical windows of the developed cells. Research of methods for thermal protection of the optical windows during the welding process was oriented to unique approach of inert gas jet cooling of AR coatings. Investigation of possibility of AR coatings homogenization was done by method of thermal baking at different temperature levels.

The work was oriented towards desing and realization of optical setup for investigation of dissociated molecules of ultra-pure iodine 127I2 throught laser spectroscopy methods. The absorption cell was equipped with special-design thermostatic box with possibility of precise control of thermal dissociation of absorption media atoms and molecules.

Microbiological diagnostics of urogenital mycoplasmas

This bachelor thesis focuses on the diagnostics of urogenital mycoplasmas, namely Mycoplasma hominis, Mycoplasma genitalium and Ureaplasma urealyticum. These bacteria are the smallest freely living organisms capable of replication on artificial cell-free media. They are transmitted especially by unprotected sexual intercourse. They can then cause variety of urogenital infections, for example uretritis, bacterial vaginosis, pyelonephritis. Mycoplasma can also cause early fetal outflow, early labor, fetal death or postpartum fever. The thesis informs about the current state, basic morphology, bacteria occurrence, reproduction. It also deals with urogenital infections that occur in men, women or about potential transmission from mother on the fetus. The overview of the current state concluded with the description of - methods used for idenitification of these bacteria, e.g. direct or indirect detection. The methods used for examination of samples in the practical part od this thesis are discurred in detail. The practical part was conducted in the Department of Clinical Microbiology of the Na Bulovce Hospital, Prague. The Department provided me with samples from the genitourinary tract. The samples examined originated - from the time period July 2014 to December 2014. Altogether there were 380 samples available. 159 samples for examination of Mycoplasma genitalium and 221 samples for examination of Mycoplasma hominis and Ureaplasma urealyticum. Overall, - there were more positive samples from women, although there were more samples to examine from men than from women. In case of Ureaplasma urealyticum 43.6 % women were positive and 12.7 % men were positive, in case of Mycoplasma hominis 12.8% women and 7.7% men were positive. In most cases these were patients that showed clinical problems, patients, who had not been - previously examined and treated properly or thein condition was a chronical problem.

Effect of cryopreservation on mouse sperm.

Cryopreservation or freezing of sperm in the reproductive biology is still actual topic. Today is the only method used for sperm storage, whether for the purposes of assisted reproduction, or for scientific purposes. However, this method has a negative impact on such stored cells and is therefore still a subject of many studies. Among the main causes of sperm damage in rodents there are inappropriately selected cryoprotective agents, poor or no elimination of oxidative stress generated during cryopreservation, as well as a poorly chosen speed and temperature of freezing. Correctly chosen procedures and the composition of the media in which sperm are stored, can significantly affect the quality of sperm. This bachelor theses focuses on the influence of cryopreservation on mouse spermatozoa. The main impact of this method includes the effect of freezing on sperm DNA, plasma membrane, acrosome, and sperm motility.

Occurrence of irregular red blood cells antibodies in pregnant women examinated on immunohematological laboratory of UHKT

Blood is one of the main part of the internal environment of the organism. By composition and function is vital fluid which as mobile media connects all the organs and tissues in the body and plays a crucial homeostatic význam.1 circulating in the blood vessels (arteries, capillaries, and veins), composed of plasma and platelets (cells). Plasma is primarily water, electrolytes and blood proteins (albumin, globulins, a number of special protein). The stents blood include red blood cells (erythrocytes), white blood cells (leukocytes) and platelets (thrombocytes) .2 The total volume of circulating blood corresponds to about 8% by weight. For an adult it is approximately 4.5 - 6 liters of blood.
