Microscopic Simulation of the junction Malá Amerika

This bachelor thesis deals with the modeling of traffic flow in several variants of the transport network. Variants are modeled in Aimsun. Current status data was obtained by field observation. In the created models, the traffic flow of both current and future traffic intensities is simulated. Based on the outputs of the simulated operation, the variants are assessed. The proposed variants are compared and the most suitable arrangement chosen.
Processing Optimization of a Small Brewery

The bachelor thesis is focused on the section of information technology, specifically on the processing optimization by an information system design for a microbrewery. The thesis is divided into three parts. The first part is focused on the study of the current situation, especially deep data analysis. The second part deals with designing the information system architecture based on the clients requirements. The third part then implements the previous point to the final product, which is extended by an automated program for evaluating the most suitable work actions. The conclusion is a validation and verification of the final produduct and suggesting possible extensions.
Determination of chlorides in pharmaceutical substances

The topic of this bachelor work is a validation of the method for „Determination of chlorides in pharmaceutical substance“ is made in a form of validation of an analythical method. This titration method with potentiometric indication was developed in a company Sicor and it is used for determination of the residual chlorides volume in the substance called Pemetrexed. Validation is a process which assess suitability of a given analythic system for gaining relevant data (results). Validation constists of four tests: precision, reproducibility, lineraity and test of solution stability, although the test of reproducibility was realized in other laboratory (QC). For determnination of the chlorides was applyed titrator 716 DMS Titrino with argent combined electrode. For a data evaluation were used software tiamo 1.1 and MS Excel.
Problems of the mirror shutter in the LED module of the headlight

This bachelor thesis deals with methods of attaching the mirror shutter in LED modules in headlamps. The aim is to compare three methods of attachment which are used in one of the platform variants. The following are the methods of attachment, followed by the description of the light sources and functions of the LED modules; they are: riveting, hot stamping and fixation with clips. Risk associated with the assembly of mirror shutter into the light source are analysed via FMEA analysis, including also measures and actions for risk reduction – implementation of lighting of produced light sources in a special jig in the light laboratory. Vibrations tests were carried out to find out the influence of vibration on the mirror shutter in LED modules in the traffic. The tests revealed that even if the clip is incorrectly fixed (not attached to the end position) it does not affect the functionality of the module.
Aerosol and microbial microclimate of clean rooms

Master´s thesis deals with the issue of internal microclimate of clean rooms. The experimental part focuses on aerosol and microbial microclimate. Different operating and intervention rooms were measured and then evaluated whether the cleanroom would comply with the standard cleanliness class even in normal operation after previous validations. The microbial examination of different clean spaces investigated the extent to which the exact passive fall method and its evaluation were time-dependent. From the data the correct desing of the air-conditioning system and its subsequent realization and regultion were verified. The theoretical part describes requirements for clean rooms according to SÚKL-LEK 17 and ČSN EN ISO 14644-1. In addition, a detailed methodology of the procedure of air-conditioning implementation is elaborated to the final validation and testing of clean rooms.

Využitím diskrétní simulace programu Witness Power with Ease se v diplomové práci optimalizuje logistický tok materiálu ve společnosti Hella Autotechnik, s.r.o. Práce přibližuje metody a jednotlivé fáze tvorby modelu včetně jeho validace a navrhuje vylepšení, díky kterému by mělo dojít ke snížení nákladů na dopravní služby o 24 400 Kč měsíčně.
Determination of content of selected nonsteroidal antiphlogistic – Ibuprofen in drugs using UV/Vis and FTIR spectrometry

The main aim of bachelor thesis was to validate the content of the nonsteroidal antiphlodstic in drugs using spectrometric methods and subsequent comparison of results. Specifically, the thesis is focused on Ibuprofen, which is used mainly as analgesic and antipyretic. This substance was chosen for its significant ability of absorbing electromagnetic radiation in ultraviolet and infrared regions of spectra. For this purpose, various generics were selected and analyzed by molecular absorption spectrometry (UV/Vis) and infrared spectrometry with Fourier transform (FTIR). Very important factor influencing chemometric properties of the obtained results was optimalization of the spectrometric methods. After designing the working procedure, sample solutions were prepared from Ibuprofen-containing dosage forms using suitable solvent and these samples were analyzed spectrometrically. The content of Ibuprofen in individual samples was calculated from the measured UV/Vis and FTIR spectra using method of standard addition. The results show that infrared spectrometry seems to be more suitable method for determining the Ibuprofen content. Nevertheless, it should be noted that the choice of a suitable extractant is very important step in the determination. The information obtained from this bachlor’s thesis could be used in future routine inspections and monitoring of drugs.
The chosen validation parameters of method for assessment of aroma compounds in processed cheese analogues

This Bachelor‘s thesis deals with the issues of method validation for assessment of aroma compounds in processed cheese analogues. The method is based on the isolation of volatiles from the sample using Solid Phase Microextraction (SPME) and their subsequent determination by gas chromatography. The theoretical part focuses on general characteristics, classification and producing technology of processed cheese analogues. Further, aroma active substances contained in cheese analogues, differences from the classic processed cheese and processed cheese analogues occurring on the Czech market are mentioned. The methods for determination of aroma active compounds with a focus on Solid Phase Microextraction and gas chromatography are briefly mentioned. Within experimental part of the work the linearity and repeatability of the SPME-GC method were verified and detection limits and limits of quantification of selected standards were determined. Then the method was applied to chosen samples of processed cheese analogues with addition of different types of vegetable fats.
Covalent geometry of nucleic acids

IV Abstract Nucleic acids (NA) are substances that play a fundamental role in all living organisms. They store genetic information and control the process of protein synthesis. These macromolecules are composed of chains of nucleotides. According to the type of sugar in the nucleotides, nucleic acids are divided into DNA (Deoxyribonucleic acid) and RNA (Ribonucleic acid). Nucleotide sequences also determine the spatial conformations of nucleic acids. At the level of dinucleotides, conformations are described by classes of dinucleotide conformers - NtC. To study nucleic acids, it is crucial to determine their spatial arrangement. The most common method of three-dimensional observations is X-ray crystallography, which uses the interaction of X-rays with the electron cloud of atoms in the crystal. The result is crystallographic data, which further goes through the process of refinement and validation. One of the essential requirements for reliable refinement and validation are stereochemical standards. These are vocabularies that characterize specific bond lengths, bond angles, torsion angles, planes, and chirality. Determination of bisphenols in selected beverages by high-performance liquid chromatography

Bisphenols are chemical compounds with a wide range of uses in the chemical industry. They are used in the production of plastic materials as softeners for polymer materials or epoxy resins, as monomers in the synthesis of polycarbonates, or for the elimination of hydrochloric acid in the production of polyvinyl chloride. This thesis deals with the development, validation, and optimization of an analytical method for the determination of selected bisphenols, specifically bisphenol A, bisphenol B, bisphenol F, bisphenol A diglycidyl ether and bisphenol F diglycidyl ether (BPA, BPB, BPF, BADGE, BFDGE), and their metabolites using the high performance liquid chromatography with mass spectrometry, HPLC/MS, and the application of the method to real samples in selected canned beer samples. The following conditions were optimized for chromatographic separation. The Sigma-Aldrich discovery C18 column (150×2,1 mm, 5µm) was chosen. The mobile phase consisted of a mixture of mobile solvent A, which contained 0,05% ammonium hydroxide, and mobile solvent B, which contained of methanol. The flow rate of the mobile phase was 0,240 ml/min. The total analysis time was 22 minutes, using a 5 μl injection of the sample solution with gradient elution.
