The bachelor thesis is focused on the study of antioxidants and detection methods of their antioxidant activity. Antioxidants are substances able to scavenge the activity of oxygen radicals. This ability protects critical cellular structures from oxidation. In nature, plants belong to the important sources of antioxidants. In this thesis we are specifically focused on the detection of antioxidant activity of Japanese knotweed extracts using FRAP (“Ferric Reducing Antioxidant Power”) with automatic UV-Vis spectrometer. The data obtained were treated with the mathematical program developed in MATLAB.
The dynamics of the MRP1/2 complex and the function of intact MRB1 core for RNA editing in \kur{Trypanosoma brucei}

This thesis describes the dynamics of mitochondrial RNA-binding protein 1 and 2 (MRP1/2) complex in different cell lines of Trypanosoma brucei under an optimized immobilized condition. This study reveals the influence of RNA on the complex's dynamics. Furthermore, the function of RNA-binding complex 1 (MRB1) core has been studied via reverse genetic, biochemical and molecular techniques, with its role in RNA editing being proposed.
Polyfenolické látky modrých hroznů a červeného vína a jejich význam ve výživě člověka

This bachelor thesis is focused on the topic Polyphenolic substances of blue grapes and red wine and their importance in the human nutrition. It was written from 2013--2014 at the Department of Post-Harvest Technology of Hurticultural Products, Faculty of Horticulture, Mendel University in Brno. Literary section pays attention to polyphenolic compounds contained in grapes and red wine. It describes their characteristic, chemical structure, formation, distribution and their antioxidant effect on the human organism. Experimental section analyzes antioxidant capacity, volume of anthocyanins and the total amount of polyphenols inside grapes, musts and wines of two varieties 'Cabernet Moravia' and 'Lemberger'. The results are presented in tables and expressed in graphs. Graphs show us the condition of observed factors in a process of grape processing into the wine and compare different samples, varieties and methods. Results are statistically evaluated and compared with other authors.
Mobility of photosynthetic proteins

Mobility of pigment-protein complexes (phycobilizomes and photosystem II playing a key role in photosynthesis) was studied by FRAP method (Fluorescence Recovery After Photobleaching). FRAP represents a fluorescence based microscopy method enabling measurement of protein mobility in living systems. The protein complexes are bleached by a laser pulse. And mobility of unbleached proteins is measured as a fluorescence recovery in the bleached area. Currently we have only limited knowledge about the mobility of photosynthetic proteins. This work was aimed at optimization of the photosynthetic protein mobility measurement by FRAP. I have performed several methodological experiments which led to the successful assessment of phycobilisome and chlorophyll-containing proteins diffusion coefficients in selected red algae (Porfyridium cruentum, Cyanidium caldarium) and cyanobacteria (Synechocystis PCC6803, Acaryochloris marina). The methodology developed and validated in my thesis was then applied in further research projects.
Stanovení antioxidačního statusu u potkanů při zkrmování pšenice Citrus

This thesis examines the influence of wheat Citrus with higher content of lutein in comparison with common wheat on antioxidant activity of the rat blood plasma and liver tissue. In the experiment, 32 male rats of Wistar strain were used. The influence of the feeding of yellow wheat varieties Citrus on antioxidant activity was measured in the blood and liver tissue. The experimental group (N = 16) was fed with dried granules of 100% wheat meal from Citrus. The control group (N = 16) was fed with similar feed prepared from two common wheat. Content of crude protein was the same in the both group. Body weight gain were followed in three-day intervals and feed consumption was followed daily. Antioxidant activity was measured by DPPH, FR, FRAP and ABTS methods. Values are expressed in gallic acid equivalent. In the blood significant differences (P < 0.05) of antioxidant activity was determined only by ABTS method between "Citrus" group (405.51 +- 7.64 mg /ml) in comparison to "Control" group (479.99 +- 22.54 mg/ml) in contrary to expectation this value of control group was of 18% higher. Values of other antioxidant methods were not significant. In the liver tissue only average value of ABTS method was little higher in Citrus group (409,439 +-13,626) in comparison with control group (389,493 +- 9,271 mg/ml). Similar situation was with DPPH method (20.711 +- 0.867 in Citrus group and 20.037 +- 1.109 mg/ml in the Control group) and difference was also not significant (P> 0.05).

Planning of restoring Critical Infrastructure on local level

Critical infrastructure (CI) is one of the most important branches in crisis man-agement. In recent years, CI demonstrated its importance on many occasions. My work is focused on the analysis of risks which can be a possible threat to CI. This is because the most important part of securing CI is the prevention of, and prepara-tion for, interruptions or damage. The risks in this part were specified using a FRAP analysis. I also identify the main reciprocal dependencies of all CI sectors to demonstrate which sector is the most important one. Finally, I specify the process of recovering or restoring these CI sectors. A new method was intentionally used for analytical-synthetic models. This method, in conjunction with the use of a FRAP analysis, is new to the branch of crisis management and was used for first time. The main aim of my work is to improve knowledge about CI at a regional level {--} city of České Budějovice. I submit new ways of identifying risks and resolv-ing problems during the recovery phase {--} specialized in CI.
