Purification and characterization of selected human microsomal reductase

Area of human liver microsomal reductases hasn't been fully explored yet. Purification and characterization of still undescribed enzymes is an important step in finishing of this task. There is separation and purification of this reductases described in my work, with usage of hydrophobic interaction chromatography on low pressure chromatography instrument ÄKTA purifier. There were two columns with different substrates used, Phenyl sepharose (low sub) and Octyl sepharose. Separation conditions were the same in both cases. Results obtained by incubation of fractions and measurement on HPLC showed significant differences between both columns and their substrates. But it isn't possible to determine, which column is better for purification, because big area still remain for adjustment and optimization of purification conditions.

Dietary habits and lifestyle in metabolic syndrome

ABSTRACT Title of the work: Eating habits and a lifestyle by metabolic syndrome This bachelor thesis deals with the eating habits and the lifestyle in patients with metabolic syndrome treated in diabetologic clinics. The work is divided into two main parts. The first (theoretical) part deals with the existing and newly discovered knowledge in the area of this issue, the eating habits and lifestyle. Primarily, I studied the issue and changes in MS. In the initial chapter I mention the main components and consequently I deal with most discussed changes in the recent years. The last specified part deals with the factors affecting the MS; here,I have been concerned with the most important categories, including eating habits, physical activity, smoking, genetics and stress. The second part (empirical) should clarify the problems in complying with good eating habits and lifestyle. It should prove the correctness and effectiveness of education in persons showing symptoms of MS. The work was designed as a quantitative research in which the target file was constituted by a selected sample of 120 respondents showing MS in diabetologic clinics. To obtain the necessary information, a questionnaire survey was used. The research confirmed a relatively good education and eating habits in patients with MS. During questioning it was found that 77% of respondents claim to have a sufficient amount of information and good eating habits. But a deeper examination of their dietary habits revealed that only 67% of them actually know how to eat properly. On the other hand, 23% of the observed patients lack of the information from a doctor, therefore they cannot have good eating habits. A positive result is that two thirds of patients educated in the diabetologic clinics are really able to eat properly and work with the information they receive. Unfortunately, the remaining 10% have enough information from their doctor, but they cannot work with the information so as to achieve the correction of dietary habits. A part of this bachelor thesis forms a health educational material that is destined for patients who do not have enough information or those who are familiar with the issue, but are interested to learn more.

Qualitative research in the aplication of desinfection and sterilization of nursing unit

Abstract The thesis focuses on the application of disinfection and sterilization in the surgical unit. These methods are an integral part of nursing care and they prevent creation, transmission and spread of infection. The qualitative research was carried out in the surgical department in the Hospital Prachatice a.s.. The method used in the research is an unstandardized in-depth interview, in the second phase extended to a structured observation. There were set three objectives. Objective 1: to determine the ways of application of disinfection in the surgical unit, Objective 2: to determine the ways of application of sterilization in the surgical unit, Objective 3: to determine whether nurses observe the principles of disinfection and sterilization in the surgical unit. On the basis of these objectives, there were formed three research questions: What types of applications are used for disinfection in the surgical unit? What types of sterilization applications are used in the surgical unit? What principles of disinfection and sterilization are observed in the surgical unit? From the results of the in-depth interview and observation, it was discovered that physical and chemical disinfection is applied in the surgical unit. The employees use the services of central sterilization. In the surgical unit, there is not used any type of sterilization, except of the pre-sterilisation preparation. While disinfection, the employees follow the correct procedure, but they do not always use the correct amount of disinfectant. As the possibility of improvement, I would suggest to carry out training and remind that the recommended concentrations of disinfectants are sufficiently effective. There is no need to increase their amount hence it increases the financial costs of the surgical unit.

The usage of modern methods of moist healing in practice

ABSTRACT The usage of modern methods of moist healing in practice Introduction: The method of moist healing has been reintroduced in practice during the last years. This revolutionary change does not only concern the usage of specific materials and a complex approach to solving the problem, but also doctors, medical staff and even patients have started to be more interested in this specific topic. The development of modern way how to heal wounds in practice has divided medical staff into two groups; the ones who specialize in this method and the ones who respect new possibilities, however, have little experience with the issue. Objective: The aim of my bachelor thesis is to find out whether nurses make a choice in kinds of material used for moist wound healing according to the phase of wound healing (TIME system), if the materials are used more preventively or therapeutically and finally, whether nurses cooperate with doctors on the choice of material. The research sample was formed by 89 nurses working in standard departments (e.g. department of surgery, department of internal medicine, department of subsequent care) in Hospital Ceske Budejovice, JSC and Hospital Prachatice JSC. Results: I have discovered that my hypothesis (H3) ? nurses cooperate with doctors on the choice of material used for moist healing is confirmed. Hypothesis (H2) said that the materials used for moist healing are used mostly therapeutically in practice. According to the survey, I have discovered that nowadays these materials are mostly used either therapeutically or preventively. The result is presented in graph 15. As it was found out, materials used for moist wound healing are used in 57 % therapeutically as well as preventively in hospital care and only in 43% therapeutically. Hypothesis H2 is not confirmed. My hypothesis (H1) said: ?The choice of material used for moist healing is performed according to the phase of wound healing?. The results surprised me hence the hypothesis is not confirmed. Only 48 % of the respondents use TIME system. The difference between the usage and no usage of TIME system is only 4 %, which amounts to 3 people out of the total number of 75 respondents (100 %). Conclusion: The results of my research could be used for an extended research, as a source for standards and seminars for nurses and also for an article published in a specialized magazine.

Transdermal in vitro permeation of cyclosporine IV.

Rigorosal Thesis 2006, Hana Krejčová Abstract The theoretical section gives a brief overview of anatomy, physiology and function of the skin, transdermal formulations and options for increasing transdermal absorption of drugs. The main part of the chapter is focused on cyclosporine includes updated information published on knowledge of the human immune system, and immunosuppressants. Experimental work presents the results of in vitro permeation experiments on pig skin. Donor and acceptor phase was in most experiments consisting propylene glycol and Tris-buffer pH 7.1 at a 1:1 ratio. Only a few experiments have been used as the donor phase of 10% dispersion of CyA in isopropylmyristate. Permeant determination was performed by HPLC with UV detection at 220nm. As a donor samples were used in the supersaturated system without CyA Enhancer, then a 5% addition of DMI or arlasolve, pyrrolidone or transcutol, or 1% transkarbam 12. All enhancers in the used concentrations icreased a depot formation in the skin, transkarbam 12 in the lowest concentration. For transdermal in vitro permeation experiments CyA appears to be administered in more lipophilic donor phase. The ideal length of the permeation experiments, it appears time 15 hours.

Abstract: The main topics of my BSc. thesis are the modern non-contact temperature measurement methods of various materials, specifically building materials. The thesis deals with temperature as a physical quantity, with its contact measurement and, most importantly, the principles of accurate non-contact temperature measurement by a thermal imaging camera. A substantial part of my thesis deals with the evaluation and description of thermograms. These results can be used both for further enhancement of this modern methodology, and also in practice ? to search for heat leaks in faulty thermal insulation as a precaution measure in the engineering or energy industries.

Druhý (měnový) pilíř měnové politiky ECB, jeho charakteristika a relevance pro měnovou politiku ECB

Abstract Monetary policy of the ECB was created under the conditions of extreme uncertainty. This was probably one of those reasons, why Otmar Issing ? the mastermind of monetary pillar of ECB´s monetary strategy decided not to go the conventional way. His decision and the key principle of the monetary pillar ? to give the money a prominent role - faced and still face the criticism. Nevertheless, the monetary policy of the ECB works quite successfully more than eight years after the implementation of the two pillars approach. Thanks to the stable price level, the ECB is these days one of the most credible institution in the world. If the credibility of the ECB is also a merit of the monetary pillar, how the monetary pillar was created, how it works, its contribution to the price stability in the Euro zone, and if implementation of this pillar was a right decision ? this all is going to be discussed in this bachelor paper. To byl pravděpodobně jeden z hlavních důvodů, proč se Otmar Issing, mentor měnového pilíře, rozhodl pro netradiční strategii měnové politiky pro nadcházející měnovou unii. Jeho rozhodnutí a zároveň základní princip měnového pilíře ? prominentní role peněz čelí neustále kritice. Měnová politika ECB navzdory kritice splňuje stanovený cíl cenové stability už více než osm let a vzhledem k tomu je nyní jednou z nejdůvěryhodnějších institucí. Zásluha měnového pilíře v dosáhnutí zmíněné dlouhodobé cenové stability je však téma k diskuzi. Historie měnového pilíře, jeho fungování, charakteristika a zejména relevance pro měnovou politiku ECB je předmětem této bakalářské práce. Possibilties of the use of an automated warehouse in a chosen food processing company

The bachelor?s thesis topic is ?Possibilties of the use of an automated warehouse in a chosen food processing company?. It focuses on the present functioning of the automated warehouse in Budějovický Budvar, n. p. The solution of the problem of 5% rejected products into the automated warehouse due to packaging defects was proposed on the basis of discovered facts. The proposed system of the solution succeeded to eliminate the problem of 5% of rejected products into the automated warehouse. The most suitable alternative was chosen on the ground of the cost analysis of the particular solutions. It is necessary to ensure the possibly highest quality of the performed work in the phase of packaging due to increasing wage costs and the need of the company to effectively handle the costs in the future. Only adequate controlling can ensure it. Currently, the packaging quality control is set to the moment of receiving the products into the automated warehouse. In this way, the defect cannot be eliminated on time, it can only be notified. The solution informs about the possibility of a defect in time when it can be eliminated easily and without additional costs right by its origin. The shift of the control to the packaging phase depends only on time and on the amount of additional cost for the control launch compared to the costs of the repeated packaging.
