Quality of agricultural products from vending machines for agricultural products and evaluation of their use

The bachelor thesis addresses the issue of selling agricultural products through vending machines. It deals with the characteristics of individual products, legisla-tive regulations and standards governing their sale, as well as the companies operat-ing vending machines. Publicly available sources, personal survey conducted in selected businesses and questionnaire distributed tough the social media platforms were used to obtain this information. The main finding of this research is that for large businesses the income from vending machines is not the main source of in-come, but small businesses have higher income from the vending machines. How-ever, the public has insufficient awareness of the machines.

The thesis deals with the relationships between the course of milking and the order of milking during the day, milk yield, stage of lactation, breed affiliation of cows and changes in the course of milking and other monitored indicators in cows with masti-tis. The literature review discusses current trends concerning the milking process, milk flow, monitoring of milking parameters, management software and factors that affect the whole milking process. The overview, which focuses mainly on mastitis and the risk factors associated with it, also covers various udder diseases. Data from each milking was collected for two farms and then the effect of the different grading cri-teria on the observed milking parameters were assessed. These were milking order, number of days in lactation on milking day, milk yield, average milk flow (in the first 15 s, 15 to 30 s, in the second 30 s and in the 2nd min of milking), proportion of milk yield in the first 2 min of the total milk yield, maximum and average milk conductivi-ty and temperature, average and maximum milk flow, total milking time and milking time at milk flow below 1 l/min. Subsequently, the data for DIM, milk yield, average milk flow (in the first 15 s, 15 to 30 s, in the second 30 s and in the 2nd min of mil-king) and the proportion of milk yield in the first 2 min were calculated. The diffe-rence between the maximum and average conductivity values and the difference between the milk flow values in the second 30 s of milking and in the second 15 s from the start of milking were also calculated. From the results it was found that Holstein cows, compared to dairy cows of the Czech Spotted Cattle breed, have a higher average milk yield, higher average milk flow and shorter total milking time. Also, milk yield in the morning milking is not always higher than in the afternoon milking. The average milk flow was highest from day 61 to day 120 of lactation. In the case of excessive flow, total milk yield, milk yield in the first 2 min of milking and its share of total milk yield were higher. In bimodal milk flow were found higher milk flow in the first and second 15 s of milking and also higher maximum milk flow and lower flow in the second 30 s of milking. A puzzling finding was the milk con-ductivity, as it decreased on the day of mastitis, when the conductivity should be highest. According to some authors, milk conductivity should always increase with the temperature of the milk sample, but this claim was not confirmed from the results.
Barcoding společenstva makrozoobentosu Křesánovského potoka

Molecular genetic methods, including barcoding, have been increasingly utilized in recent years for the analysis of communities of macrozoobenthos and many other organisms. Morphological identification of precise species of mayflies, caddisflies, or stoneflies in their larval stage is demanding and requires a significant amount of time and expertise. Barcoding analysis can help to classify disputed species more accurately. The aim of this thesis is to use the barcoding method to determine selected representatives of macrozoobenthos and verify the morphological determination of macrozoobenthos representatives. The studied locality is Křesánovský Stream in southern Bohemia. DNA was extracted from the collected samples and using mitochondrial gene CO1 and primer pairs LCO1490, HCO2198, jgLOC1490, and jgHCO2198, sequences were created and tested in the BLAST program. The most similar sequences from publicly available databases were obtained, compared to sequences from this thesis and the exact species were subsequently determined. A total of 57 samples were studied, from which 29 species were morphologically identified (six species of mayflies, eight species of caddisflies, seven species of stoneflies, three representatives of beetles, four species of dipterans, and one specimen of Gammarus fossarum), and 30 species were identified through barcoding (11 species of mayflies, eight species of caddisflies, five species of stoneflies, two species of dipterans, three species of beetles, and one species of fleas). Morphological determination was successful in 63% of cases. The success rate was 74% for mayflies, 67% for caddisflies, 33% for stoneflies, and 25% for dipterans. The specimen of Gammarus fossarum was morphologically identified correctly. Barcoding appears to be a suitable method for studying macrozoobenthos and an excellent complement to morphological analysis of samples. However, it has its limitations, and it is not appropriate to consider it as the sole method for determining macrozoobenthos samples.
The Situation of Literature Teaching in English Language Classes in Czech Secondary Schools

Although literature can be seen as a means of improving students' English proficiency, its role in secondary education in the Czech Republic is often insignificant. Literature should be an important and integral part of the comprehensive development of English as a second language for students. Therefore, the aim of this thesis is to investigate the role of literature in secondary teaching of English as a second language in the Czech Republic. This thesis includes a theoretical introduction to the topic of the importance of literature in language teaching and learning and research based on questionnaires completed by both secondary school English language teachers and students, which provides an insight into the perspectives of participants from both sides of the educational process on the examined topic.
The Ideal City Brno: The New Stará Osada

My master thesis deals with the urban locality Stará Osada, which is an artery of the urban district of Brno- Židenice. It is a social and transport hub for the Brno - Židenice district. Stará Osada is part of the wider city centre. This traffic junction is defined by a development of prefabricated houses, where the dominant feature of the environment is a thirteen-storey building. Stará Osada could be defined similarly to Mendel Square in Brno. Compared to Mendel Square, the space of Stará Osada is very fragmented, disorganised and people only use it as a transfer point for public transport. In connection with the construction of the New Armoury and the traffic tunnel, traffic calming on Gajdošova- Svatoplukova Streets, which currently form the city ring road, will be addressed in the future. Stará Osada has the potential to be a full-fledged centre of Židenice, which supports the development of the urban district, which the district strives for. The aim of the thesis is to design a public space in the form of a square with new buildings, supporting the development that is lacking in the urban district. The buildings, by their mass, will enclose and integrate the space of Stará Osada and will be a manifesto of a possible architectural and urbanistic solution for Stará Osada. The square and the adjacent buildings will provide the opportunity for increased social events and new housing.
Study of the interaction of lung surfactant with selected polymers

This thesis deals with the study of interactions between pulmonary surfactant (Curosurf) and selected polymers (N,N,N-trimethylchitosan, polyvinylpyrrolidone, sodium hyaluronate). This work also focuses on the inactivation of lung surfactant using bovine serum albumin, which causes an increase in the surface tension of the surfactant, whereby the inactivated surfactant is unable to fulfill its function in the lungs. The addition of polymers to this mixture has been shown to be an effective means of restoring the surface tension of the surfactant. First of all, this work focuses on finding the optimal concentration ratios of polymer and Curosurf, at which their mutual interaction occurs. The method of dynamic light scattering was used to obtain optimal ratios. In the second part, the work is devoted to the determination of surface tension using a du Noüy ring tensiometer. It was found that the addition of all selected polymers led to a reduction of the surface tension of the inactivated surfactant to values close to native Curosurf. This work provided useful information to understand the mechanism of lung surfactant surface tension recovery.

Nowadays, it is essential to find ways to integrate the old urban fabric into the contemporary city and to fight against the emergence of mono-functional zones in small towns like Rosice. In addition to the large cities, smaller communities within commuting distance are also struggling with a process of densification that has for the most part been solved by exporting dense patterns from cities or building typologies that do not always fit these suburban conditions. Often, it is precisely by improperly intervening and simply copying dense urban models that the distinctive identity of a place is lost, and this intervention is not adequate to the complex context of these suburban areas with their seemingly disordered heterogeneous landscapes. By exploring alternative forms of housing, different and new forms can emerge, which can promote precisely diversity and new approaches, while at the same time leading to the integration of this heterogeneous urban landscape and combating the diagnoses of postmodern society.
Kohnova cihelna (Kohn's brickyard ) – a carbon-neutral city

In recent years, there has been increasing talk of carbon-free districts, cities and even continents in the context of global warming, with intensive research into the possibilities and limits of this phenomenon. Also in this context, specific urban and architectural proposals are being developed more often than before. These proposals work with carbon-free technologies and building or urban elements that can reduce production of carbon emissions and its absorption into the atmosphere. This work also deals with this. Specifically, this work locates a new (mostly residential) district on the eastern side of Červený kopec, in the area of the former Kohn's Brickworks, and through the use of various urban design approaches and building technology solutions, it seeks to move towards achieving carbon neutrality for this complex. Author does so in the knowledge that he is not able to achieve an ideal state under the current conditions, but he considers it necessary to at least come as close to it as possible. In doing so, he applies the findings of his research on the topic of carbon neutral cities.
The Sustainable City: The Žižka Barracks in České Budějovice

The subject of the diploma thesis is the proposal for the transformation of the former Žižkov barracks complex in České Budějovice. The area is located near the historical core. Currently, the complex is inaccessible to the public, and almost all buildings are vacant. The ensemble of buildings is not a cultural monument. The architectural-urbanistic study proposes a multifunctional urban development in the area. The theories of Aldo Rossi, Jan Gehl, and David Sim are applied in the urban design. The aim was to create conditions for construction that support the circular economy of building structures. The concept of these principles is demonstrated in the design of a community center building. Economic, social, and ecological values of the project are compared in the conclusion.
Nursing care for alcohol-intoxicated patients in the traumatology unit

The thesis addresses the issue of nursing care for patients intoxicated with alcohol in the trauma centre, while being divided into two parts, meaning theoretical and practical. The theoretical part focused on defining the field of traumatology, injuries, ethyl alcohol, and the connection between injuries and alcohol. It also provided a description of patient care in traumatology, the specifics of nursing care for alcohol-intoxicated patients, and communication with intoxicated patients, including communication with aggressive patients. The aim of the research part of the thesis was to describe the specifics of nursing care for alcohol-intoxicated patients and to evaluate the communication between the patient and the nurse. Qualitative research was chosen for the research part. Data collection took place through written and verbal methods in the form of semi-structured interviews in early April. Data analysis was conducted using the pen-and-paper method. Subsequently, the data were organized into a system of open coding, comprising three categories and twelve subcategories. The research sample consisted of ten general nurses working in the trauma centre, six in the standard ward, and four nurses in the intensive care unit. The results indicated that except for one nurse, the surveyed nurses regularly encounter patients intoxicated with alcohol in their workplaces. According to the statements of the informants, the most common types of traumas associated with alcohol intoxication include falls, traffic accidents, suicides, assaults, visits to catering establishments or work in construction. Furthermore, the interviews revealed that the surveyed nurses perceive differences in nursing care and communication with patients who have trauma unrelated to intoxication and vice versa. They find the approach to intoxicated patients more demanding in this regard. In the interviews, the nurses mentioned the frequent practice of dealing with aggressive, uncooperative patients. Some nurses admitted that their personal attitudes towards intoxicated patients often influence their approach to these patients. It also emerged from the interviews that the surveyed nurses are not offered any further education in this area. The obtained results provided insights into the specifics of the issue, indicating potential communication shortcomings with patients for correction. The thesis will serve as a resource for creating simulation training scenarios (Appendix 1) to educate general nurses working in the trauma centre.
