Calcium homeostasis and modulation of nociceptive synaptic transmission

This study was designed to improve our knowledge regarding mechanisms of nociceptive signaling at spinal cord level. One of the forms of spinal cord synaptic transmission modulation is central sensitization, a manifestation of synaptic plasticity at spinal cord level, which was found to be present at many chronic pain syndromes. This study deals mainly with a development of calcium imaging technique with a final goal to study mechanisms of central sensitization in vitro on population of dorsal horn neurons. We have analyzed synaptically evoked intracellular Ca changes as a result of dorsal root stimulation in a superficial dorsal horn area in spinal cord slices and found two types of Ca responses: one synchronized with electrical stimulation and a second one, delayed response due to Ca release from internal stores. The delayed Ca release was not previously shown to be present in these neurons and it was not dependent on activation of ionotropic glutamatergic receptors, suggesting involvement of metabotropic receptor pathway. The presence of this delayed type of Ca response could have a significant role in the induction of some types of chronic pain syndromes, since intracellular calcium increase is thought to be a key trigger point in spinal cord neurons sensitization. Capsaicin is the main component of chilli peppers responsible for pungency. In this bachelor thesis, the presence and concentration of natural capsaicinoids (capsaicin and dihydrocapsaicin) extracted from 12 different varieties of chilli peppers (Jalapeño, Bhut Jolokia Yellow, Habanero Orange, Carolina Reaper, etc.) were determined by high performance liquid chromatography. Each variety was prepared without seeds and partitions and with seeds and partitions in order to compare yields of capsaicin and dihydrocapsaicin depending on sample preparation. Finally, for individual varieties of chilli pepper, the range of pungency were evaluated in SHU units based on capsaicin concentration.

The aim of this thesis was in cooperation with commercial grower to evaluate the experiment with available varieties of chilli peppers from the perspektive of yield and growth. Furthermore, to analyze selected substances and the results obtained statistically process, evaluate and determine the differences between the different varieties of chilli peppers. Chilli peppers have quickly become a popular crop, especially due to their pungency and high vitamin C content. Pungency is due to the presence of capsaicin, whose content can reach up to 2 200 000 SHU. From chilli peppers you can prepare a wide range of products from jams to sweets to drinks. Chilli peppers are very perspective species, especially in the pharmaceutical industry.

This diploma thesis “Evaluation of growth and qualitative parameters of chosen chilli peppers varieties” is about the comparison of the cultivars of chilli peppers. There have been 13 cultivars involved in the experiment. The first part of the thesis is about literary overview about the technology of growing. In the second, experimental part, the qualitative and quantitative characteristics of the varieties have been evaluated – product, growth and contents substances (vitamin C, capsaicin, dry basis) of the plants. The experiment was successfully conducted in cooperation with the company World of chilli, s.r.o. in 2018 and subsequently it was evaluated in the laboratory of ZF Mendela in Lednica.

The first part of study involves historical chilli pappers origin and the sekond part focuses on evaluative floricultural values experimental and substances of chilli pappus. This thesis was elaborated in cooperation with World of chilli s.r.o.
The role of nociceptive synaptic transmission modulation at the spinal cord level in different pain states

Pain is a common symptom of many clinical syndromes and diseases. In particular, the treatment of neuropathic pain represents a serious public health issue because currently available analgesia is ineffective in many cases or it has adverse effects. Treatment of pain-related suffering requires knowledge of how pain signals are initially generated and subsequently transmitted by the nervous system. A nociceptive system plays a key role in this process of encoding and transmission of pain signals. Modulation of the nociceptive synaptic transmission in the spinal cord dorsal horn represents an important mechanism in the development and maintenance of different pathological pain states. This doctoral thesis has aimed to investigate and clarify some of the mechanisms involved in the modulation of the spinal nociceptive processing in different pain states. Modulation of synaptic transmission in the development of painful states

My thesis introduces the topic of nociceptive signalisation and processes involved in the formation and spreading of neuropathic pain. This study focuses on the mechanisms of nociceptive synaptic transmission mechanisms in the level of spinal dorsal horn and its modulation by paclitaxel, a chemotherapeutic drug inducing neuropathic changes. The attention is put especially on the possibility of glial activity participation in paclitaxel side effects. This idea stems from the existing hypothesis of the functional connection between TLR4 and TRPV1 receptor activity. TRPV1 is well known for its participation in chemical, thermal and nociceptive sensory transmission. Minocycline antibiotic is considered as an inhibitor of microglial activation therefore it was used for blocking neuroinflammation. The experimental part is comparing an impact of substances applied to the model of tachyphylaxis used for monitoring of nociceptive transmission changes according to decreasing activity of TRPV1 receptors. Electrophysiological recording of miniature excitatory postsynaptic currents from neurons in the Rexed laminae I. and II. of spinal dorsal horn was used. Effects of selected biologically active natural compounds on mammalian energy metabolism

Obesity is associated with many diseases, particularly type 2 diabetes, hypertension, dyslipidemia , hyperglycemia and cardiovascular diseases. If a patient experiences at least a trio of these diseases are diagnosed metabolic syndrome. The prevalence of obesity and related diseases has increased at an alarming rate in recent years and has become a worldwide health problem. Cardiovascular disease is the leading cause of death worldwide. Some minor components which are commonly found in our diet (eg some plant metabolites), directly or indirectly affect energy metabolism of our body and thus can reduce or even prevent developing health complications associated with the metabolic syndrome. This thesis, aims to clearly summarize the findings and results of scientific studies on selected plant metabolites occurring in extracts of specific plants that have preventive or therapeutic effects on obesity and other components of the metabolic syndrome. This work describes the use of these substances in the pharmaceutical industry and the food industry, the primary natural resources , and mechanisms of action , including effective doses. Modulatory mechanisms of nociceptive TRP channels

Detection of painful stimuli in the periphery is mediated by temperature-sensitive transient receptor potential (TRP) channels which are expressed in primary afferent endings of free sensory neurons called nociceptors. TRP channels in nociceptors are involved in the detection of thermal, but also mechanical and chemical stimuli. Out of seven known types of temperature-sensitive TRP channels, three are responsible for detecting painful temperatures: vanilloid receptors TRPV1 (> 42 o C) and TRPV2 (> 52 o C) detect noxious heat, and ankyrin receptor TRPA1 detects noxious cold (< 17 o C). Better knowledge of TRP channel mechanisms of action is essential for understanding TRP channel functions and ultimately for the design of potential analgesics. New findings presented in this thesis clarify mechanisms of action of TRPV1 and TRPA1 receptors, focusing on camphor and voltage sensitivity of TRPV1 channels and calcium modulation of TRPA1 channels. The first topic discussed in this thesis is the mechanism of camphor sensitivity of TRPV1 receptor. Camphor is a naturally occurring substance known since time immemorial for its effective analgesic properties, yet its mechanism of action is not understood.
