High cycle fatigue life of Ti6Al4V alloy produced by direct metal laser sintering

High cycle fatigue life of Ti6Al4V alloy specimens manufactured by Direct Metal Laser Sintering (DMLS) was experimentally determined. The DMLS fabrication process was characterized by a 400 W laser power and 50 micrometer layer melted thickness. Post-fabrication heat treatment consisted in stress relieving for 3 h at 720 °C in vacuum with subsequent cooling in argon atmosphere. Fatigue testing of specimens oriented in three different directions with respect to the material build direction was performed with the aim to examine the influence of the layered microstructure on the fatigue behavior. Results of measurement of surface roughness, metallographic examinations of the layered material and fractographic investigation of the fatigue fracture surfaces were employed in the discussion of fatigue crack initiation in DMLS fabricated Ti6Al4V alloy.
Difficult temperament in early childhood

The presented dissertation consists of both a quantitative part and a qualitative part. The quantitative part deals with the relationship between 5-HTTLPR S/L, MAOA H/L, and COMT Val158Met polymorphisms, the stress reaction of new-born infants after a heel stick blood draw (measured by determining salivary cortisol at three time points) and temperament assessed at age three months by Rothbarth's Infant Behavior Questionnaire-Revised in a sample of 84 infants. Observed polymorphisms were related both to the course of the stress reaction and to temperament. The short allele of serotonin transporter polymorphism was connected to higher scores in the secondary scale of Negative Affect and lower scores in the secondary scale of Attention/Regulation. Homozygotes for the more active allele of MAOA polymorphism (HH) had the lowest scores in Negative Affect compared to both of the remaining groups, they also had higher scores in the secondary scale of Extraversion and Attention/Regulation and a greater decrease of cortisol in comparison to HL heterozygotes. The presence of low-active L allele predisposed their carriers to higher scores in Negative Affect and lower scores in Attention/Regulation. LL homozygotes had the highest increase of cortisol after a heel stick blood draw. The Met allele of COMT Val158Met polymorphism was connected to higher Extraversion and Attention/Regulation and a greater cortisol decrease. It was possible to predict all three secondary scales of IBQ-R from the stress reaction after the heel stick blood draw. Negative Affect was predicted by a higher increase and a lower decrease of cortisol. Extraversion and Attention/Regulation were predicted by a greater cortisol decrease. The magnitude of cortisol decrease partially mediated the influence of COMT Val158Met polymorphism on Extraversion. The qualitative part of the dissertation is a multi-casuistic study of six couples parenting infants with difficult temperaments. It is based on semi-structured interviews that were analysed in accordance with qualitative procedures. The most difficult infant displays to manage were unsoothable crying in the first six months and early sleeplessness and a later escalation of sleeping problems. Mothers were esentially not able to gain control over the amount of crying, but some of them managed to influence their experience to achieve a greater acceptance of it. To do this, it was necessary for them to eliminate their feelings of failure in the parental role. The parents' biggest dilemma concerning their infants' sleeping problems was whether to use the "cry it out" strategy or not to manage them. For some parents parenting a difficult infant was an opportunity to re-evaluate their approach to parenting and the parental role, significantly broadening the concept of both. Caring for a difficult infant significantly strained the marital relationship; four couples experienced marital crisis during the care of their child. The father's involvement in infant care seemed very important in this respect. Insufficient involvement led to dissatisfaction in the mother, the way the mother communicated her demands further influenced the marital relationship. Particular behaviour that the mother understood as the father's involvement in infant care emerged.
Alkaloids of Papaver rhoeas L. (Papaveraceae) and their biological activity related to Alzheimerʼs disease II.

Čakurdová, M.: Alkaloids of Papaver rhoeas L. (Papaveraceae) and their biological activity related to Alzheimer's disease II. Diploma thesis, Charles University in Prague, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany and Ecology, Hradec Králové, 2017. The aim of this work was focused on isolation of alkaloids from the fraction 1 (joined fractions 20-27), which was obtained from the summary alkaloid extract of Papaver rhoeas L. (Papaveraceae). The alkaloids were identified by structural analysis NMR, GC-MS, optical activity and melting point. Two alkaloids were identified as (-)-stylopine and (+)-rhoeadine. Isolated substances were tested on ability to inhibit the enzymes acetylcholinesterase, butyrylcholinesterase and prolyloligopeptidase. Obtained data were expressed as IC50 values: (-)-stylopine (IC50 AChE = 522 ± 67 µM, IC50 BuChE = >1000 µM, IC50 POP = >790 µM); (+)-rhoeadine (IC50 AChE = 915 ± 64 µM, IC50 BuChE = >1000 µM, IC50 POP = >790 µM). None of the isolated substances showed so potent cholinesterase inhibitory activity as the alkaloids galanthamine (IC50 AChE = 1.71± 0.065 µM, IC50 BuChE =42.30 ± 1.30 µM) and huperzine A (IC50 AChE = 0.033 ± 0.001 µM, IC50 BuChE = >1000 µM) or rivastigmine (IC50 AChE = 0.037 ± 0.001 µM, IC50 BuChE = 3.3 ± 0.3 nM)....

The principle of the reaction is displacement of aluminum ions from a complex formed by reaction of aluminum chloride solution with aluminon (ammonium salt of aurintricarboxylic acid) and formation of a complex of aluminon and fluorides. During the reaction the solution changes its color which is caused by changes in absorbance of the solution with fluorides and of the blank solution. The absorption spectrums of both the solutions were measures at the wavelength of 522 nm. Initially, the reaction conditions were optimized in a test tube, where we found linear dependence of the color change on concentration of fluorides, and the ratio of the added agents was optimized. The role of a pupil and social ASPECTS of the school physical education lessons. Aim: The thesis deals with the social dimension and the role of the student in school physical education. The social dimension is particularly focused on the relationship between individual pupils and between student and teacher. In an additional dimension to the role of pupil detects changes position of the pupil as an individual in an hour, the possibilities of active participation and decisions. The study group are primary school pupil, which have been teached two different clocks. Habitual hour (Traditional), to which they were accustomed pupil teachers and two o'clock hour was progressive (creative), more focused on the student, his autonomy, participation in management, self-esteem but also his own choice of difficulty Exercises like. Methodology: The reference is Sobor schoolgirl fourth to ninth grades (522 female students) elementary Schools in Prague that we have a better comparison divided into two groups: 4 to 6 Class (261 žákyň) and 7 to 9 Class (261 female students). Production of connector

The diploma thesis deal with the proposal of connector manufacturing from bronze sheet CuSn6, thickness 1.5mm in production volume of 1.600.000 parts per year. From considered manufacturing variants there was chosen shearing and forming technology of thread in the progressive stamping die. For part production was selected pressing machine Bruderer 1250 with nominal pressing force 1250 kN from company E. Bruderer Maschinenfabrik AG. In the economical evaluation there were determined expenses 21.6 Kč per one piece including demanded profit. The turning point was calculated at production of 165.522 parts.
Environmental Aspects of Pyrotechnic Mixtures.

The submitted bachelor thesis deals with the study of pyrotechnic compositions and their influence on the health and environment. It explains the nature and ingredients used in pyrotechnic compositions. During burning these compositions release different compounds, toxic or non-toxic into the atmosphere in large quantities. In this work the issue of toxicity of selected alkali, alkaline earth metals and the other harmful metals was explored. The pyrotechnic compositions containing Ba, As, Pb and Cd were investigated in more detail. When burning the pyrotechnic compositions release noxious substances into the air first. Then these substances reach due to wet or dry transport mechanism the other components of the environment and mix with them. These compounds have diverse negative effects both on humans and the environment and other animals.
Creating a business plan

The goal of this bachelor thesis relates to issues in pre-school education in the Czech Republic. The work comprises a business plan which reacts to the present gap in the market concerning the given social sector, caus ed by the insufficient number of the government-run child care facilities. The overview of literature used in drawing this bac helor thesis comprises spe- cialized literature and laws concerning the issues in this case. The selected information has been summed up and used in drawing the text of this thesis. It will also support the practical part of the work. The sources for the selection of the needed data an d information will be both primary and secondary materials. The primary source represents specialized literature, information obtained from the internet and public d ata provided by the governance and government institutions such as the Magistrate of t he city of Č eské Bud ě jovice, the Dis- trict Health Department, the building and construct ion authorities, etc. The primary information will be collected by means of a public survey by filling in the prepared questionnaire. The main idea underlying the project and the busine ss plan is the creation of a pre-school facility named Child care centre RADOS T (JOY). Children of the age of 3 to 6 years will be admitted to attend the centre. T he conception of this child-care centre is the individual approach towards education leadin g to a healthy lifestyle and overall health.
Education of women with functional disorders of the motion system to the physiological activity of the pelvic muscles using physiotherapy practice

This work deals with pelvic floor muscle issues. The topic is Educating woman with functional disorders of the locomotor system for the physiological activity of the pelvic floor using physiotherapy techniques. The first part focuses on the anatomical relationships in the pelvic area in terms of single organs, breathing mechanics, pelvic floor work and abdominal musculature. Further chapters are about pregnancy, childbirth and the postpartum period in relation with pelvic bottom and dysfunction of the pelvic floor mechanism. Theory of functional disorders has been explained as a result of the changes woman's body goes through. The aim of the theoretical part was to summarize knowledge about the function of the pelvic floor, dysfunction mechanism and the possibility of a therapeutic effect on functional disorders of the musculoskeletal system. The second part has developed the methodology and therapeutic procedures for two research files with respect to PF activity as part of the deep stabilization system. The first group were women before childbirth in the 2nd and 3rd trimester of their pregnancy; the second group consisted of women after childbirth - during the postpartum period. Characteristic for both research files were the presence of functional disorders of the musculoskeletal system. For the research were used qualitative methods of data acquisition; case history, observation, input and output kinesiology analysis and interview. The main aim of this work is to determine the rehabilitation plan for women in the 2nd and 3rd trimester of pregnancy and the determination of the rehabilitation plan for women in childbed. Both rehabilitation plans contain special set of PF exercises, which include education for their proper implementation. Selective clients have opportunity to insight into the understanding of this muscle group work, not only useful during exercises, but even during every day activities. The Bachelor thesis can be used in physiotherapists' clinical practice. Also, as a source of information for professionals and the general public.
