Isolation of yeasts from the vineyard and their using in the production of wine

The aim of this thesis was the isolation and identification of yests obtained from the wine berries by different molecular methods. The yeasts for identification were collected from the species of Veltlínské zelené white wine from the wineyard from Milotice, district Hodonín. Identification based on analysis DNA by PCR method and followed by RFLP using restriction enzymes HaeIII, HinfI, HhaI a TaqI. To obtain values lenghts of the restriction fragments was used gel electrophoresis. For getting genetic similarity of analyzed yeasts was used BioNumerics software, that processe the results using cluster analysis using Jaccard´s coefficients and the yeasts were identified by comparing the characteristic lenghts with number of articles and educational databases.
Validation of PCR-RFLP method for determination of lactose intolerance

The lactose intolerance is the most common food intolerance occurring worldwide. Individuals having the lactose intolerance cannot produce the laktase enzyme in thein small intestine. This enzyme is able to digest the lactose contained in milk. There are several wals for this disabilitty. The practical part of this bachaleor thesis is focused on the genetic origin of the lactose intolerance. In the population of Europe we can find as the most frequent the two polymorphism of the MCM6 gene that affect the production of laktase C/T13910 and G/A22018. In principle it concerns the new gene station which ensure the production of laktase enzyme even in adult age. The aim of this bachaleor thesis is to creat the research based on the recent knowledge Publisher in the specialized literature. There are gin to be describe all type sof the lactose intolerance and the ways of its diagnostics, futhermore e.g. the princupli of Alleny, the used laboratoř methods, the rule sof validation methods etc.. The pros and cons of every testing approach will be explained. The practical part is aiming at the implementation of the validation of the genetics test used for petting down the lactose intolerance and creating the validation protokol that will serve as a base of my bachaleor thesis. The genetic Cheb-up is based on the PCR-RFLP principle, within the metodology the laboratoř procedures being used in the genetic lab will be described. The validaton will flow into a detection if the applied method is sufficiently valid fot the diagnostics. The genetic analysis will focus on the two selected polymorphism C/T13910 and G/A22018. The sample with a familiar genotype that has already been verifie by sekvenation will be used for validation.
Rezistence genes against fungi deseases in cereals and methods for their detection

The paper deals with the possibilities of detection fungal disease resistance genes in the original European cereals, such as wheat, barley, rye and oats. It provides an overview of the most important resistance genes for breeding. The significance of the resistance genes is evaluated on the basis of the harmfulness of the individual cereal fungal diseases, as well as the importance of the individual cereal species. It describes the possibilities of detecting resistance genes using molecular markers and compares various types of molecular markers. It seeks answers to the question of where the breeding on resistance and therefore the use of molecular markers brings the greatest benefit.
Gene NQO1 polymorphism analysis using the PCR method

This bachelor thesis deals with the detection of polymorphism of the NQO1 gene by PCR-RFLP methods. The thesis describes the exact methodology of detection of polymorphism commonly used in the clinical laboratory. The methodology including DNA isolation, determination of DNA concentration, PCR, RFLP, and electrophoresis. The detection was performed in 120 patients in genetic laboratory GENLABS s.r.o. in České Budějovice. The results of the analysis were evaluated and compared with the literature.
Restrikční endonukleázy a genetické testy

This bachelor thesis Restriction endonucleases and genetic tests engages in the issue of restriction enzymes. In the literary review I describe the character of restriction endonucleases, their classification and mechanism of operation. The thesis furthermore touches a topic concerning the effect of the restriction reaction conditions and discusses techniques of utilization of these enzymes, where for example DNA cloning, analysis of variability or genome mapping belong. The practical part of the thesis includes some work with a software for an automatic search for restriction sites and for restriction DNA evaluation.
Aldosterone synthase in arterial hypertension and possible influence of its genenetic polymorphism on left ventricular hypertrophy

Part I. The aldosterone synthase gene (CYP11B2) polymorphism T-344C in blood pressure and left ventricular hypertrophy. BACKGROUND: Aldosterone is a key cardovascular hormone, it significantly influences volume, pressure and electrolyte balance. Aldosterone plays an important role in development of left ventricular (LV) hypertrophy and myocardial fibrosis. The aldosterone synthase gene (CYP11B2) is an important candidate gene region in essential hypertension. DESIGN AND METHODS: We assessed the influence of the T-344C polymorphism of aldosterone synthase - the rate-limiting enzyme in aldosterone biosynthesis - on the structure of the left ventricle in young normotensive men. The population included 113 normotensive mid-European Caucasian men aged 18-40 years (mean 27 +/- 5 years). We also studied the association of -344T/C polymorphism of the CYP11B2 gene with the presence and severity of hypertension in 369 individuals, of whom 213 were hypertensive patients (139 controlled hypertensive, 74 resistant hypertensive) and 156 were healthy normotensive subjects. Studium SNP genu DGAT1 jako kandidátního genu pro kvalitu vepřového masa

This work was focused on the study of variability DGAT1 gene in the pig population Czech Large White breed. Subsequently, association analysis was performed of the gene and the different production indicators of quality of pork meat. Examined polymorphism was found at position 103 of intron 2 of chromosome 4. DGAT1 gene has a major role in the synthesis tryacylglycerols and may affect the storage of fat in the body. Relative genotype frequencies were: AA = 0.4222; AG = 0.4889; GG = 0.0889. The values of the relative frequencies of alleles were as follows: A = 0.6666 and G = 0.3334. The association analysis didn't show any statistically significant differences.

This diploma thesis follows the bachelor thesis and it?s task is to processing analysis of the occurrence of PSE meat in a population of the Czech white breed as a pigs stress syndrome impact. General problem of congenital diseases is processed in literature review. Further, this work deals with pigs stress syndrome and stress, which causes the meat defects as an external factor. Molecular - genetic methods, which are used in the practical part, are described as well. The research focused on genotyping of loci selected panel of samples, to determine the frequency of alleles and genotypes of selected locus and the statistical relationship between genotype and expression of PSE meat. Results were statistically processed and evaluated at the end.
Methods of detection of wheat and barley strain of Wheat dwarf virus in vector Psammotettix alienus by PCR-RFLP

Wheat dwarf (causal agent Wheat Dwarf Virus - WDV) is one of the most important cereal diseases in the Czech republic where it can cause huge economic losses in wheat, barley, triticale, rye as well as in oat. Psammotettix alienus is the only known vector of WDV while another way of transmission is unrecognized. This methodology describes a PCR assay for sensitive detection of Wheat Dwarf Virus in Psammotettix alienus, and RFLP assay for barley and wheat strains specific identification.
