Ionic currents recorded in voltage clamp experiments on enzymatically isolated rat ventricular myocytes were analyzed with the aim to study the voltage and frequency dependence of sodium current block induced by two antiarrhythmic drugs: ajmaline and propafenone. The levels of block produced by both examined agents were clearly related to membrane voltage. Interestingly, ajmaline, in contrast to propafenone, did not show frequency and use dependent effect. These differences are analyzed.

This study was aimed to analyze the effects of the antiarrhythmic drug ajmaline (a known inhibitor of the fast sodium current INa) on the potassium currents, namely the transient outward current (Ito) and the current measured at the end of a depolarizing pulse (IK) in rat ventricular myocytes. Both currents were inhibited in concentration- and voltage-dependent manner. However, the effects were frequency-independent.
