Poláková, Magdaléna ; Rybářová, Kateřina (advisor) ; Tichá, Zuzana (referee)
Title: Pilot Usability Testing of the Box and Block Test in Occupational Therapy by People after Stroke: Creating a Video Manual Abstract: Stroke often results in impaired upper limb function. Occupational therapists use standardized tests to objectively assess hand dexterity. An example is Box and Block Test (BBT), which assesses mainly fine and gross motor skills. The tested subject's task is to sequentially move as many blocks as possible from one side of the test box over partition to the other side using only the tested limb within one minute. For this test, only working versions of Czech translations of instructions by Mathiowetz et al. from 1985 were used in the Czech Republic. Therefore, in 2021 Rybářová et al. created Extended Czech Version of Manual for BBT with instructions in Czech. It also contains new more detailed rules that should ensure greater objectivity in the standardised implementation of BBT. According to these rules, it is possible to evaluate, for example, the situation when block falls outside the test box. The main goal of the bachelor thesis was to create video manual for correct administration of BBT according to the new manual. Therefore, video documentation was obtained using two healthy subjects as well as the author of this thesis. The video manual was then created in...
Evaluation of executive functions in patients with acquired brain damage: translation and clinical use of the "Executive Function Performance Test"
Pepřová, Tereza ; Krivošíková, Mária (advisor) ; Tichá, Zuzana (referee)
Title: Evaluation of executive functions in patients with acquired brain damage: translation and clinical use of the "Executive Function Performance Test" Abstract: Patients with acquired brain damage that have affected the frontal lobes usually develop a deficit in executive function (Kulišťák, 2017). Within self-care activities, they are limited for example in initiation, organization, sequencing, emotional control, memory, or self-evaluation. Occupational therapists can use a range of standardized tests to describe the extent of the deficit and select an appropriate rehabilitation. Executive Function Performance Test, which I used in my Bachelor's thesis, is one test from this category. The aim of the Bachelor thesis was the clinical evaluation of executive functions in patients with acquired brain damage using the Executive Function Performance Test. The minor goal was the one-way translation of the Executive Function Performance Test to the Czech language and the subsequent adaptation of the tasks for Czech patients. Furthermore, the Bachelor thesis contains a description of screening tests, clinical tests, and other standardized tests that are evaluating executive function. As well as a description of the executive functions in healthy adults. The practical part of the thesis contains a description of...
Alterations in protein expression induced by valproic acid in human T-lymphocyte cell line MOLT-4
Tichá, Zuzana ; Kvasničková, Eva (advisor) ; Řezáčová, Martina (referee)
Univerzita Karlova Farmaceutická fakulta Katedra biochemických věd EFFECT OF VALPROIC ACID (VA) ON CHANGES IN EXPRESSION OF PROTEINS OF HUMAN T-LYMPHOCYTE LEUKEMIA CELL LINE MOLT-4 Valproic acid (VA) is histondeacetylase inhibitor (HDACI) and it leads to inhibition in proliferation and apoptosis induction during incubation with leukemia cells. The aim of our article was to determine the changes in expression of proteins after incubation leukemia cells with VA. After 4-hours-long incubation of MOLT-4 cells with 2 and 4 mmol/l VA, the nuclear extracts from MOLT-4 cells were used to detection of changes in levels of proteins. The nuclear extracts were separated for first dimension-IEF in immobilized pH 4-7 gradient. Immediately after being focused, the proteins were separated for electophoretic way in the second dimension, 9-16% gradient DS was used. Proteins separated by 2D electrophoresis were visualized by silver staining , and silver-stained gels were digitized using a laser personal densitometer. Protein maps were quantified using a Melanie 3 software package. Proteins were identified by MALDI mass spectrometer. The finding protein process was finished by using ProFound and PeptIdent programs. Identification process of protein spots exhibiting significant abundance alterations induced by VA help to...
Rozložitelnost plastů v aerobním a anaerobním prostředí
Tichá, Zuzana
The increasing use of plastics in our modern society, which are used in many sectors of a human activity, caused the need of material development which will be friendly to the environment, so the topic of the thesis is aimed at this area and deals with the degradability of plastics in aerobic and anaerobic environment. The main aim of this work is to verify the possibility of degradation of biodegradable plastics made from material known as the brand name Mater - Bi, which is a mixture of the bioplastic that is made from maize starch, in the condition of aerobic and anaerobic environment. Testing of the plastics biodegradability consists of the test series in laboratory and pilot and operational tests. All tests were chosen with regard to the plastic cycle in the environment, its distribution possibilities and potential impact on the environment. In laboratory tests disintegration of the test material in terms of the composting process, assessing of the earthworms escape behaviour, germination of lettuce or black soldier living conditions in the environment with degradable plastics and assessing the process of anaerobic fermentation with the degradable plastics are evaluated. In the pilot tests are examined conditions of the domestic composting in the small piles and operational tests include testing of the plastic material in the composting process at the composting plant. In the first part of the thesis are defined theoretical knowledge of plastics and their degradation, aerobic process of composting and anaerobic fermentation. The second part focuses on the description of the experiments and their methodology. In the final part of the thesis the test results are evaluated and assessed and with respect to the obtained knowledge the design and development of the methodology for the plastic use and recommendations for practice are described.
Senior's View on the Use of Information and Communication Technologies in Practical Life
TICHÁ, Zuzana
The development of the imformation and communication technologies is at the contemporary time a very actual theme. The object of this bachelor work is to find out the senior´s taking of the information and communication technologies and their using in the practical life. The theoretical part of this work is formed by three basicly chapters, concerning the problems of the age, growing old and defining of the therm senior. Also are they intending on the senior´s education, going nowadays forward, involving practiculary the Virtually university of the third age. The last chapter is intending on the information and communication technologies, on its characterication and the senior´s reference to these technologies. In the empiric part of the work occures the work destination, that has to find out, what is the senior´s way to the information and communication technologies the main research question: "What way are the seniors to identify with the using of information and communication technologies?" To get the most exact answere of the main research question with the next secondary questions were the ground for the interview. Like the best for this acivity was chosen the questions research strategy. To get the data helped the asking method and the fast phase was the ahead research and then the proper one. The research group formed the students of the Virtually University of the third age, elected by designed quota election. From the research resulted the good senior´s orientation concernant the information and communication technologies terms and they are using all kinds of it daily. They accept the advantage of the open informations, of the communication and the rate of speed, which these technologies are desposing, on the other side the seniors are speaking about the disadvantage of the social isolation. Nowadays they want to keep these technologies because they are standing for them the certain relieve in their life, but they don´t want to prolonge the study because the také their knowledge for safficient of this domain.
Documentary piece 2.0 and its dominant features regarding the viewer
Tichá, Zuzana ; KOPECKÝ, Lubor (advisor) ; VLACH, Jiří (referee)
This thesis deals with documentary pieces 2.0 Its first and second part specifies the role of this term in the context of modern documentary formats, which are gaining in popularity lately but less so for theorization. The subject of the first part of this paper is to explain what the term documentary piece 2.0 actually means, to describe the principles upon which it works and in what ways it is possible to divide it. The third part analyzes the dominant features of documentary pieces 2.0 and demonstrates them with specific examples of projects. Features which this thesis describes are: mosaic arrangement, collage concept, the possibility of exploration, investigative approach, subjective view and viewer usage data. The aim is therefore to specify dominant features, which documentary pieces 2.0 carry within themselves, and to describe through them how does this whole modern production works with the audience.
