Cell Viability Measurement

This work discusses the viability of cells. It describes the principle of fluorescence, fluorescent methods, fluorescent microskopy and the method of dyeing of the cells with a special kit that contains calcein AM and ethidium homodimer dyes. Further it describes the program for analysis and evaluation of viability on two different cellular cultures.
Study of fluorescence lifetime and spectral changes of nanoparticles in cell biology

This work deals with the study of fluorescence lifetime and spectral changes of nanoparticles in cell biology. It describes the principle of fluorescence, fluorescence microscopy and laser confocal microscope Leica TCS SP8. The classic FLIM method, the Lambda Square mapping and the division of nanoparticles are introduced there. In the practical part, the created program for the evaluation of fluorescence lifetime and spectral changes is described. The program can show two-dimensional lambda maps, the fluorescence lifetime and spectral shift in the space area. In the final part of the thesis, an experiment with rhodamine nanoparticles is carried out and it is evaluated using the created program and then discussed.

Malé a střední podniky v oblasti celnictví do vstupu České republiky do Evropské unie a po vstupu do Evropské unie

Práce analyzuje a hodnotí oblast celnictví před vstupem ČR do Evropské unie a po jejím vstupu, se zaměřením na problematiku provozování celních skladů. V oblasti spotřebních daní se práce věnuje rozsáhlé problematice vydávání povolení souvisejících s činnostmi MSP a dopravě vybraných výrobků v režimu podmíněného osvobození od daně. Bez vydání příslušných povolení není možné ve specifické oblasti spotřebních daní provozovat podnik na výrobu vybraných výrobků. Další povolení, udělovaná celními orgány, jsou rozhodující pro činnosti podniků jak při výrobě, tak obchodování s vybranými výrobky.
