v anglickém jazyce Charles University Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Student: Anežka Sladká Supervisor: Mgr. Pavel Bárta, Ph.D. Thesis title: In vitro saturation study of radiolabeled monoclonal antibody carotuximab Angiogenesis, or the formation of new blood vessels from existing ones, is currently one of the processes targeted by cancer therapy, as the formation of new blood vessels is a prerequisite for successful tumor growth and eventual expansion in the body in the form of metastasis. One of the receptors involved in signalling leading to angiogenesis is endoglin (CD105). This receptor is also targeted by the chimeric monoclonal antibody carotuximab. The aim of this thesis was to perform indirect labelling of the monoclonal antibody carotuximab with the radionuclide gallium-67 using the bifunctional chelator 1,4,7-triazacyclononane,1-glutaric acid-4,7-acetic acid (NODAGA). During the labelling process, the NODAGA:carotuximab molar ratio was optimized (1:20, 1:50 and 1:100). The prepared radiopharmaceutical [67 Ga]Ga-NODAGA-carotuximab was subsequently determined for its radiochemical purity by HPLC with radiometric detection. This was followed by in vitro binding experiments on human renal tumor cell culture involving internalization,...
Comparison of radiolabelled fatty acid (18F-FTHA) and 18F-FDG in imaging of brown adipose tissue

Charles University in Prague Faculty of Pharmacy in Hradec Králové, Department of Pharmacology and Toxicology Student: Mgr. Tereza Vašků Supervisor: Dr. Matthias Bauwens: Maastricht University, Netherlands Consultant: PharmDr. Jana Ramos Mandíková, PhD Title of diploma thesis: Comparison of radiolabelled fatty acid (18 F-FTHA) and 18 F-FDG in imaging of brown adipose tissue Brown adipose tissue (BAT) is highly metabolically active tissue, which consumes glucose and free fatty acids (FFA) during the process called thermogenesis. Due to these characteristic features, it is possible to quantify the activity of the BAT by non-invasive imaging methods (by using radiopharmaceuticals). Nowadays, one of the most frequently used substances is the radiopharmaceutical called 18 F -FDG (radiolabelled glucose by fluoride). The 18 F -FDG is in clinical practice used for metabolically active tissues diagnosis, notably tumours. We focused in this study on synthesis of radiolabelled fatty acid, namely on the radiopharmaceutical 14(R,S)-[ 18 F]Fluoro-6-thia-heptadecanoic acid (18 F -FTHA). Fluor-labelled fatty acid is used notably for myocardial metabolism observation. The goal of the thesis was a synthesis of radiopharmaceutical 18 F -FTHA using a semimanual module in an environment of sufficient purity and yield....

Charles University in Prague, Faculty of Pharmacy in Hradec Kralove Department: Department of Pharmacology and Toxicology Candidate: Zbynek Novy, MSc. Supervisor: Assoc. prof. PharmDr. Frantisek Trejtnar, PhD. Title of doctoral thesis: In vitro study of interactions of radiolabeled monoclonal antibodies with epidermal growth factor receptor Various types of cancer are characterized by overexpression of epidermal growth factor receptor (EGFR) compared to normal tissue. Anti-EGFR antibodies utilize this phenomenon to treat certain oncological diseases. Intensively studied field is verifing of possibility to increase usability of these antibodies by incorporating suitable radionuclides into thiers structures, results can serve in radioimmunodiagnosis or radioimmunotherapy of mentioned malignities. Despite the fact that EGFR recepor and his signaling intracelluar pahtway is object of intense research, there is a lack of knowledge in the area of factors, which can significantly affect binding of radiolabeled antibodies to EGFR in vitro. The aim of this doctoral thesis was to introduce method of radiolabeling anti-EGFR monoclonal antibodies (cetuximab, panitumumab) with iodine-131 and to use this radiolabeled antibodies to determine expression rate of EGFR in four selected cell lines (A431, HaCaT, HCT116...
Parasitostatus of red deer bred in selected overwintering enclosures in Eagle Mountains
Nový, Zbyněk ; Lamka, Jiří (advisor) ; Ducháček, Lubomír (referee)
TTHHEE AABBSSTTRRAACCTT The thesis: Parasitostatus of red deer bred in selected overwintering enclosures in Eagle Mountains Made by: Zbyněk Nový The thesis describes qualitative and quantitative parasitostatus of lungworms at red deer populations bred in three overwintering enclosures of Eagle Mountains. The effectivity of therapeutical intervention was calculated from gained data. The treatment efficacy was 80 - 100 %. Larvoscopic examination showed presence of three lungworm species (Elaphostrongylus cervi, Dictyocaulus viviparius a Varestrongylus sagitatus), its extensity of appearance is changing during longterm monitoring.
