Municipal Services Building in Žďárná

The master’s thesis deals with the design of Municipal services building in Žďárná. The building is located in the center of the village Žďárná in the South Moravian region. The building is set in a sloping terrain. Municipal services building is divided into 2 buildings: SO.01 Municipal building and SO.02 Health Center. SO.01 Municipal services building can be divided into two parts – the first part is two-storey with partial basement and it is covered by sloping roof made of wooden trusses. The second part is single storey with partial basement and it is covered with flat roof. SO.02 Health Center has one floor and it is roofed with a flat roof. The building SO.01 Municipal services building is functionally divided into 4 parts – 01 municipal office, 02 hairdresser, 03 post office and 04 garage. The building SO.02 Health Center is functionally divided into 2 parts – 01 pharmacy and 02 doctor’s office. The building is designed as masonry construction. Horizontal load-bearing structure is made of monolithic reinforced concrete ceiling. Each floors and connected by reinforced concrete stairs and by an elevator. The building SO.01 Municipal services building is insulated with contact thermal insulation system. The building SO.02 Health Center has ventilated façade, the insulation is made of mineral wool boards and the cladding is made of compact HPL laminate boards.
Apartment building in Boskovice

The bachelor’s thesis deals with the design of a new apartment building located in Boskovice. The apartment building is designed as a detached building set in northeast part of the city Boskovice between existing surrouding buildings of detached houses and apartment buildings. The aparment bulding is set in a moderately sloping terrain. The apartment building has a basement with three floors and it is roofed with a flat roof. The main entrance to the bulding is from the south side and it has front door steps. There is a total of 7 dwelling units in the above ground floors which sizes are 1+KK, 2+KK and 3+KK. In the basement are located Technical facilities and storage space. The building is designed as mansory construction. Horizontal load-bearing structure is made of monolithic reinforced conctere ceiling. The individual floors are connected by a half-turn reinforced conctere stairs and by a lift. The building is insulated with contact thermal insulation system.
Capillary monolithic columns based on copolymer of styrene, divinylbenzene and methacrylic acid and their application for separations of small molecules.

This work is focused on the preparation and characterization of capillary monolithic columns based on a copolymer of styrene, divinylbenzene and methacrylic acid for the chromatographic separation of small molecules. The capillary monolithic columns were prepared in the quartz capillaries by one-step radical copolymerization reaction of monomers of styrene, divinylbenzene and methacrylic acid in the presence of toluene and isooctane as porogenic solvents and azobisisobutyronitrile as an initiator. The effect of incorporation of methacrylic acid into the polymerization mixture on the separation behavior and efficiency in reversed phase capillary liquid chromatography (CLC) and capillary liquid electrochromatography (CEC) was studied using a test mixture of compounds containing thiourea, phenol, aniline, benzene, toluene, ethylbenzene, propylbenzene and butylbenzene. The Walters test for reversed stationary phases was employed to evaluate the hydrophobicity of the prepared columns. Based on the calculated value of hydrophobicity index of 10.76, the prepared monolithic columns can be classified as very hydrophobic. Isolation and characterisation of biologically active substances

This Bachelor thesis aims at isolation and partial identification of biologically active substances which are produced by actinomycetes and can be potentially applied in medicine. Cultivation broths of actinomycetes containing their metabolites were purified and pre-concentrated by solid phase extraction. Then, the bioassay of the extracts by Kirby-Bauer test using the sensitive strain Kocuria rhizophila was performed. Biologically active metabolites were analyzed and isolated by ultra- performance liquid chromatography with photo diode array detector. Isolated substances were assayed by mass spectrometry, which yielded relative molecular mass values of the unknown compounds. The values were compared with relative molecular masses of compounds listed in a chemical database, which involves natural products including antibiotics. We revealed that the unknown biologically active substances do not refer to any already discovered compound present in the database suggesting that the unknown compounds may be novel. More mass spectrometry and nuclear resonance experiments have to be carried out in order to elucidate their structure.
