Sulfation and desulfation of neuroactive steroids in normal and neoplastic primate brain tissues

A novel methodology for the determination of steroid sulfatase (STS) and sulfotransferase (SULT) activities in primate brain tissue have been developed and evaluated. STS activity determination was based on enzyme assay followed by liquid-liquid extraction ofthe reaction product, its subsequent derivatization and gas chromatography-mass spectrometry analysis. Indirect determination including product hydrolysis and determination by radioimmunoassay was used for assessment of SULT activity. For both, STS and SULT assays, optimal reaction conditions were found while strong structure-activity relationships for both enzyme activities were observed. Additionally, so far unidentified high-substrate inhibition for STS was revealed wíth a hypothesis suggesting its physiotogícal relevance' Determinations of STS and SULT activities in monkey brain regions revealed differences between genders and brain regions. Due to low number of samples, significance was not described. For STS kinetic characteristics were determined. In human brain tumors, STS and SULT activities showed highly significant differences according to diagnoses. Furthermore, tendency to cluster formation with respect to both enzyme activities emerged. Implementation of project management in IT company

This thesis deals with the implementation of project management in an IT company from the sector of Small and Medium Enterprise. Its goal is to prepare the project management framework based on the robust methodics PMBoK, by which the company can begin to perform its business cases by using projects. The minor goals are the clear definition of project management and the summary of PMBoK and its features customized for the SME sector. This thesis consists of three parts: first one defines the basic terminology related to project management and includes the selection of the ideal organizational structure for project management in smaller company. Next part analyses the methodics PMBoK and it is used to derive a customized methodics for Small and Medium Enterprises.This methodics is then described more in detail in the last part with emphasis on process description. The main contribution of this thesis lies in creation of customized methodics of project management, which will be used in real life. Next minor contribution is the process of customization of PMBoK for SME.

Zhodnocení současného stavu využívání systémů pro podporu rozhodování ve státní správě

Tato práce pojednává o systémech pro podporu rozhodování ve státní správě. Práce obsahuje teoretické zhodnocení oblasti systémů pro podporu rozhodování a problematiky české státní správy s přihlédnutím na informatický pohled a roli Ministersva informatiky. V praktické části je zahrnuto zmapování využívaných systémů pro podporu rozhodování v ústředních správních úřadech a dále tvorba metriky s její následnou aplikací na konkrétních příkladech. Na závěr je vytvořeno zhodnocení s praktickými doporučeními pro další rozvoj systémů pro podporu rozhodování ve státní správě.
