Energy metabolism and enzymatic activity in the Ips typographus in relation to diapause.

The thesis describes the development and survival of immature Ips typographus specimens at low temperatures under laboratory and field conditions. Further, the focus was identifying and characterizing the digestive enzymes present in the gut of adult I. typographus, their location in the gut and enzymatic fluctuation over a full calendar year, with a specific focus on digestion of cellulose.

My work concentrates on feeding behavior of overwintering diapausing and post {--} diapausing bark beetles and developmental treshold. This is done either biochemically by measuring the enzymatic activity in the midgut and by assessing the feeding status from the size and consistence of the food bolus in the gut. Detailed knowledge of feeding behaviour and development treshold may help to predict the overwintering success of local populations with all the consequencies for spring dispersal and reproduction.

The project broadens our knowledge on ecophysiology of the spruce bark beetle, mainly in relation to practical application of the results. As a main practical result of the project, several alterations that improved the effectiveness of currently applied protective measures have been proposed, tested and accepted by the headquarters of the Forests of the Czech Republic, state enterprises.

Effect of 7-oxo-dehydroepiandrosterone on oxygen consumption of human PBMC

Effect of 7-oxo-dehydroepiandrosterone (7-oxo-DHEA) on resting and noradrenaline stimulated oxygen consumption of human peripheral blood mononuclear cells (PBMC), isolated by gradient centrifugation, was studied using Clark oxygen electrode. It was found that, the 7-oxo-DHEA in concentration 10 umol/l has a thermogenic effect and increase resting metabolic rate of PBMC by more than 80 %. Oxygen consumption stimulated by noradrenaline was potentiated by 7-oxo-DHEA in concentration 0,1 umol/l by 39 %.
