Title: Case study of a patient with the diagnosis of burns II.-III. grade to 40% of body Objectives: The main goal of this bachelor thesis is to work out a detailed case study of rehabilitation care of a patient with burns and literature search of this topic. Another goal is to acquire comprehensive teoretic and practical knowledge about this diagnosis. Methods: I used assay and physiotherapeutic approaches which I learned during the bachelor studies at FTVS UK. The general part informs about anatomy and physiology of the skin, about diagnostics of degree and extent of burns, methods of treatment, physical and psychological rehabilitation. Special part includes methodology of the work, anamnesis, opening kinesiologic analysis, individual therapies, outcoming kinesiologic analysis, the effect of the therapy and end. Results: I worked out a detailed case study of a patient with this diagnosis and a literature search has been worked out. The main contribution of the work was the possibility to gain new experiences and knowledge to this diagnosis.
Biofuel from aqueous weed

Presented bachelor thesis is focused on renewable energy sources - specifically biofuels made from algae. At the beginning there will be discussed the legislative measures related to biofuels within the European Union and Czech Republic. The other part will follow up algae and technologies of algae cultivation in raceway ponds and photobioreactors. The cultivation of algae is directly connected with production of biofuels such as bioethanol and biodiesel and other valuable products which are discussed in the last part of this thesis.

Tomáš Křížek: Applications of Capillary Electrophoresis in Life Sciences (Dissertation thesis) ABSTRACT This thesis is focused on the applications of capillary electrophoresis in two important areas of life sciences, proteomics and enzyme assays. In the first part, Pluronic F-127 copolymer was studied as a sieving matrix for proteomic applications of capillary gel electrophoresis. The effect of thermoassociation of Pluronic F-127 on the separation selectivity was investigated and no difference in selectivity of the separation below, inside and above the thermoassociation temperature region was observed. The performance of Pluronic F-127 in capillary gel electrophoresis was compared with dextran as a commonly used sieving matrix. The results showed, that Pluronic F-127 offers superior performance for low-molecular-mass proteins because it provides higher separation power than dextran with significantly lower viscosity of the background electrolyte. The lower viscosity makes the polymer easier to replace after each analysis, which leads to remarkably higher repeatability of the experiments. Separation and determination of possible products of enzymatic cleavage of 4-nitrophenyl-N,N'-diacetyl-β-D-chitobioside using capillary electrophoresis

This work deals with the development and optimization of conditions of a method that can be used to compare the activity of the enzyme β-N-acetylhexosaminidase in hydrolysis of a natural substrate and a chromogenic substrate, which is often used in the study of enzyme kinetics. As a substrate, 4-nitrophenyl-N,N'-diacetyl-β-D-chitobioside was selected for cleavage. This oligosaccharide contains bond, which the enzyme cleaves in the natural substrate, and the bond that occurs in the chromogenic substrate. To determine the products arising from enzymatic hydrolysis of 4-nitrophenyl-N,N'-diacetyl-β-D-chitobioside, capillary zone electrophoresis was used. First, it was necessary to find the optimal composition of the electrolyte, its pH and concentration. The optimal background electrolyte was a solution of sodium tetraborate at a concentration of 25 mmol/l and a pH of 10.25. Subsequently, repeatability, calibration curves and linearity, limit of detection and limit of quantification were investigated. Repeatability of migration times ranged up to 0.6%, the repeatability of peak areas between 2.5 and 6.3%. Limits of detection were ranging from 0.005 to 0.120 mmol/l. Finally, the optimized method was successfully used to monitor the actual enzyme cleavage.
