Laccase activity profiling in Trametes versicolor cultures degrading endocrine-disrupting compound Delor 103

In this work endocrine disrupting potential of Delor 103, a commercial mixture of PCB congeners, was studied along with its effect on production of laccase by the ligninolytic fungus Trametes versicolor. Using a gene-reporter yeast assay for evaluation of hormonal activity Delor 103 showed an androgenic activity with an EC50 value of 2.29. 10-2 mg/l. Chlorbenzoic acids, Delor 103 potential metabolites resulting from microbial degradation, displayed on the other hand an estrogenic activity, indicating possible changes in hormonal activity of Delor 103 during its microbial degradation. The addition of Delor 103 to mineral medium T. versicolor cultures resulted in an up to 257times higher laccase activities detected in fungal cultures. Delor 103 induced enzymes showed different pI values from those of control cultures. In a complex malt-extract glucose medium (MEG) the stimulation effect of Delor 103 was kept down. Further, the production of laccase and synthesis of different pI forms depended strongly on the growth phase of fungal cultures. Exponencially growing cultures of T. versicolor were able to produce up to 7 different pI forms of laccase in responce to Delor 103 whereas stationary cultures produced only 4 enzyme forms with higher pI values. Existují na poli historického zkoumání vždy určitá místa, která nebudou nikdy zcela zaplněna. K takovým místům patří bezpochyby ta týkající se náboženského cítění daného národa. Tu nastupuje archeologie, aby se za pomoci hmotných nálezů a literárních pramenů pokusila oživit ducha dávno mrtvého lidu. Přitom vyplují na povrch překvapivé podobnosti v určitých rysech kultu mezi jednotlivými národy, a rozproudí debatu o vzájemném ovlivňování se či nezávislém vývoji těchto prvků. Mezi takové prvky se v oblasti východního Středomoří řadí i kult kamenů. S uctíváním kamenů se setkáváme jak v prostředí krétsko-mykénském, tak na území Blízkého východu, jakož i severní Afriky všude tam, kam zasáhly stopy foinické civilizace. Známe betyly z mnoha foinických měst, např. Byblu. Rovněž frýgická matka bohyně Kybélé byla uctívána jako kámen, snad meteorit. Taktéž Bible zmiňuje město zvané Beth-el, kde Jákob vztyčil kámen jako " Dům Boží". Tato víra byla známa i ve starověkém Řecku, ať už se jednalo o sloupky boha Apollóna či různé omphaly, z nichž nejslavnější byl umístěn v Delfách. Za Severovců se kult dostal i do Říma, jistě i zásluhou Julie Domny, ženy Septimia Severa a dcery vysokého kněze z Emesy. Media representation of Czech nobility in the context of restitution claims

The question of integration of nobility into the larger society has come to light over the last 15 years when representatives of Czech nobility addressed public attention concerning restitution claims. As a research shows, the attitude of Czech society towards nobility carries xenophobic features. [CYYM]. In today's society, for many participants media is often the only source of information about certain people or events. Representatives of noble class are among those "unknown quantities" because the majority of the Czech population usually has only superficial personal experience with their lifestyles, standards and values of nobility. These are the exact cases where the media may acquire the potential to infl uence the attitude of the public. The aim of the thesis was to identify how Czech media covers the noble class in the context of their restitution claims and whetherthey confirm or deny existing stereotypesand prejudice. For historic reasons opinion of nobility within Czech society is rather negative. Media has the power to change this by representing nobility as people who are responsible for their property, carry certain moral values, and respect their family and traditions. ANTIBACTERIAL EFFECT OF SELECTED NANOPARTICLES AS REVEALED BY DOUBLING TIME OF TREATED ANTHOMONAS CAMESTRIS PV. CAMPESTRIS CULTURES

Besides many possibilities of applications of nanoparticles in the field of medicine, diagnostics, molecular biology, bioorganic chemistry or remediation of environment, there is also a potential of employment of nanoparticles as a tool for elimination and control of bacteria invading plant tissue. In this experiment an antibacterial activity of selected nanoparticles based on silver, gold and bimetallic silver/copper was tested on bacteria Xanthomonas campestris pv. campestris (Xcc) (strain 1279a). The strongest inhibitory effect represented by doubling time of treated cultures was measured in the presence of the smallest silver nanoparticles (9 nm) at the highest concentration (5 ppm).
Effect of a cultivar and thermal treatment of seeds in selected vegetables at early growth phase on the sensitivity to infection by important fungal pathogens

The methodology is focused on the effect of selected factors on the development of important vegetable illnesses: Alternaria brassicicola dark leaf spot of cabbage, A. dauci carrot leaf blight, and Botrytis aclada onion neck rot and Peronospora destructor onion downy mildew. In the case of A. brassicicola, A. dauci and B. aclada, the plants in a defined stage of development were infected by the strains of fungal phytopathogens originating from acknowledged collections. In the case of P. destructor the plants were infected by a fungal strain obtained from a field plant culture massively affected by this phytopathogen. The effect of the cultivar used and of the seed thermal treatment on the development of the above given phytopathogens was investigated. It was found that, in the case of the cultivars used, the interpretation of the results had to include the factor of the cultivar´s earliness that was important with respect to the resistance to the fungal pathogens.
Occurrence of PBDE compounds and Bisphenol A in rivers with the occurrence of cryfish

The thesis deals with result evaluation of a project under the auspices of the EAA and Norway Grants, which ran from 2015 to 2016. The project's goal was to evaluate water quality at sites with native and invasive crayfish. The project also included monitoring organic substances such as Polybrominated diphenyl ethers (PBDEs). I mainly focused on a subset of PBDEs, the brominated flame retardants (BFRs). These substances have been monitored since the 1970s, when food was accidentally contaminated leading to the contamination of the nearby area in the US. What is alarming is their persistence in the environment, which leads to their constantly increasing levels. Information about the transformation and biodegradation of PBDEs in the environment is still limited and is continually analyzed in new studies. These substances are poorly volatile and insoluble, which makes dust and particles arising from the products containing BPAs the main sources of contact. Another agent, whose presence in the environment of the Czech Republic I analyzed, is Bisphenol A (BPA). BPA is used in the production of epoxy resins and polycarbonate plastics. BPA is a toxic substance with the ability to disrupt hormone function. Baby feeding bottles have been confirmed to contain these compounds. In April 2004, the Czech government approved a (program to reduce pollution of surface waters by dangerous harmful substances). In August 2004, the EU enacted a ban on the sale and use of penta-BDE and octa-BDE in all fields. Since 2011, Bisphenol A may not be used in the manufacture of baby feeding bottles. The goal of this thesis is to analyze and evaluate the amount and possible consequences of these substances in our environment, which are alarming even after full or partial bans on the use of these substances.
Cultural standards and their impacts on business negotiation - case study of China

This bachelor thesis is focused on the impact of cultural standards on business negotiation focusing on China. In this work were used descriptive and comparative methods of investigation. The first part deals with culture as it is, provides the demarcation of concepts as culture, cultural relativism or organizational culture. Further provides partitions of some specifics of culture and a brief description of history and its evolution. Consequently contains a detailed description of cultural dimensis theory from Geert Hofstede, which is consisted of five parts: power distance, collectivisme versus individualisme, masculinity versus femininity, uncertainty avoidance and long term orientation. In the second part is application of these dimensions on Chinese culture. Below are specific features of Chinese culture, like mianzi - face, guanxi - connections and also are described some aspects of business etiquette with particular focus on negotiations in China. Practical part demonstrates that the Chinese culture plays a significant role in trade negotiations with Chinese partners.
Phonological and morphological skills of preschool children

This bachelor's thesis is focusing on the development of preschool children, primarily on their phonological and morphological abilities. The main objective of this thesis is to examine further risks of reading abilities,indicating possibilities of reducing all these risks by the methodical work with children of preschool age. In the theoretical definition the physical, psychological and social development of children is defined, as well as their maturity and readiness for school in regards to speech as well as their phonological and morphological abilities. The practical part is focused on quantitative research of preschool children, with a focus specifically on the observation of variables, mostly in phonological and morphological abilities, techniques, practices. Based on obtained results are recommended exercises for the development of these abilities.
