Stabilometric platform Nintendo Wii and program HomeBalance were used during the therapy and enable the visual biofeedback to the pacient. Another specification of this therapy is that the whole process was situated to home environment. The therapy lasted one month and every patient should exercise every day. Time of one exercise depended on the patient's skills. The patient's task was to change position of the center of pressure to the specific place. Every day the patient exercised identical therapeutic scene consisting of a balanced position sequence in the space, that the patient should have achieved. The success of the therapy was evaluated objectively by intrumentation (FootScan) and objective tests, but also by patients' subjective feeling. Two patiens, after brain damage occurred as a result of the traumatic injury, were selected for this study. It objectively evaluates the effect of the chosen therapy, which is balancing Chung Shi shoes brand. Balance shoes are modern and revolutionary element that is used in physiotherapy. It is objectively evaluated on the footscan device, biomechanical noninvasive imaging equipment, measuring the disintegration of static and dynamic pressures on the soles of the feet. The theoretical part deals with the incidence, prevalence, epidemiology, consequences and rehabilitation of the traumatic brain injury and the possible effects on the postural stability. The practical part is compiled by a qualitative research, this is a pilot study. Two groups took part in the measurement: experimental groups of two women after traumatic brain injury at the age of 20 and 24 and a control group of two women showing no signs of postural instability, aged 21 and 23. Abstract: The bachelor thesis is focused on disorders of postural stability of patients after brain damage. The brain damage is classified as very serious health condition, which can be caused by traumatic brain injuries, post-stroke or as a consequences of cancer, neurodegenerative diseases, etc. The brain damage may develop disorder of postural stability, which is manifested by feelings of insecurity of standing, walking, increased frequency of falls and dizziness. This work is not based on classical physiotherapy techniques. Aplication of this therapy uses stabilometric platforms and 3D scenes in virtual reality. The therapy consists of testing the stability of standing and therapy in program StereoBalance. Therapies were absolved by two patiens post-stroke. To be able to compare the results of patients with postural stability disorder, healthy population was also examined (10 probands). The author chose Synapsys Posturography System device for evaluating the effect of therapy. The theoretical part of this thesis defines basic concepts, especially brain damage, balance disorders and posturography. Practical part describes metodology and results of the research. 11 people participated in this research and two of them were patients after brain damage. Values measured in a group of patients and in reference group were compared in practical part. In the end of this thesis, there are some recomendations and final summary of results of the research. It tries to sum up the fundamental information, propose an effective therapy and other possibilities of using 3D scenes. 3D scenes were used in this thesis which affects the postural stability of the patients and the Wii Balance board which was used to monitor the selected parameters of the patient's stability. All data were recorded by a PC. A group of 15 healthy individuals and 2 patients after the brain injury underwent the measurement. The results of the patients were compared to the results of the healthy individuals and the differences were pointed out. Afterwards, the two patients were participating in the training of stability using 3D scenes for 12 weeks, during that their stability parameters were recorded using a PC. Characterization of phosphatases in tobacco plants (Nicotiana tabacum L.)

Phosphateses (EC 3.1.3.x) are a group of enzymes that hydrolyze phosphoesters. That way they affect the energetic metabolism of a cell and its regulation. Phosphatases that dephosphorylate proteins are an integral part of signaling pathways, stress responses and they modulate enzymatic activity. This thesis deals with the study of phosphatases obtained from tobacco leaves (Nicotiana tabacum, L.). Solution of enzymes was prepared by extraction in both acidic and alkaline buffers. Through the use of the chromogenic substrate pNP-phosphate it was determined that there is a higher phosphatase activity in the glycosylated fraction than in the fraction that did not bind to Con A Sepharose. The research of the ions effect on the phosphatase activity has determined that Mg2+ and Ca2+ show positive effect on the phosphatase activity while the effect of Co2+ and Mn2+ is inhibitory. The Zn2+ ions have shown no effect whatsoever. The glycosylated phosphatases also dephosphorylated low-weight-molecular substrates phosphoserine, ATP and glucose-6-phosphate. The research of protein phosphatase activity discovered the affinity to the substrate phosvitin, although neither to casein nor its tryptic cleaves. 3 TITLE: Ways of creativity development in elementary mathematics education SUMMARY: This thesis deals with creativity - from a general concept to a more specific perspective - creativity in mathematics lessons and textbooks at elementary schools in relation to the curriculum method and transformation of a number of textbooks towards unique teaching methods. The aim of this thesis is to describe the current situation on a selected sample of experimental groups - elementary school classes (two of them second year and three of them fourth year) which specialize on integration of pupils with specific learning and behavioral disabilities as well as extraordinarily talented pupils and thus provide the broadest possible range of pupils, which enables to find possible links between the used methods and creative teaching approach including creating one's own teaching materials and consequently searching for possible ways to develop creativity of pupils in relation to used methods, textbooks, other materials and forms of work. Study of acid phosphatase in pea and tobacco

Phosphatases catalyze the hydrolytic fission of orthophosphoric acid ester. They are generally divided into acid and alkaline phosphatases according to their pH optimum. There was preparated the extract of leaves Nicotiana tabacum L. and Pissum sativum. Furthermore, there were determinated pea and tobacco phosphatases's rate constants. Reaction speed katalyzed by tobacco phosphatase is 10,0 mol/min.ml and by pea phosphatase 38,0 mol/min.ml using paraNP - phosphate as a substrate. Using a substrate phospho - L serine the reaction speed katalyzed by phosphatase of tobacco is 1,8 mol/min.ml and by phosphatase of pea is 0,4 mol/min.ml. Michaelis constant Km is 1,8 mM for tobacco phosphatase and 8,5 mM for pea phosphatase using paraNP - phosphate. Using a substrate phospho - L serine Michaelis constant Km of phosphatase of tobacco and pea is the same - 4,0 mM. The pH optimum of pea phosphatase is 5,0 using paraNP - phosphate as a substrate and 6,0 using phopho - L serine. pH optimum of tobacco phosphatase is 5,4 using paraNP - phosphate and 7,0 using phospho - L serine. Temperature optimum for pea phosphatase is 60 C and for tobacco phosphatase 55C.
The effect of biotic stress on the metabolism of saccharides in tobacco plants (Nicotiana tabacum L.)

Plants have developed a number of ways how to minimise negative influence of the environment. As a consequence of stress action, plants carbohydrate metabolism is quite often influenced, esp. on the level of expression and activities of different enzymes and also several metabolites concentration. One of key enzymes of carbohydrate metabolism is invertase. The aim of this work was to find out, whether the activity of its isoforms (cytoplasmic, vacuolar and extracellular) in tobacco plants is influenced by Potato virus Y (PVY). It was shown, that activity of cytoplasmic invertase was not affected, but the activity of vacuolar and extracellular isoform was enhanced during potyviral infection. Hence, it is likely, that vacuolar and extracellular invertases are related to plant antiviral defence. The effect of PVY on other enzymes of carbohydrate metabolism and several metabolites content was studied. Activity of α-amylase and phosphorylase, starch-degrading enzymes, was strongly enhanced during potyviral infection. That is probably how plant cells get glucose, which is a key source of energy and metabolites for biosynthesis of different compounds. It may also serve as a signal molecule.
