Development and optimalization of methodology for detection of GMO potatoes

Genetically modified (GM) or transgenic crops, now more often called ``Biotech crops{\crqq} they are commercially cultivated since 1996. And also since 1996, the first year of commercialization of biotech crops, GM potatoes were cultivated in USA, Mexico, Canada and later in South Africa, China and India. The global area of approved biotech crops in 2006 was 102 million hectares and 22 countries grew biotech crops, 11 developing countries and 11 industrial countries. The Czech Republic is on of the six EU countries where biotech crops are cultivated at present. The most compelling case for biotechnology, and more specifically biotech crops, is their capability to contribute to: increasing crop productivity and stability of productivity and production; conserving biodiversity, as a land-saving technology; the production of renewable resource based bio-fuels. This diploma paper was focused on developing of fast, precise and cheap method based on PCR to detect the presence of transgenes in potatoes {--} tubers and leaves, allows monitoring the presence of GM potatoes in market, environment, etc. and to quantify ``contamination{\crqq} of ware potatoes (tubers) with GM ones.

Natural Gas Consumption Modeling: Data as a Key Factor

Natural gas companies have a numerous group of small customers, whose consumption is known only cumulatively for a period of approximately one year. Thus, almost all activities of the companies require high-quality modeling of such customers’ consumptions in the period between two meter-readings. The paper discusses one of the most crucial key factors of the modeling: the used data, their accessibility and quality.

Understanding consumption behavior of customers is essential for natural gas distribution and trading companies. For large customers, consumption is measured daily or, at least, monthly. However, meter readings are taken and consequently billed in approximately 12-month intervals in small commercial and resident sectors. This period is too long with respect to the needs of gas companies. Furthermore, the companies have to estimate unbilled revenues at the end of an accounting period. Thus, there is a strong interest in mathematical modeling of the consumption patterns as a basis of such estimates. This paper discusses requirements for unbilled revenues estimation, and summarizes experience with mathematical model GAMMA used by West Bohemian Gas Distribution Company in Pilsen, Czech Republic. In addition, plans for further development of model GAMMA are presented in connection with a research grant awarded by the Grant Agency of the Academy of Science of the Czech Republic.

Efficiency of the gas separation with porous ceramic membranes might be expected higher for gas mixtures of adsorbable and nonadsorbable components in comparison with mixtures of only nonadsorbable gases. The reason is the additional specific contribution of surface diffusion to the gas transfer through a membrane in the case of the existing adsorption of permeating substances. The transport of pure nonadsorbable gases in porous material was already well described [1]. This work deals with the transport of binary adsorbable gases through porous Vycor glass. The transport of adsorbable gases in such pores is considered to be contributed by both gas phase and surface diffusion. The measurements were performed using a modified Wicke-Kallenbach cell possessing a closed and an open chambers [2, 3].

The aim of my thesis is to analyse selected enerprise, which is agricultural co-operative in Dolní Cerekev by using specific methods. Implement external analysis and analysis of internal sources, introduce the market in which the firm operates and analyse situation in this market. On account of gained information propose concrete suggestions to firm's management, which should lead to next development of this firm.
