Utilization of Diffusive Techniques in Study on Reactivity of Biocolloids

The main aim of this thesis is the utilization of simple diffusion techniques for the study on transport properties of copper ions in the systems containing humic acids with respect to the other parameters, which can affect the process (the structure of diffusion environment, the interactions between transported specie and diffusion matrices, selective blocking of binding sites of humic acids). The first part of experimental works was focused on characterization of studied materials (humic acids, humic sol and humic hydrogel). The main part of the thesis was dealing with the optimization of simple diffusion techniques, which were suitable for the study on transport of copper ions in matrices containing humic acids, taking into account the mutual interactions between studied components in the system. The obtained diffusion characteristics were compared to the data determined using sorption experiments. Consequently, the minor goal of the experimental works of this thesis was also the assessment of the influence of basic physico-chemical parameters of studied materials on transport phenomenon.
Utilization of flow-coulometry in study of transport metal ions in humic gels

This diploma thesis deals with the study of non-stationary diffusion of cupric ions in humic gels. The main objective of the work was the optimalization of conditions for using the electrochemical analyzer EcaFlow 150 GLP for the study of diffusion experiments. Next part was focused on reaching of constant concentration profiles of cupric ions in humic gel. Last aim of the study was the bond strength between cupric ions and humic acids. Diffusion was characterized by the values of the diffusion flux and cupric ions concentration profiles in humic gel. The data necessary for calculation were obtained by the electrochemical analyses of the extracts of each slice after diffusion on the EcaFlow 150 GLP. It has been proved that after application of several adjustment is this method suitable for the study of diffusion experiments. By using of different extraction agents was obtained the fractionalization of cupric ions according to the bond strength to humic acids.
SEC-MALLS study on hyaluronan-albumin interactions

This work is focused on interactions of hyaluronan (HA) and bovine serum albumin (BSA). These interactions were studied by SEC-MALLS method, where molar mass, root mean square (rms) radii, hydronamic radius and intrinsic viscosity of particles of the system were defined. Additionally, interactions were examined by measurement of DLS and zeta potential, where hydrodynamic radius and a value of the zeta potential were defined. The interactions were confirmed at any of the used media, but with different efficiency. Furthermore it was found out, that the high ionic strength minimizes the range of the interactions, magnifies the radii of the complexes, their intrinsic viscosity and zeta potential. It was proved that complexes become smaller in their radii and viscosity with growing BSA concentraton.
Dynamic light scattering and electroanalytical investigation of hyaluronan-amino acids systems

The bachelor thesis deals with interactions between low-molecular and high-molecular weight hyaluronic acid (HA) and protonized amphiphilic amino acids Lysine and Arginine. The interactions were observed in the area of low aminoacids concentrations with in the range 0–15 mmoldm-3. The interactions occur between the carboxyl groups of HA and the amino group of the protonated amino acids. Proving these interactions would allow us to physically modify HA and further more, use such a system as a carrier of pharmaceuticals. The resistence towards the ionic strength at the concentration of 0,015 and 0,15 moldm-3 NaCl was investigated. Previous results show that the system with unprotonated amino acids at low concetrations of electrolyte in the solution is rather disturbing. Therefore, the interactions were reinforced by the aminoacids protonation using HCl. The amino acids were protonated not only completely, but also partly because of negative influence of chloride anions. To study the interactions pH-metry, conductance and Dynamic Light Scattering were used.
New Methods of Cross-linking of Humic Substances

This bachelor thesis was focused on the study of crosslinking of humic substances. The main aim of this work was to find and optimize utilization of new crosslinking agents for humic acids. Commercial product – lignohumate – was also used in the work as comparative sample. Two groups of crosslinking agents were studied in this work – multivalent metals (CaCl2, MgCl2) and citric acid. In all cases the studied hydrogels were cross-linked by means of chemical valences. All prepared hydrogels were characterized using the determination of their inner pH, density and concent of dry matter. The process of aging of hydrogels – syneresis – was also studied. The main experimental method applied for hydrogels characterization was rheology. Prepared hydrogels represent interesting materials for following studies and also for possible future practical applications.
Influence of polyelectrolyte biopolymers on transport of ions

This bachelor thesis was focused on the study of interaction of cationic biopolymer with model diffusion probe (basic organic dye) in the gel media by diffusion processes. The main aim was optimization of simply laboratory diffusion technique (unsteady diffusion in a cuvette) because of study on interactions of chosen biopolymer with model compound depending on temperature. Cationic chitosan was chosen as model biopolymer. The model diffusion compound was chosen organic anionic dye Acidal Black 10B (C.I. 20470). Diffusion processes in plastic cuvettes were realized by transport of dye in a hydrogel media based on thermoreversible polysaccharide – agarose. The important content of this method is a monitoring of the time progression of diffusion profile by UV-VIS spectrophotometry. Important part of this work was characterization of hydrogel medium and biopolymers by basic physical-chemical methods. This bachelor thesis is focused on reactivity of chitosan and its behavior in different systems. Unsteady diffusion in cuvettes appears to be a universal method for the study on reactivity of biopolymers.
Stability of Soil Organic Matter

This bachelor thesis summarizes available information about current research of soil organic matter stability and it divides them into two logical units. The first part includes environmental, physical and chemical factors, including human activities, which affect the stability or decomposition of soil organic matter. The methods used for the study of organic matter in the soil are summarized in the second part of this thesis. There the attention is paid to the accumulation of organic matter in specific pools, the ways of extraction of organic compounds and the use of quantitative and qualitative analytical methods to describe various characteristic of organic matter.
The Study of Aggregation of Humic Acids in Different Xenobiotic Environments

Theme of the presented bachelor thesis is the study of the aggregation behavior of the humic acids (HA) in the environment of extraneous substances. As extraneous substances, three cationic surfactants were selected (CTAB, TTAB and Septonex). In the experimental part of the thesis, the humic acids were characterized in detail using different methods (elemental analysis, ash content determination, titration acidity determination). The experimental part of the thesis is dealing with the structure of created HA-surfactant systems. These systems were characterized using the results of the elemental analysis and FT-IR spectroscopy. The key part of the experimental part of the study is represented by the selection of proper methods and by determination of the critical aggregation concentration of the humic acids in the surfactant environment. For these purposes, two key methods were selected in the frame of the presented bachelor thesis: isothermal titration calorimetry and dynamic light scattering. The first method is sensitive owing to the energetic fluctuations, in the case of the dynamic light scattering we are able to detect well the changes in the particle size caused by mutual interactions of particular substances in the system.
Study on Reactivity of Cationic Biopolymer by Simple Diffusion Techniques

This bachelor thesis is focused on study of chitosan reactivity by simple diffusion processes realized in hydrogel media based on agarose. Mechanical properties studied by oscilatory measurements and the pore size were determined at agarose gels with the addition of chitosan as the main parameters influenced the transport properties. The main goal of this work is the investigation the reactivity of chitosan. Agarose is used only as a supporting matrix for the formation of a hydrogel. The state of the art is aimed primarily on the study of chitosan reactivity based on already published scientific papers.
The Study of Biopolymers Stability Using Light Scattering Techniques

The stability of chosen biopolymers (or simple carbohydrates and amino acids) was in-vestigated using light scattering methods. Samples were prepared by dissolving the sub-stance in deionised water or in diluted acetic acid. Firstly, the effect of increasing concen-tration on zeta potential was observed and the optimal concentration for the next measure-ments was determined. Afterwards, the stability and the molecular weight of used sub-stances were studied during long-time storage in different time intervals. It was found out that chitosan, CMC, hyaluronate and sodium alginate were subjected to degradation whereas increase of the molecular weight and the zeta potential was observed in case of BSA. In the end, the samples were exposed to several different external effects, namely the influence of different temperatures, UV radiation and increasing ionic strength. The results showed that rather higher temperature has a significant impact on biopolymers stability. However, used biopolymers were hardly affected by UV radiation except BSA whose molecular weight increased considerably. Decrease of the zeta potential absolute values and the molecular size usually occurred with increasing ionic strength. The change of molecular weight was not detected.
