Microbiology of wine

The aim of this work was the investigation of the amount and the composition of microflora during the winemaking of red wine. The study was performed in 3 factories in Velké Pavlovice region, where each of them utilized its own technological approach. According to this, during the period of preliminary fermentation, 3 different technological procedures were used, which differed in application of commercial yeasts and enzymes, and sulphur dioxide. They differed also in the temperature during the period of alcoholic fermentation and the application of commercial bacteria during the period of malolactic fermentation. From the experimental results follows that the application of different technological approaches leads to the differences in the number of both yeast and bacteria viable cells and the total number of microorganism cells in the period of preliminary, alcoholic and malolactic fermentation.
Principles and utilization of cosmetotextiles

The aim of this thesis is to create a literature review about cosmetotextile. Cosmetotextile is the combination of the cosmetic industry and the textile industry. The future of the textile industry is represented by the progress of new kinds of technical textiles, that are based on revolutionary technologies, innovative materials and utilization. This means developement of new products with high added-value properties, for example application of antimicrobial and UV-protective agents, fragrances, anti-cellulite treatments or skin softeners. Such textile finishes are both important commercial target and challenge for technical developement. This thesis focuses on the field of intelligent textiles and combination of textile and cosmetics. It digests agents, that are used and their effects and describes the principle of microencapsulation, which is a very important principle applied here.
Microorganisms with lipoplytic activity and their applications

Lipases are hydrolytic enzymes that are produced by many types of microorganisms. This thesis describes not only the microorganisms which produce lipases but also different possibilities for the industrial utilization of lipases. Lipases are widely used in reactions where the combination of a lipophilic substrate with a hydrophilic one is necessary – in the synthesis of ascorbic acid fatty esters, sugar esters, lipoaminoacids and in the lipophilization of phenolic derivatives. Lipases are also important in relation to environmental protection. For example, they are used for the purification of waste water. In this thesis, five different preparations containing microorganisms with lipolytic activity were tested for lipolytic activity. There are many ways of determining lipolytic activity. The spectrophotometric determination of lipolytic activity that uses the ability of lipases to divide p-nitrophenyllaurate to p-nitrophenol may serve as an example. After this p-nitophenol is detected spectrophotometricaly.
Plants polysaccharides and their importance in food production

Some findings about plant polysaccharides, their characteristics and structures, modifications, changes, their biosynthases, isolationes and primarily their importance in food-stuff industry are the subject matter of this review. Because of their modifications and because of the significant role they play in food-processing industry it is focused on starch and cellulose. For better understanding of whole meaning it was necessary to attend to the theme from the initiation – mainly the cell structure and then syntheses, especially photosynthesis, ways that are monosaccharides and polysaccharides generated.
Utilisation of plant origin waste

This thesis describes properties and utilization and specifies the composition of organic plant waste produced during industrial processing. By cultivation of specific micro-organisms from this waste material it is possible to produce enzymes that have further beneficial industrial utilization. The experimental part of this thesis deals with the production of enzymes from the applied waste material grape pomace with the help of chosen micro-organisms. Waste, its processing and utilization is and always will be a key issue in our society.
Comparison of sensitivity of Bacillus subtilis and Clostridium difficile against selected chemical ingredients.

Sporicidal activity of disinfectants is tested on Bacillus subtilis spores according to ČSN EN 13704 standard. Until December 2007, 10 countries encountered outbreaks and 7 additional countries had endemic cases of Clostridium difficile-associated disease due to hypervirulent strain C. difficile PCR ribotype 027. This outbreak of CDAD with increased severity and mortality is spread as typical nosocomial infection. Clostridium difficile spores can persist on dry surfaces for 5 months. Therefore it is important to compare the effectiveness of disinfectants on both strains and find out whether Bacillus subtilis is the only proper microorganism for testing the sporicidal activity.
Determination of chlorides in pharmaceutical substances

The topic of this bachelor work is a validation of the method for „Determination of chlorides in pharmaceutical substance“ is made in a form of validation of an analythical method. This titration method with potentiometric indication was developed in a company Sicor and it is used for determination of the residual chlorides volume in the substance called Pemetrexed. Validation is a process which assess suitability of a given analythic system for gaining relevant data (results). Validation constists of four tests: precision, reproducibility, lineraity and test of solution stability, although the test of reproducibility was realized in other laboratory (QC). For determnination of the chlorides was applyed titrator 716 DMS Titrino with argent combined electrode. For a data evaluation were used software tiamo 1.1 and MS Excel.
Hyaluronic acid and possibilities of its utilization

This bachelor thesis presents contemporary knowledge gained in the research of hyaluronic acid, describing its physical-chemical and biological properties as well as principal possibilities of its utilization both in medicine and cosmetics. Hyaluronic acid is a linear polysaccharide formed from disaccharide units containing N-acetyl-D-glucosamine and glucuronic acid. Hyaluronic acid represents one of the main components of the extracellular matrix and is present in almost all biological fluids and tissues (mainly in connective tissue). Its high molecular mass and unique viscoelastic properties offer a wide range of use of this molecule in cosmetology, ophthalmology, rheumatology, otolaryngology, dermatology, plastic surgery, wound healing and drug delivery.
