Směrovaná polymerní léčiva pro terapii nádorových onemocnění
Kovář, Marek ; Mrkvan, Tomáš ; Šírová, Milada ; Strohalm, Jiří ; Šubr, Vladimír ; Etrych, Tomáš ; Ulbrich, Karel ; Říhová, Blanka
We have presented data on development of polymer-bound cytostatic drugs and their therapeutic efficacy in selected tumor models

Cytotoxic drugs based on HPMA polymer show direct cytotoxic and cytostatic effect together with an ability to mobilize the immune system. Two types of linkage of the drug to the polymer are discussed, as well as linkage-related differences in tumour cell accumulation and fate of the cells
Protinádorová účinnost konjugátu na bázi N-(2-hydroxypropyl)metakrylamidu, obsahujícího doxorubicin a normální lidský imunoglobulin

Polymeric conjugate based on HPMA containing cytotoxic drug (doxorubicin) and human “non-specific” immunoglobulin has a significant anti-tumour effect and does not impair immune functions of the host. C57BL/6 mice with EL-4 lymphoma, when completely cured with the conjugate, were protected against the same tumour due to a specific anti-tumour immune reaction
Diffusion of PHPMA polymers in brain extracellular space

The movement of bulky PHPMA polymers through ECS is limited in contrast to linear PHPMA macromolecules which have the same ability to explore ECS as small molecules.
Antracyklinová antibiotika na bázi protilátkou směrovaného HPMA

Conjugates based on N-(2-hydroxypropyl)methacrylamide represent a new generation of polymeric anticancer drugs with improved therapeutic potential and considerably decreased nonspecific side effects. Antibody -targeted derivatives show specific active targeting and nonspecific passive accumulation in solid tumors.
