Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Botany and Ecology Candidate: Novotná Magdalena, Mgr. Consultant: Cahlíková Lucie, Ing., Ph.D. Thesis title: Biologically Active Metabolites of Plants. IV. Alkaloids from Chelidonium majus L. and Their Neurotropic and Antioxidant Activity The aim of this thesis, which follows the diploma thesis, was to process the second half pool of alkaloids of Chelidonium majus, chlorides insoluble in chloroform, to isolate the remaining alkaloids in a pure form, to test their biological activity against two human cholinesterases (HuAChE, HuBuChE) and to determine their antioxidant activity. After division by column chromatography, the two alkaloids were purified using preparative chromatography. Based on structural analysis (NMR, MS) and comparison of data from the literature, the substances have been identified being chelidonine and homochelidonine. The activity of both alkaloids against two human cholinesterase (HuAChE, HuBuChE) was determined. Experimentally obtained IC50 values were compared with the standard drugs, that are used in the treatment of the Alzheimer's disease (galantamine and huperzin A). Chelidonine exhibited promising IC50 values towards HuAChE and HuBuChE (IC50 26.8±1.2 µM HuAChE and 31.9±1.4...
