Effect of year on field germinability of opium poppy seeds

The aim of this thesis was to perform an experiment with opium poppy control and drought stressed plants. Gained seeds were tested for germinability in normal conditions and during simulated drought stress induced with PEG 6000. It was found plants for seed production provides less seeds with lower quality, but the vigour and germinability of these seeds are better. In this thesis I also tested the method of surface plasmon resonance as potentialy suitable method for dehydrin accumulation analysis. I managed to verify method for termostable proteins isolation and at the same time I confirmed, the SPR method is potentially usefull method for dehydrin accumulation analysis in opium poppy seedling plants.
Molecular detection of genes involved in drought stress reaction in opium poppy

Drought belongs among one of the main abiotic factors that influence the yield and quality of agricultural crops. However, plants fight these negative environmental impacts by mechanisms developed during evolution. One of these mechanisms is the plants´ response on molecular level, which is gene expression. These genes potentially reacting to drought include the following genes: MLP43 and SDD1. For the purposes of molecular detection of these genes, the DNA from opium poppy plants (Papaver somniferum L.) Orel and Onyx type, was necessary to be isolated. Further, primer sets for the genes of MLP43 and SDD1 were designed. The chosen primer sets were used for analysis purposes using the real-time PCR method. The PCR method was also optimized.
Optimization of PCR detection of biosynthetic alkaloid pathway genes in opium poppy

The thesis deals with the creation of the biosynthetic pathways of selected alkaloids of opium poppy. Then is following the design primers for selected genes and optimization of PCR reaction of these genes for amplification and sequencing of the longest fragments. The PCR reaction was optimized for the 7OMT, TNMT and CODM genes. These genes are involved in biosynthetic pathways of morphine, codeine, papaverine, noscapine and sanguinarine. Each gene was split in half. The primers were designed separately for each half of the gene. Altogether were designed 13 pairs primers. 5 pairs primers were optimized by gradient PCR and gel electrophoresis. These primers were used for sequencing analysis.
Poppy peeds and Cannabis in the international Trade

The bachelor thesis focuses on the international trade with opium poppy and cannabis. At the beginning of the thesis the term of the international trade is explained and the instruments for regulation of the international trade are described here. Also the information about the legislation that regulates international trade with opium poppy and cannabis are provided here. This information is gathered from the books and internet sources. The primary aim of the thesis is to analyze the international trade with opium poppy and cannabis and discover the competencies of the customs administration. The secondary aim is to find out the knowledges and opinions of farmers in the Czech Republic about the legislation that regulates the cultivation and international trade with opium poppy and cannabis by using the form of questionnaire. The data about the export and import of opium poppy and cannabis in the Czech Republic and European Union are gathered from the official site of the Czech Statistical Office and Market Access Database. The collected data are analysed and processed into the graphs in Microsoft Excel. The results from the graphs are evaluated on the basis of the stated goals.
The methodology of in vitro cultuvation of opium poppy and production of homozygous materials for breeding

The methodology includes several options for preparing and maintaining initial donor F1 plants with respect to breeder’s possibilities. The next part presents the basic techniques for explant preparation, establishing of in vitro culture, regeneration of shoots and whole plants, transferring to non-sterile conditions, multiplying, identification and selection of diploid materials for further breeding. Attached there are specifications of cultivation media used. Important stages are documented with the help of figures.
