The effect of ionizing radiation on the complex of estrogen receptor with DNA

Estrogen receptor protein forms a specific complex with DNA bearing estrogen response element. The study of the effect of ionizing radiation on the stability of the complex is presented. Method of agarose gel electrophoresis was used to follow radiomodifying effects of estrogen, which is in conditions in vivo necessary for complex formation. Method of retardation polyacrylamide electrophoresis was used to determine overall stability of the complex as a function of absorbed dose.

Monte Carlo modelování radiolýzy bází DNA

An attempt to explain differences in measured yields of modified bases determined in free bases and in the double stranded DNA is made with help of theoretical calculations. A theoretical model is applied to predict yields of reactions of the most important radiolytic product, OH• radical, with individual nucleobases, nucleosides and short DNA oligomers. The relation between spatial limitations for radical approach to particular reaction sites within DNA and the yields of damaged bases is discussed.

Radiolysis of DNA-protein complexes

The summary of experimental measurements and theoretical predictions of radiolysis has been presented for two prokaryotic DNA- protein complexes: i)lac operator - lac repressor complex, ii) DNA bearing abasic site analog - Fpg protein.
