Optimization of isolation and PCR methods for the identification of CMS in onion
PETERKA, Václav
This bachelor thesis deals with the optimization of different methods of DNA isolation from different types of onion (Allium cepa L.) samples and the subsequent detection of the Ms locus and the detection of cytoplasmic male sterility (CMS). The different types of DNA extractions performed and their results were compared and evaluated. The best quality DNA was isolated from leaves using the CTAB-PVP. These test samples were also used for PCR for the detection of CMS and Ms locus using several different markers, which were compared with each other. The most suitable markers were then used in the PCR for the main samples. DNA isolation by CTAB-PVP was also used for further DNA isolation from the main samples of onion, which were two sterile lines and four fertile lines. The extracted DNA from these samples was first used to detect the Ms locus. Detection was performed using the marker AcSKP1. The samples were collected over several weeks. The Ms locus was detected in all samples (C1C6), regardless of the age of the plant. However, the Ms locus was detected in various forms of zygosity in samples from lines designated C2, C4 and C5. Furthermore, the DNA samples were used to detect CMS in plants and the type of CMS was determined. The markers cob and orfA501were used for this detection. Using these markers, it was found that the lines that were declared sterile (lines designated C1 and C4) were indeed sterile and had the CMS-T sterility type. The lines C2, C3 and C6 that were labelled as fertile were also detected as sterile samples. This means that these lines are not 100% fertile. In the line designated as C5, no sterility was detected in any of the samples.
Technological, economical and ecological aspects of high efficient cooling
Hort, Martin ; Peterka, Václav (referee) ; Píška, Miroslav (advisor)
The diploma thesis is aimed to the influence of the high-pressure and low-pressure coolant system according to the machining operation. The comparison was carried out with different types of materials and cutting tools using the main machining operations – turning, milling, and drilling. The study is focused on the chip and heat removal from the cutting zone, tools life and wear of the tools. A description of the basic principle of the high-pressure coolant system meant is described in this thesis. The economical analysis of the high-pressure compared to low-pressure system was determined in the last part of this work.

The comparison of websites of insurance companies on czech market
Slunka, Václav ; Sigmund, Tomáš (advisor) ; Peterka, Václav (referee)
The aim of this work is creating a methodology for comparing web presentations of insurance companies. The methodology is completely subjective and evaluates website of the Czech insurance company, insurance company Allianz and Generali. Site accessibility has the greatest weight. In the theoretical section there are three main areas. These areas include the history of language for web, security at sites and Web payments. In conclusion of this work is evaluated user questionnaire, corresponding to the criteria in the methodology. Thanks to its results it should be clear whether some results deviate significantly from the methodology.
Comparison of selected websites in the sphere of Europe tourism
Pálek, David ; Toman, Prokop (advisor) ; Peterka, Václav (referee)
The main goal of this work is to create metodics which will compare choosen tourist websites. Firstly, I will choose 3 big tourist cities of Europe. I will make a lot of views on websites and each websites will recieves points depends on quality. In the end of metodics, the one websites will be the winner. The second part of this work is user ability test which will give participants qestions to find something on choosen website. I will compare the results of this test and make a conclusion which websites are best. After that I will compare results from metodics and the results from user ability test. In the end of this work I will highlight the main things which every tourist websites of big Europe city should have.
