The importance of determining the prognostic COVID score for the stratification of patients with COVID-19.

COVID-19 remains a life-threating disease with a very heterogenous clinical presentation for risk patients. Recently, there has been an effort to find a suitable laboratory marker for early and reliable assessment of prognosis of this disease. The scoring systems seem to be suitable markers with more complex evidential value. This diploma thesis is engaged in the importance of COVID-19 prognostic score then, which was developed and validated by the Sysmex Europe GmgH corporation in Hamburg (a subsidiary of Sysmex Corporation in Kobe) in the collaboration with European university hospitals. It is a score calculated from ten hematological parameters, which were proven to change dynamically at the onset of COVID-19. This score and the changes in selected hematological and biochemical parameters were observed depending on the course of COVID-19. The observed laboratory markers included WBC, Lymf, Neu, Eo, IG, Delta-He, DDI, FBG, NLR, CRP, PCT, IL-6, ferritin and troponin T. Ilona Fátorová Title of diploma thesis: Changes in selected laboratory parameters in patients with complicated COVID-19 The aim of the diploma thesis is to write a research on the topic using available literary publications. Comparison of changes in selected laboratory markers in COVID-19 disease in hospitalized patients and patients with a mild course of the disease and then statistical evaluation of the obtained data. The theoretical part contains a briefly description of the COVID-19 disease and its originator - SARS-CoV-2 virus. It deals with the description of the disease, transmission, pathogenesis and the course of the disease. Furthermore, risk factors for the serious course of the disease, its complications, clinical and laboratory diagnostics and finally treatment. In the experimental part of the work, a statistical analysis of the measured data is performed. Possibilities of hematopoietic stem cells determination in the peripheral bood.

In this study the hematopoietic stem cell, its properties, sources, utilization and possibilities of its determination in the peripheral blood are described. The detection of CD34+ cells by flow cytometry is the reference method for determining the number of hematopoietic stem cells. The aim of my work was to verify whether it is possible to identify such cells can also use another method. This method is the determination of hematopoietic progenitor cells by a haematology analyser Sysmex XE 2100. In the practical part of the study, data of patients obtained from the analyzer Sysmex XE 2100 and flow cytometer are processed. This is the number of hematopoietic progenitor cells, CD34+ cells and total number of leukocytes. The data are compared by using the Spearman correlation test. It has been shown that the numbers of hematopoietic progenitor cells and CD34+ cell counts, there is dependence. Correlation coefficients of 0.55 and 0.58 can be considered very well. The analyzer Sysmex XE 2100 can be used as an alternative method to determine the number of hematopoietic stem cells in peripheral blood and leukoconcentrate. Numbers of the CD56+ cells were gained by flow cytometry measuring the expression of antigen CD56 labeled with antibody against CD56. The correlation was reviewed with the numbers of LGL cells, which were found in the periferal blood of the patient by microscopic evaluation with common panoptical staining of blood coat. There were used the absolute counts of detected parameters for this comparison. The leukocytes from the measured blood count of the patients on the analyzer Advia 2120i were used for calculation the absolute counts of LGL cells. In the group of patients with immunodeficiencies and various forms of allergy where established an increased number of CD56+/CD3- cells (NK cells), we performed a correlation with the number of LGL cells defined microscopically. In the peripheral blood smear and in the bone marrow smear, we find the typical leukemic cells which are characteristic by their size, morphology of cell nucleus and cytoplasm. They are bigger than healthy lymphocytes, and they have most frequently the kidney shaped nucleus with fine net-like chromatin and the cytoplasm projecting into fine salients all over the cell perimeter. In the blood count we usually find a mild pancytopenia. To demonstrate the presence of the pathological cells in the peripheral blood smear and the bone marrow smear, we use the panoptic staining method according to Pappenheim. Jana Šklíbová: The Occurrence of Prothrombotic States in the Ústí n. L. Region and Their Treatment Monitoring This thesis deals with thrombophilia - either an inherent or acquired defect in haemostasis mechanisms which causes heightened inclination to thromboembolism. Early recognition of prothrombotic states as well as general risk markers in patients and an early treatment can reduce the occurrence of thrombosis and thromboembolism, thus saving many a life. The focus of this Bachelor Thesis has been on performing selected thrombophilic marker tests - a blood-platelet count, prothrombin test, activated partial thromboplastin time test [aPTT], antithrombin test, protein C test, protein S test and activated protein C test) on 2 groups of patients and a control group. The first group consisted of people who had not yet suffered from thrombosis, but demonstrated symptoms of risk factor presence (e.g. a family history, repeated miscarriages, pregnancy complications, etc.). The second group included patients who had recently had deep vein thrombosis. The data obtained from the tests have been evaluated statistically in order to find out whether the two sets of subjects are significantly different from each other. Comparison of Results of Coagulation Parameters Measured by Different Methods

The present thesis deals with the laboratory determination of the functional activity of coagulation factor VIII. It is one of the coagulation factors involved in hemostasis. Its deficit or functional deficiency leads to a severe bleeding disorder - haemophilia A. The aim of the theoretical part of this thesis is to create a brief overview of the coagulation factor VIII - heredity, structure, biosynthesis, secretion and function in the organism. It also describes haemophilia A, which is a disorder caused by a lack of synthesis, functional defects or the presence of inhibitors against the coagulation factor VIII. The history of the disease, clinical classification, etiopathogenesis, diagnosis and treatment are described in this part. Laboratory methods for determination of coagulation factor VIII and its specific inhibitor are described in the last part. The practical part deals with the determination of the functional activity of the coagulation factor VIII by the commonly used coagulant method and also by the chromogenic method.
