Gene expression in chicken embryo: micromanipulation and visualization methods

The aim of this work was to obtain better insight into the principles of cell structures and organs in the chicken embryo development. To reach this goal special methods of micromanipulations and visualization in vitro, ex vivo, ex ovo and in ovo were implemented and adjusted. These methods were used to study gene expression in neural crest development and eye development. In the course of long term research in our laboratory we observed that oncoprotein v-Myb influences the development of the neural crest and has the capacity to change natural cell fate. We performed a series of experiments to investigate v-Myb protein influence on neural crest cells differentiation, especially melanocyte lineage development, and its influence on gene expression in the neural crest. Therefore we focused on Gremlin 2 (PRDC), the gene upregulated by v-Myb in the neural crest. The established procedure of electroporation in ovo was adjusted to transfect cells of the developing eye and used to study gene expression during lens induction. The results obtained from chicken embryo experiments endorsed the study performed on mouse embryos. Authoritarianism of social workers

The aim of the Master Thesis is to identify the proportion of authoritarian personality of future social workers. In line with the objective of the work, the research question has been formulated in the following way: On what factors does the proportion of the authoritarian personalities of future social workers depend? Based on the research question the following hypothesis has been determined: Students of the programme Social Work in Public Administration are more authoritarian than the students of the programme Rehabilitation Psychosocial Care of Handicapped Children, Adults and the Seniors. Students of part-time studies are more authoritarian than students of programme Rehabilitation Psychosocial Care of Handicapped Children, Adults and the Seniors. With the increasing age, the proportion of authoritarianism grows. The quantitative research strategy, which was implemented using the questionnaire method through a standardized questionnaire, was selected to achieve the goal. The questioned team consisted of future social workers, namely students of Faculty of Health and Social Sciences in České Budějovice programme Social Work in Public Administration and Rehabilitation Psychosocial Care for Children, Adults and Seniors. Translation of F-scale was taken from the publication of T. W. Adorno. Data analysis were evaluated using descriptive statistics in MS Excel and hypothesis testing was performed in the SPSS statistical program. The research shows that in the first hypothesis statistically significant relationship has been captured in the properties of conventionalisms, authoritarian submission, authoritarian aggression, anti-intraception, power and toughness, destructiveness, projectivity and sexuality. In the second hypothesis, it was tested whether the students studying part-time are more authoritarian than students of full-time studies. A significant relationship was noted in attributes of subordination and authoritarian aggression. In the third hypothesis, it was demonstrated that a statistically significant relationship was detected in the attributes of subordination, authoritarian aggression, and superstition. The outputs of the Master Thesis research can be provided as a feedback for students or social workers. The results could be of interest to the students of the Faculty of Health and Social Sciences at the University of South Bohemia in České Budějovice. Authoritarian personality has not yet been processed for social workers.
Oncoretroviruses: tumor transformation mechanisms of hematopoietic cells resulting in leukaemia

HTLV-1 and FeLV are retroviruses, which are able to transform host cells and cause cancer, mostly leukemia, in infected organism. Belonging to Retroviridae family and both using very similar genome, these viruses developed different ways to reach transformation of infected cells. While FeLV uses insertional activation close to cellular proto-oncogenes in order to regulate transcription of these genes or carries cellular oncogene in its genome, HTLV-1 codes viral proteins which are able to regulate many processes of the cell. One of these proteins is Tax, which regulates many events in the cell, such as signalization, cell cycle, apoptosis and others. Another protein responsible for oncogenesis is HBZ, which is transcribed from antisense strand of proviral DNA. In the end HTLV-1 and FeLV strategies causing cancer are compared with some other leukemic retroviruses in order to show, that molecular strategies described on examples of HTLV-1 and FeLV are more or less common also for other oncogenic retroviruses. Influence of concentrated oxygen inhalation on pulse rate and saturation at short - term anaerobic load

Název: Vliv inhalace koncentrovaného kyslíku na tepovou frekvenci a saturaci při krátkém anaerobním zatížení. Cíle práce: Zjistit, zda hyperoxie ovlivní hodnotu měřených parametrů oproti normoxii při stejné zátěži. Nastínit problematiku inhalace kyslíku a souvislost hyperoxie se sportem. Metodika: Probandi (n = 10) absolvovali dvě 30 s jízdy maximálním úsilím na bicyklovém ergometru. Před druhou jízdou inhalovali po dobu 15 minut 100 % kyslík. U každé jízdy jsme měřili počáteční a konečnou tepovou frekvenci a saturaci hemoglobinu kyslíkem. Výsledky: Studie ukázala signifikantní rozdíl (p  0,05) hodnot počáteční i konečné tepové frekvence a počáteční saturace za podmínek hyperoxie oproti normoxii. Pokles saturace v průběhu zátěže byl vyšší u podmínek hyperoxie. Nárust tepové frekvence v průběhu zátěže nebyl věcně významný. Závěry: Inhalace koncentrovaného kyslíku pozitivně ovlivňuje aktuální saturaci, čímž vytváří v organismu vyšší zásoby kyslíku. Stereotypes about social worker on the basis of inhabitans of Tábor

This bachelor thesis deals with stereotypes of social worker at the residents of region Táborsko. In theoretical part, factors which influence public perception, especially their attitudes, stereotypes and prejudices are defined. Thesis further describes conception social work as such, its professionality, values, help and control. It focuses on the relationship between social work and mass media. Media have the power to influence its audience to different behavior and thus have impact on reorganization of society. In one of the chapters the social worker is closely introduced. Especially his competences, roles and authority. Social worker helps to represent social work and takes different roles in it. With social work stereotypes are deeply wedded which occur everywhere around us and are ordinary part of our lives. However stereotypes are sources of creating prejudices of which is in our society countless. Goal of this work is to find out how is the social worker stereotyped at the residents of region Táborsko. In other words goal is to explore opinion of inhabitants about social worker and analyze which stereotypes are represented the most. With quantitative research the research survey was implemented. Within this research the questioning method was used with semantic differential. For fulfillment of the work, two research questions were set: VO1: On which demographical factors depend the public perception of social workers? VO2: What are the sources of negative public perception of social worker? Following hypotheses were set toward above- mentioned questions: H1: Women perceive social worker more positive than men. H2: The higher education person has the more positive is stereotype perception of social worker. H3: People who experienced direct contact with social worker have more positive opinion of him. The research sample represents overall 309 inhabitants of region Táborsko. These representatives belongs to age group of 15 35 years. Concretely, it comprises of 309 respondents who live in region Táborsko. Collected data were analyzed by descriptive statistics in MS Excel programme. Testing of hypothesis was realized in the programme SPSS with method t-test and correlation. Characteristics, given to social worker by respondent, were defined by semantic differential. Respondents noted their opinion about particular object by choosing the mark on scale from one to seven. Endpoints were defined by pair of contradictory adjectives. From results of research survey the findings are following. For the first hypothesis significant statistic relation were found at human characteristics: careful careless, kind evil, responsible irresponsible, friendly unfriendly, strong personality weak personality, willing selfish, even tempered unsettled. Research shows, concretely average from the t-test, that at these characteristics women perceive social worker more positive than men. For the second hypothesis was tested whether with growing education the positive perception of social worker grows as well. Test demonstrates that the more people are educated the more they think that social workers are priceless, important and they have strong personality. Last but not least, for the third hypothesis significant relation was found at characteristics responsible irresponsible, friendly unfriendly, willing selfish, even tempered unsettled and unrecognized appreciated. From outcome of t-test it can be seen that people with own experience with social worker have more positive opinion than people whose experience is derived from media or they have none. Results from research of this bachelor thesis may be used as a source of information for public and also may interest social workers from Tábor region. To limit the occurrence of stereotypes I would recommend to create campaign where the public will be familiarized with profession of social worker.
