Meristem cultures in garlic (Allium sativum) breeding

The diploma thesis deals with the use of meristem cultures in garlic (Allium sativum) breeding. The source material was used variety Tantalum of garlic. The using material, before the isolation of meristem, was tested for the occurrence of viral diseases by immunological tests ELISA (Enzyme-Linked Immuno-Sorbent Assay), also known as EIA (Enzyme Immunoassay). The method used to detect antibodies and antigens. The material was tested for viruses onion yellow dwarf (OYDV - Onion Yellow Dwarf Virus) and virus genus Carlavirus (GCLV - Common Garlic Latent Virus). Then the sound material was sterilized, cultured and then it was obtained meristem. Cultivation of preparation meristem was realised according available methodologies. After preparation meristem put on MS medium with vitamins and growth regulator IAA, NAA and BAP.
Purification of patatin proteins from tubers of selected potato (\kur{Solanum tuberosum} L.) cultivars with subsequent characterization of isolated patatin proteins

The complex of patatin proteins were extracted from potato tubers (Solanum tuberosum L.) and its purification process is described in this work. The initial material for patatin protein isolation were potato fruit juices extracted from cultivars Laura, Sibu and Westamyl. Based on the known patitin´s properties, the ion-exchange chromatography (DEAE medium) coupled with affinity chromatography was applied. We used the column containing biological ligant of Concanavalin A, which was able to bind the carbohydrates in the molecule of patatin proteins. The used purification process were successful and according to the SDS-PAGE analysis we obtained patatin proteins of a high purity from each the potato cultivars. The antioxidant activity was observed for all samples and range from 0,007 to 0,086 mg ascorbic acid concentration / g protein.
The development in Rwanda after genocide in 1994

This diploma thesis deals with the post-conflict development in Rwanda and its main aim is to evaluate the tools adopted after the genocide in 1994 for the purpose of preventing recurrence of any similar conflict in the future. The thesis mostly concentrates on the process of justice (on the international, national and local level) and on the process of reconciliation of the polarized society in Rwanda as two key components of the post-conflict reconstruction. On the basis of the results derived from the detailed analysis of the tools from security, social, economic and political area, the thesis answers the asked question if the future conflict in Rwanda is nowadays hardly probable (thanks to effective and successful activity of the adopted measures) or if there is, on the contrary, some ethnic tension in Rwanda which could become the reason for repeating of any wave of violence in short- or medium-term.
The Zapatista Rebellion in Mexican Chiapas: political - historical context

This bachelor thesis deals with the Zapatista rebellion in Mexican Chiapas from the year 1994 and its main aim is to evaluate the achievements of the whole rebellion. Firstly, the thesis concentrates on the bases for the formation of the rebellion by studying crucial events from Mexican history. Furthermore, it describes the course of the rebellion in chronological sequence and it lays stress on the armed actions of the Zapatista, their non-military activities and peace negotiations with the government. In the last chapter, the thesis presents a deeper analysis of different aspects of the rebellion by the means of the analysis of structure, origin and members; the analysis of ideology, demands and aims and finally the analysis of means of struggle where the role of media is highlighted above all. The final evaluation of the achievements is devided into Mexican, global and theoretical level.
