Grapevine cryoconservation
Faltus, Miloš ; Kotková, Renata ; Bilavčík, Alois ; Zámečník, Jiří
Storage of grapevine germplasm is endangered by biotic and abiotic stressors in the field conditions, and it is connected with the risk of valuable accession loss. Cryoconservation is a method which eliminates the risk and makes a safe long-term storage of plant germplasm possible. Cryoconservation method is based on storage of plant material at a temperature of liquid nitrogen when all processes are practically stopped but the viability of plant material remains. This methodology describes preparation of plant explants before cryoconservation, apical shoot tip dehydration by cryoprotectants, procedure and principles of long-time plant storage and plant recovery for evaluation of explant survival and regeneration. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize it in the framework of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

Conservation of Allium germplasm collection by cryopreservation
Zámečník, Jiří ; Grospietsch, Martin ; Kotková, Renata ; Faltus, Miloš
Allium is important crop in the Czech Republic. Allium sativum L. germplasm is maintained in the field collection and this fact increases risk of accidental lost of genotype. Conservation of Allium sativum L. germplasm by means cryopreservation decreases risks of genotype lost. Allium sativum L. samples are stored at ultralow temperature that stopped all biochemical processes and the samples are stored without any changes for many years. This methodology describes procedure of material preparation, explant preculture and dehydration by mixture of cryoprotectants, shoot tips cryopreservation and their recovery. Principles of safe genotype storage and recovery are defined. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize the Allium sativum L. cryopreservation method in the frame of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

Character of transitions in frozen sucrose solutions
Sikora, Antonín ; Dupanov, V. O. ; Kratochvíl, Jaroslav ; Zámečník, J.
The classical DSC methods was employed to study aqueous solutions of sucrose in the whole concentration range and the temperature interval of -120 to 30 °C.
Thermal properties of dehydrated vegetable seeds
Zámečník, J. ; Černošková, E. ; Sikora, Antonín ; Bilavčík, A.
On the base of DSC measurements of seeds with defined water activity was found, that radish seeds have lower amount of strongly bonded water that chives seeds.
Repeated dehydration of chive and radish seeds at room temperature
Černošková, E. ; Zámečník, J. ; Zima, Vítězslav
Water content in seeds of radish and chive was studied by using of DSC and TG methods.
Thermal stability of plant seeds with low storage stability
Zámečník, J. ; Černošková, E. ; Sikora, Antonín ; Faberová, I.
DSC method was used to study of changes on seeds during storage at low temperatures.
Extracellular space volume fraction and geometry in human cortex and glial tumors
Homola, Aleš ; Vargová, Lýdia ; Zámečník, J. ; Tichý, M. ; Beneš, V. ; Syková, Eva
Ecs diffusion parameters in human temporal cortex similar to those previously determined in rat cortex. Volume fraction ? and tortuosity ? are increased in astrocytic gliomas and the higher the glioma´s WHO grade the greater the ECS volume and tortuosity increase. Migration and invasiveness correlate with an increase in ECS volume.
