Biotic elicitation of explant culture of Trifolium pratense L.

1 ABSTRACT Lenka Zdražilová Biotic Elicitation of the Explant Culture of Trifolium pratense L. Summary The paper examined the effect of four concentrations of salicylic acid on the production of flavonoids and isoflavonoids by the Trifolium pratense L. suspension culture (variety DO-8, variety DO-9). The cultures were cultivated in the Gamborg nutrient media with the addition of 2 mg.l-1 of 2,4-dichlorophenoxyacetic acid and 2 mg.l-1 of 6-benzylaminopurine, at the temperature of 25řC, 16-hr light / 8-hr dark period. The elicited and the inspection samples underwent the photometric determination of flavonoids in accordance with the Czech Pharmacopoeia 2005 and the determination of isoflavonoids via the HPLC method. The results show that the optimal effect of salicylic acid on the production of flavonoids was manifested after a 6-hour application of a concentration of 10 mmol (variety DO-8) and after a 168-hour application of a concentration of 0,01 mmol (variety DO-9). The effect of this biotic elicitor on the production of isoflavonoids was manifested after a 48-hour application of a concentration of 0,01 mmol.
Study of the Metabolism in in Vitro Cultures of Canabis Sativa L.

I. Summary The literature review concerning Cannabis sativa L. was prepared with focus on the biologically active compounds. The scope of cell cultures biotechnology and application of modern instrumental analytical methods in the field of metabolomics was discussed. Based on the NMR and MS results, no cannabinoids were detected in the control or elicited cell suspension cultures of Cannabis sativa L. Also other secondary metabolites were not detected by NMR. Other more sensitive analytical methods and their combinations should be used for absolute evaluation of other secondary metabolites, if they are present. It can be concluded, that biosynthesis of the cannabinoids was not induced by pectin or jasmonic acid as elicitors in the suspension, callus or embryogenic tissue cultures of Cannabis sativa L. Main metabolites of certain growt phases were evaluated based on the quantification of produced metabolites and prepared growth phase curves. The impacts of elicitations were interpreted by PCA statistical method, with subsequent metabolite identification or structure elucidation. Maxima of possible 1 H NMR signals were identified and 2D NMR experiments were applied for signals of unknown compounds. Eye movements trainer

This work deals with proposal and realization prototype of electronic ophthalmogyric muscles trainer, designated to conventional therapy for some forms of strabismus, or like device for rehabilitation muscles after surgical intervention. Apparatus is based on stimulation ophthalmogyric system by the help of watching " running" lighted LED diode. Diploma work includes a short dissertation of strabismus, ophthalmogyric eye system, detailed description of mechanical and electronic parts of apparatus and first experience in the workplace with it.
