Martin Schwarzer, Ph.D. Title of diploma: Impact of selected probiotic bacteria on juvenile host growth in gnotobiotic mouse model This thesis expands understanding of the ability of Lactiplantibacillus plantarum WJL bacteria to support the growth of gnotobiotic hosts in models of chronic malnutrition. It was investigated whether other bacteria also have similar capabilities and whether these traits are species- and strain-specific. Strains of Bifidobacterium longum ssp. longum and Bifidobacterium adolescentis were tested in a gnotobiotic juvenile mouse model of chronic malnutrition and their impact on the host's somatotropic axis was monitored. Concurrently, it was analysed how malnutrition affects the bacteria in the host's gut and the level of its colonization. Germ-free C57BL/6J mice were mono-colonized via intragastric gavage with the aforementioned bacterial strains. The mice were mated after confirming stable colonization and F1 male offspring were weaned on the 21st day after birth onto an experimental diet with low protein and fat content. The effect of the combination was evaluated using the FIC index. Based on the calculation of the FIC index, the mutual interaction was further classified as synergistic, additive, indifferent or antagonistic. Methods: To evaluate the interaction of antibacterial drugs in combination, four antibiotics were selected and employed to a pair-wise combinations. The main antibiotic was daptomycin, which was combined with tigecycline, linezolid and vancomycin. The effect of pair-wise combinations was monitored against the clinical isolate Enterococcus faecium and against the collection bacterial strain Enterococcus faecalis (ATCC 29212). Hotel with sports background

The main subject of the diploma thesis is a new construction of the hotel with sporting facility in Kuřim. The Thesis is done in a form of a project documentation with all requests by the law and regulations included. The project is Composed of two buildings, a hotel and the sporting hall. The building is Located in slope in direction to south direction. The hotel building have four Storeys. The whole disposition solution of both buildings is connected. In a first floor is located the hotel restaurant, staff background, wellness and fitness Secon and third floor is supposed to be Used Mainly for accomodating of Sportsmans, and the last fourth floor is for holding meetings and conferences. The main Supporting structure is done with as monolitic frame construction of beams, columns and continuous slabs. All roofs are done as mono-layer roofs. First floor of the sporting hall is used by people during the sporting activity. This area is composed of changing rooms with a social background and other spaces for the proper function of the hall. The main Supporting construction of the hall is designed as glued lamellar beams supported by columns.

This model seems to be more financially favorable and more ethically acceptable, which is an essential fact in the current era of increasing pressure to abandon testing on vertebrates in particular. The experimental part of this thesis is focused on optimizing the utilization of the G. mellonella larvae as a model organism within the research of fungal burn wounds fungal infections induced by the yeast Candida albicans. Within these experiments, larvae were used to preparation of tissue explants on which the formation of a single- species yeast biofilm was investigated. Methods: Before obtaining of tissue explants, the larvae were decapitated. In the experimental work, the toxic impact of DMSO on larvae was evaluated not only from the point of view of mortality. Much attention was also paid to the characteristic features that are commonly observed during tests on Galleria mellonella larvae, such as the degree of melanization, the impact on activity and cocoon formation. Methods: For individual experiments, three test groups of six individuals each were created based on weight, one group of larvae weighing 250 ± 10 mg, the second group weighing 350 ± 15 mg and the third group weighing 450 ± 15 mg. Using a Hamilton syringe, a volume of 10 µl of the test solution with the appropriate concentration of DMSO was administered to each larva directly into the haemocoel through the last left proleg. This bachelor thesis deals with the issue of social farming in the Czech Republic and in Europe, and the approximation of the functioning of social farming in practice on a selected subject. The bachelor thesis has a theoretical and practical part. In the theoretical part, a literature research on the topic was developed and in the practical part, the functioning of the social farm Květná Zahrada z.ú., was described and evaluated in detail using the method of field survey, semi-structured interview and SWOT analysis. The SWOT analysis identified a predominance of strengths that the farm can use to fulfil the opportunities and the outputs of the SWOT analysis, semi-standardised interview and field survey confirmed the effective functioning of social farm Květná Zahrada in the concept of social farming in the Czech Republic.

Other aim was to divide the patients into groups according to the serological results, then evaluate the prevalence of EBV and evaluate the incidence of primary EBV infection in the tested group of patients for the years 2019-2022. Methods: Chemiluminescence immunoassay and indirect immunofluorescence were used to determine specific antibodies against EBV. In some cases, the examination was completed by determining the avidity of anti-VCA IgG antibodies and the microblot array method. Real-time PCR was used to detect EBV DNA. Microsoft Excel 2016 was used to process data and to create tables and graphs. The goal of the theoretical part of the thesis was to focus on a comprehensive assessment of the issue of resistance to the current therapeutics used in the fight against resistant mycobacterial strains, to summarize the basic characteristics of the genus Mycobacterium, to address resistant TBC and NTM, and to describe the mechanisms of action and resistance known so far for individual therapeutics, which are often interrelated.
