This bachelor thesis deals with physical phenomena in electron microscopy, the construction of a scanning electron microscope and its operation. It also describes sample preparation and its specifics in cryogenic electron microscopy. An important part of the work is the description of image and contrast formation, including charging and curtaining artifacts. The experimental part is dedicated to the preparation of lipids and their imaging in the cryogenic scanning electron microscope under different imaging conditions. The emphasis is mainly on the study of contrast and charging artifacts.
