Využití biopreparátů na bázi mikroorganismů k biologickému ošetření osiva vybraných kulturních plodin

The aim of the thesis was focused on the biological treatment of corn, soybean and cucumber seeds using the entomopathogenic fungus Beauveria bassiana, mycopara-sitic fungus Trichoderma virens and antagonistic bacteria Bacillus mycoides. A 0.5% solution of carboxymethylcellulose was used for seed coating, in which the concentration of spores in 1 ml was 1x106. After seed coating, the number of spores of filamentous per one seed was determined and also the influence of microorgan-isms on seed germination and seedling development was evaluated. The results show that the antagonistic bacterium B. mycoides and entomopathogenic fungi B. bassiana had a positive effect on the growth and development of seedlings of all tested crops. The mycoparasitic fungus T. virens slightly retarded the development of roots and shoots in corn and soybean during germination. On the other hand, cu-cumber seeds coated by T. virens showed a faster development on both parameters evaluated. By sowing coated seeds with useful microorganisms, which can be easily introduced into the soil in addition increasing soil suppressiveness. Thus, seedlings can be protected against soil-dwelling pests or plant pathogens.
Use of biochemical methods in the study of plant metabolism with emphasis on the defense system

As sessile organisms, plants cannot avoid being exposed to various biotic and abiotic stress factors. To defend themselves plants have developed complex and sophisticated defense mechanisms. In agriculture, pesticides are used for plant protection, but they can have undesirable side effects, and therefore attention is being paid to biological control agents. One such agent is Pythium oligandrum, a soil oomycete that has a positive effect on plants at several levels. On the one hand, it secretes elicitins, which stimulate defensive responses and prepare the plant for possible infection (i.e., priming effect), and on the other hand, it produces tryptamine, a precursor of auxin, thus supporting the plant's growth. Furthermore, P. oligandrum directly acts as pathogen mycoparasite or evokes antibiosis, or competition for nutrients and space with other microorganisms. The key research topic of this thesis was the analysis of the biochemical properties of eleven Pythium strains and their impact on plant metabolism. Study of enzymes secreted by oomycete Pythium oligandrum

Pythium oligandrum is one of the non-pathogenic microorganisms of Pythium species which found its use as biological control agent. The main modes of action of this oomycete lie in inducing the plant immunity and in mycoparasitism of pathogenic fungi and fungus-like organisms. The oomycete attacks the pathogens by secretion of hydrolytic enzymes into the environment. In the first part of this bachelor's thesis, activity of hydrolytic enzymes endo-1,3-β-glucanase, cellulase, chitinase and proteases was observed in a commercial product based on Pythium oligandrum, called Polyversum-Biogarden. It was conclu- ded that the direct hydrolysis of phytopathogens is probably not the main mechanism of this product because the activity of glycosidases increased only after six hour-long incubation of the Polyversum-Biogarden in water and the proteolytic activity was not detected. In the second part of the bachelor's thesis, properties of proteases secreted by orga- nism Pythium oligandrum were studied. The highest proteolytic activity was observed at pH 6,5. Ovomucoid acted as an inhibitor of secreted proteases. The stability of pro- teases was lowered by SDS, detergents present in liquid soaps and by a solution of a solid soap or NaOH. Conference proceedings on biological plant protection. Contributions were presented on the influence of biological seed dressing for wheat on the composition of soil micromycetes, efficacy of botanical insecticides based on seed oil from Pongania glabra Vent. against Plutella xylostella L. larvae, supressive effect of selected plant species on the germination and foliar necrosis, induced resistence of wheat to powdery mildew.

The complex characterisation of nematode Phasmarhabditis hermaphrodita

Ph.D. thesis "The complex chacterisation of the nematode Phasmarhabditis hermpahrodita" is focused on the study of ecology of slug parasitic nematode P. hermaphrodita, its general characterisation in the context of all nowadays knowledge. The thesis consists the general introduction, that introduces the whole nowadays research of P. hermaphrodita, and four original works, that follow the introductory chapter. The first work deals with the orientation of P. hermaphrodita in 2D and 3D space and reaction on the presence of different hosts or nutritive substrates. The other study deals with the reaction of P. hermaphrodita on various animal and plant nutritive substrates and in the first place the influence of these substrates on the development of nematodes and quality and quantity of progeny. The third scope of this thesis is the influence of intraspecific competition on development, quality and quantity of nematodes and the last part of this text comprises a short study of the persistence of P. hermaphrodita in different substrates, from mineral to organic.
