The topic of this work was studied genetic diversity and phylogeny of selected wild species Lathyrus neurolobus Boiss. & Heldr., which is closely related to cultivated pea, using modern methods of molecular phylogenetics. Genotypes of L. neurolobus were obtained from Department of Botany at Palacký University in Olomouc, genomic DNA was isolated of these genotypes and samples were subjected to analysis using SSR and iPBS markers. It was found that for assessment of genetic diversity is suitable method of iPBS markers, where the average value of polymorphic information content is very high (PIC=0,928). According to evaluated markers polymorphism and assembled dendrograms was found that for search of relationship between place of origin and placement in dendrogram, will be appropriate the method of SSR markers, but only in case of selection informative markers. There was also confirmed transferability of SSR markers designed for pea to wild species of another genus.

During the process of domestication and selection reduction of genetic diversity of cultivated crops occurred. Currently the main interest of breeders is to transfer some of crop wild relatives’ genes to cultivated crops. Those genes of interest are related e.g. with desease and pest resistance or drought resistance. Hybridization of wild and cultivated species is prevented by reproductive isolation. The frequent phonomenon in hybrids is called nuclear-cytoplasmic incompatibility, which is manifested by reduced fertility, sterility or lethality. In this phenomenon occurs conflict between nuclear-encoded genes with genes encoded in the organellar genomes. The identified accD candidate gene responsible for nuclear-cytoplasmic incompatibility in pea is highly variable due to insertions and deletions. High variability of the accD gene was also confirmed in the genera Lathyrus and Vicia. Variability was observed in gene sequence lenghts caused by presence of indels and single nucleotide polymorphisms. In comparative analysis with other regions of cpDNA commonly used in phylogenetics, the region of the accD gene had the highest value of parsimonially informative sites. The phylogeny derived from the region of the accD gene corresponds to the phylogeny based on combined chloroplast markers, therefore the accD gene may be suitable for this type of analysis.
Creation/Work of Jaroslav Foglar as a starting point for the use of dramatic education methods at the first elementary school

1 ABSTRACT The diploma thesis deals with the possibility of using Jaroslav Foglar's work in dramatic education at elementary school.The aim of the diploma thesis is to verify the influence of methods and techniques of dramatic education through Jaroslav Foglar's texts in the form of value charts and attitudes in the life of pupils of the 5th year of primary school.How dramatic education helps with methods and techniques - first fordeepest adoption of Jaroslav Foglar's work, and second - to increase understanding and adoption of given values and attitudes.The theoretical part focuses on the personality and work of Jaroslav Foglar and his timeless message. It focuses on the concept of drama education, its aims, principles, forms, methods and techniques. At the same time, it focuses on the analysis of the work in terms of dramatic education. In the practical part of the diploma thesis, three thematic units focusing on the moral qualities of the human being, using methods and techniques of dramatic education, are elaborated on the basis of books and short stories by Jaroslav Foglar. Analysis of gender roles and stereotypes on social network Instagram

This diploma thesis deals with the position of women and men on the Internet social network called Instagram. The theme is first described from a theoretical point of view, explaining what web 2.0 is, what social networks are and how social networking helps with our socialization to society and our self-presentation. In this part are also explained the concepts of gender and gender stereotypes. My own research consist of three parts. The first part is a qualitative image analysis of so-called "selfie" photos of selected Instagram users, which revealed, that the women that I analyzed supported gender stereotypes in female imaginary, while analyzed men occasionally oscillated between masculinity and femininity. The second part is the analysis of the number of the likes under these "selfie" photos and the third part is the analysis of deep semi-structured interviews, which showed that women and men have much in common on Instagram. But overall, their position on this network according to my analysis is not the same.
Prion disease in human - Creutzfeldt-Jakob disease

Magdalena Smětáková Title of diploma thesis: Human prion disease - CREUTZFELDT-JAKOB DISEASE Objective: Theoretical part of the thesis includes a comprehensive theory not only about prions, but also about Creutzfeldt-Jakob disease in general. The aim of the experimental part was to evaluate and quantify the cerebrospinal fluid and brain tissue of eight individuals suspected from prion disease, Creutzfeldt-Jakob disease specifically. Methods: To determine the presence of 14-3-3 protein in cerebrospinal fluid and prion protein in the native brain tissue an analytical method called Western blotting was used. Results were expressed only qualitatively. For better differential diagnosis, immunological examination of so-called CSF triplet (p-TAU, h-TAU, β-amyloid) was completed. The quantitative determination was performed by enzyme immunoassay (ELISA). Results: All eight subjects suspected from the prion disease were confirmed with the diagnosis of Creutzfeldt-Jakob disease.
