Cloning, expression and characterization of human serine racemase mutants

AAbbssttrraacctt Human serine racemase (hSR) is a cytosolic pyridoxal-5'-phosphate dependent enzyme localized in the central nervous system. It synthesizes D-serine, which is an endogenous coagonist for the N-methyl-D-aspartate (NMDA) receptors and plays a key role in excitatory neurotransmission in the brain. Thus, human serine racemase is a promising target for the treatment of neurodegenerative diseases connected with NMDA receptors. However, few specific inhibitors have been identified to date and the crystal structure of hSR has become available only very recently. We decided to perform a random mutagenesis to determine the amino acid residues critical for the enzyme activity. Ser 84 was reported as a catalytic residue along with Lys 56. After analysis of a double mutant S84G/P111L which retained its capability to convert L-serine to pyruvate, we prepared and characterized the single mutant S84G in order to exclude potential effect of the P111L mutation.on the activity of the analyzed enzyme. Evaluation of quality of life in patients with DM with different methods of insulin delivery.

This thesis deals with the quality of life in patients with diabetes mellitus first type with different routes of administration of insulin. The work is divided into two parts, theoretical and research. The first describes the theoretical overall diabetes mellitus, as well as its basic distribution, complications and treatment options, focusing primarily on treatment with an insulin pump. It also outlines the concept of quality of life, both individually and in association with chronic disease. The second part is devoted to research, whose main objective is based on information obtained through a questionnaire survey to determine whether the application of insulin using an insulin pump improves the quality of life in patients with diabetes mellitus first types. The Universe, Space and Man

The thesis "The Universe, Space and Man" deals with dilemma of visual phenomena in The Universe. The aim is to point out the relation between Man and the Cosmos, man's position in this apparently horizon less space and, to describe the basic cosmic visual phenomena and show their potential interpretation in the context of Art Culture. The work looks into the phenomena from an astronomical point of view (in accordance with latest findings of today's science), from an astrological and mythological point of view (integral components in history of religious cults), using visual art qualities (color, form, light) and finally, through the interpretative eyes of an artist. Work includes author's individual views on the given dilemma presented in her own creation (series of paintings). The didactic application of the topic has also been presented (teaching project).

Thesis "Foreign exchange exposure and currency risk" deals with managing foreign currency exposure and foreign exchange risks when doing business in The Czech Republic. It defines foreign exchange risk, different types of foreign exchange exposures and the possibility of its ensuring, as well as internal and external methods of reducing foreign exchange exposure and foreign exchange risk. The practical part is devoted to a particular solution, respectively to the management of foreign exchange exposure and foreign currency risk in ABC, s.r.o. company.

This bachelor thesis deals with the distribution of net profit of commercial companies. The aim of my work is to show the clearest procedure how to work with the profit in period from its recognition to its settlement. My thesis is divided into eleven parts. In the first part, I focus on a theoretical definition of commercial companies according to the Commercial Code. The remaining parts are designed to cover the whole process - from a recognition of the net profit through its approval and until its distribution. The practical part verifies the theory in practice and summarizes my findings relating to the joint-stock company Expert & Partner Engineering a.s. and copartnership DISA v.o.s.
