Development of molecular selection markers for detection of fertility restorer for CMS Shaan 2A

This bachelor thesis deals with the development of molecular selection markers for the detection of fertility restorers for CMS Shaan 2A in rape by molecular techniques. The thesis describes molecular techniques such as DNA isolation techniques, polymerase chain reaction and primer design techniques for the detection of fertility restorers. Specific PCR primers for the Pentatricopeptide repeat (PPR) superfamily protein gene and the Tetratricopeptide repeat (TPR) -like superfamily protein gene sequence amplification have been designed and aplicons have been sequenced. On the basis of the sequence differences between plants with fertility restorer fenotype and CMS fenotype the technique for fertility restorer detection was designed.
The health risks associated with the \kur{Listeria genus}

This bachelor thesis is concerned with the health risks associated with the genus Listeria, especially with the species Listeria monocytogenes and Listeria ivanovii inducing listeriosis. As for humans, listeriosis occurs rather sporadically, nevertheless, it can cause outbreaks, listeriosis can also become a hospital infection. Listeriosis is dangerous primarily for pregnant women where it can cause abortions and for individuals with weakened immune systems where it may even cause encephalitis and meningitis. Currently a growing prevalence of bacteria resistant to antimicrobials is observed, including bacteria Listeria monocytogenes which represents a serious problem. There is no listeriosis vaccination, the only prevention is a careful heat treatment of food and hygiene which minimize the risk of the infection.
