Elektrochemická DNA microarray jako nástroj pro sledování maligního potenciálu buněk

Prostate cancer is a heterogeneous tumour entity with known interindividual differences in biological behaviour regarding tumour aggressiveness and metastatic potential. To identify the molecular causes behind these differences, the gene expression profiles of three cell lines derived from prostate epithelium – PNT1A (benign), 22Rv1 (malignant) and prostate carcinoma bone metastasis – PC-3 were examined using electrochemical DNA microarray technology. Most of up-regulated genes associated with stimulation of cell cycle were found to be common in malignant and metastatic cells, while the down-regulated genes were involved in a negative regulation of cell proliferation. Further, the cell lines were also treated with sarcosine, which is a potential oncometabolite and their differential expression profiles were compared. In all cell lines examined, most of the genes affected by sarcosine are involved in stimulation of a cell cycle and negative regulation of apoptosis, which are both hallmarks of cancer. These finding further support oncogenic potential of sarcosine and may contribute to a better understanding of the mechanisms of prostate cancer and expression profiles.

The objective of this diploma thesis is to analyse and evaluate the utilisation of social and technological innovations in small and medium enterprises in Blansko. The theoretical part is based on general information and definitions about the term innovation, social innovation and technological innovation. It presents various models of theirs comprehension and a historical overview. The practical part focuses on quantitative research of enterprise's social and technological innovation use in the selected town. On the basis of the survey practical recommendations for the municipality and enterprises were defined, they should help to improve the use of social and technological innovations in Blansko.
