Interaction of protein subunits SEC10 and SEC15 of the exocyst tethering complex
Bartáková, Anna ; Ryšlavá, Helena (advisor) ; Čermáková, Michaela (referee)
The exocyst is an evolutionarily conserved tethering complex involved in the regulation of the secretory pathway in eukaryotic cells. As an effector of Small GTPases, the exocyst contributes to efficient targeting of secretory vesicles to the sites of intense exocytosis at the plasmatic membrane through interactions with specific membrane phospholipids. The exocyst complex consists of eight subunits: SEC3, SEC5, SEC6, SEC8, SEC10, SEC15, EXO70, and EXO84. Each subunit binds at least two other subunits, among them the interaction of SEC10 and SEC15 is crucial for the exocyst function. The question remains how this particular interaction is evolutionarily conserved across different plant taxonomic groups. SEC10 and SEC15 genes from the three evolutionary groups of plants (green algae - Klebsormidium nitens, mosses - Marchantia polymorpha, angiosperms - Arabidopsis thaliana) were cloned into yeast two-hybrid vectors for studying protein interactions. Testing the interaction of SEC10 and SEC15 exocyst subunits from different plant species in the yeast-two hybrid system showed that despite partially different protein sequences of the tested subunits, their interaction across different evolutionary groups of plants remains very well conserved.
Analysis of relations between NATO and Russia in terms of energy security
Bartáková, Anna ; Dubský, Zbyněk (advisor) ; Kuchařová, Alžběta (referee)
Am of the thesis is to analyze the development of the relations between NATO and Russia since the eastern enlargement of NATO in 2004 until the formation of a New Strategic Concept in 2010 with an emphasis on theenergy diplomacy of Russia in the post-Soviet region. The functioning of Russian energy diplomacy is illustrated by the example of two Ukrainian gas wars. Objective of the thesis is to analyze how is Russia using its energy resources at the bilateral level (especially in the Eastern European region) and its influence towards NATO. There is an instrumental nature in understanding of the energy policy by Russia, which subsequently affects the potential expansion of NATO in the post-Soviet space. The conclusion is that European NATO and the U.S. should seek to maximize the diversification of energy resources to stabilize their negotiating position with the Russians.
Conception of Leadership in Fascist Italy and Nazi Germany
Bartáková, Anna ; Reschová, Jana (advisor) ; Prorok, Vladimír (referee)
Principle aim of the thesis is to examine different approaches towards the distribution of power between the Leader and the party in Fascist Italy and Nazi Germany using a method of historical comparison. Through comparison of ideological bases, ways of establishment and exercise of leader principle by both regimes, the way Fascists and Nazists fulfill their ideological concept in reality is being implied . Key result of the thesis is detecting that German Nazis achieved their ideals more comprehensively than italian Fascist did. The first part defines both ideologies, the second one pertains to concept of the state, the third chapter describes the leader principle, in fourth one the function of the party is outlined and fifth part gives a definition of relation between the Leader and the party. Contribution of the thesis lies in determination of historical phenomena, which represent the potential menace to contemporary democratic systems and society.
