The role of Kit ligands in hematopoiesis of Danio rerio

Hematopoiesis is a precisely regulated process, dependent on the activity of hematopoietic cytokines and their receptors. Due to an extra round of whole genome duplication in teleost fish, two paralogs of many important genes, including some hematopoietic cytokines and their receptors, are present in the zebrafish (Danio rerio) genome. In this project, we have been investigating the role of zebrafish Kit ligands in hematopoiesis. Kit ligand is a pleiotropic cytokine, which is essential for vertebrate erythropoiesis; however, in zebrafish, no such role has been reported so far. To determine the function of zebrafish paralogs of Kit ligand (Kitlga and Kitlgb) in hematopoiesis, we performed in vivo and ex vivo gain- and loss-of-function experiments. Strikingly, we were the first to report the synergistic cooperation of zebrafish Kitlga with erythropoietin and dexamethasone, enabling the growth of kidney marrow-derived suspension cells and providing optimal conditions for the expansion of adult erythroid progenitors. It provides drug information to healthcare professionals in the form of timely and accurate answers to drug-related enquiries, including drug interactions (DI). Aim: This study aimed to analyse enquiries related to DI processed by DIC from 2015 to 2020 and to solve three drug enquiries related to drug interactions. Methods: Quantitative and qualitative analysis of drug enquiries was carried out. Data was collected from individual enquiries related, but not limited to drug-drug, drug-herbal, and drug-disease interactions. In the early phases of drug design and development, scientists must overcome many challenges involved in identifying potential drug-like or lead-like compounds. This has led to the need of creating large sets of chemical data which will aid in improving the identification of pharmacophores and active compounds. Various scientific fields especially pharmacology, medicinal chemistry and biochemistry have begun to employ the use of computer sciences to aid in the screening for potential leads with more specificity with regards to drug-like compounds' or substances' bioactivity. The emphasis of this project was to create a database containing a collection of pyrazine compounds synthesized overtime in the Faculty of Pharmacy (Charles University, Hradec Kralove) with the aim of having anti-mycobacterium (and possible antibacterial and antifungal) activity, and further utilize this database to predict certain pharmacokinetic and bioavailability properties. The diploma thesis describes the design of your own web application within the long-term collaborative project co.llaborate.online. In a small team, we reflect on our own workflow and designer work. Currently, the common goal is the design and development of MVP for processing text, images and other graphic material into any graphic output - graphic editor co.design. As part of the project, we have also focused for a long time on the concept of compatible MVPs for text editing or data management and user research. The starting point of co.llaborate.online is the principle of DRY (Don't repeat yourself) and the principles of web technologies. We test how certain workflows can be automated and facilitate the collaboration of designers, text authors, editors, proofreaders, etc., or emancipate users from unnecessary activities.

Výsledkom práce je databáza FireProtDB, ktorá poskytuje manuálne overené experimentálne dáta z dostupných zdrojov a implementuje grafické užívateľské rozhranie, ktoré poskytuje dôležité informácie o dátach spoločne s možnosťou vyhľadávania umožňujúcim vytvárať dotazy na mieru a cieliacim na užívateľov, ktorí hľadajú dáta pre vytváranie dátových sád pre nástroje využívajúce strojové učenie.

Design and implementation of online training diary for athletes is the main goal of this thesis. At the beginning, it was necessary to explain some of key words, related to the thesis topic. One of the most important things is data mining and its usability for sports data analysis. After that, existing solutions of sport applications were analyzed and also it was obligatory to analyze potential users requirements. Application design, implementation and testing were the next steps. Some of data mining methods were used for analysis of sports data intended for individual athletes and their coaches.

This bachelor thesis describes the design of relational database for a small company engaged in sales and authorized repairs of sensitive electronic devices. Main target of the database solution will be the support and increase of process efficiency focused mainly on device maintenance and its planning within the company. Thesis is composed of three consecutive parts. First part defines theoretical foundations needed for understanding the issue of data modeling. The second part deals with the analysis of a current state of the company. Data model, design of which is the matter of third part of this thesis, will be built on the outcome of analysis of the company and its internal processes. Thesis then contains evaluation and expected benefits for the company.
