Indukce triploidie u candáta obecného (Sander lucioperca)

The aim of this study was to induce the triploidy in pikeperch (Sander lucioperca) with use of a cold shock. To induce the triploidy, fertilised egg were (spawning temperature 14,5 °C) submerged in a cold bath at the temperature of 2 °C. Time of initiation was 1; 3; 5; 7 and 10 minutes post activation. The exposure time was 20 and 40 minutes. Ploidy level of freshly hatched larvae was assessed with use of the flow cytometry. In both exposure times, the hatching rate was getting lower with later time of initiation (20 minutes exposure, hatching rate: 58,4-13,4 %; 40 minutes exposure, hatching rate: 28- 9,6 %). Number of malformed larvae increased with later time of initiation and longer exposure time (20 minutes exposure, malformed larvae 0-47,2 %; 40 minutes exposure, malformed larvae 0-58,8 %). None of the tested combination of exposure time and time of initiation led to a population containing 100 % triploid larvae. However, percentage of triploid larvae grew up with longer exposure time and later time of initiation. The best cold shock combination with highest yield of triploids were after 20 minutes long treatment initiated 10 minutes post activation (57,1 +- 14,2 %) and after 40 minutes long treatment initiated 10 minutes post activation (61,9 +- 8,2 %). The most important finding of this study is that cold shock treatment leads to triploidy in pikeperch. To obtain 100% triploid larvae, shorter exposure time and different shock temperature might be applied. It may also eliminate low hatching rate and high appearance of malformed larvae.
Multiple-criteria Analysis in a Choice of an Employer

This thesis deals with the issue of decision-making process in recruitment and selection of employees using methods of multi-criteria analysis options. The theoretical part of the thesis is focused on explaining the concepts of human resources and HR management. This area describes in detail the activities of selection process. Another area defines the basics of project management and project manager job. The third part deals with the theory of multi-criteria decision and the last area describes the process models. The practical part of the thesis uses the knowledge gained in the theoretical part for the real application to selection process. First, the company selected is introduced, especially its history, organizational structure and the current selection process. Then the current selection process is analysed and the results of analysis initiate the creation of a new process. For achieving the higher quality of results are included and applied methods of multi-criteria analysis of options and the questionnaire method in the new process. In conclusion, the achieved results are discussed with the manager and the current selection process is compared with a new selection process.
Comparison of the effectiveness and efficiency of the four common anesthetic used in fisheries for burbot (Lota lota L.)

The aim of this study was to compare the effect of four anaesthetics on haematological and biochemical blood profiles in burbot (Lota lota L.). Both blood profiles of burbot were evaluated 10 min and 24 h after anaesthesia with MS-222 (100 mg.L-1), clove oil (33 mg.L-1), 2-phenoxyethanol (0.30 ml.L-1), Propiscin (1 ml.L-1) and were compared to non-anaesthetised control. The tested anaesthetics did not have any effect on haematological profile of burbot. The exposure to clove oil, 2-phenoxyethanol and Propiscin significantly influenced level of ammonia and glucose. The level of lactate was significantly increased following anaesthesia with 2-phenoxyethanol and Propiscin. The levels of total protein, aspartate aminotransferase and calcium were higher with clove oil, 2-phenoxyethanol and Propiscin compare to control. The use of MS-222 showed the smallest effect on haematological and biochemical blood profile and is recommended as a suitable anaesthetic for burbot.
