Unravelling molecular mechanisms of the abasic crosslink DNA repair

4 Abstract DNA as the primary carrier of genetic information guarantees organisms to live, grow, develop and reproduce. However, this most vital molecule in the cell is subject to various damages every moment. If it is not repaired, the cell and the organism will eventually succumb to inevitable destruction. One of the most serious damages is abasic site interstrand crosslink (Ap-ICL). Ap-ICL is formed spontaneously when an abasic site covalently pairs with an adenine on the opposite strand. The lack of information on repair mechanisms, the influence of the local sequence and its stability leads to questions about the fate, toxicity and occurrence of these lesions in cells. During evolution, several mechanisms have evolved to repair these and other damages to ensure the organism's survival. A recently discovered pathway known to repair Ap-ICL is named after the DNA glycosylase responsible for removing Ap-ICL. NEIL3 DNA glycosylase is recruited to Ap-ICL by ubiquitylation of DNA helicase, which is part of the DNA replication complex. NEIL3 glycosylase contains several zinc-finger domains, that bind to the damaged DNA and ensure its catalytic function. Design of parking place in crematorium of Brno cemetry

The subject of this thesis is to solve the problem of the lack of the parking places at the city crematorium of the Central Cemetery of Brno. The main task is to deploy the parking lots on the northern and western side of the Central Cemetery. The parking lot on the western side should take into consideration the proposal for a new entrance from this side. This thesis also includes solutions for wheelchair access and for building a new bus stop.

The Bachelor´s thesis analyzes both general progress, as well as a detailed analysis of the specific contract, with an emphasis on quality and cost. Consecutively it indicates that should help to promote consumer awareness and effectiveness during the engagement.

Parking garage in the University hospital in Brno – Bohunice

The thesis is draft a multi-storey parking garage in the area of University Hospital in Brno Bohunice to insufficient capacity of the parking spaces. The aim of thesis is to create a sufficient number of parking spaces, which would solve the deficit of parking spaces and connecting the street Jihlavská. The parking garage is located on the south side of the University Hospital and ensures pedestrian connections to the hospital. The parking garage should serve for patient, visitors and hospital staff. This thesis takes into consideration the solution of terminal of public transport in Bohunice’s hospital and for patients and visitors to the hospital is also designed park.

Connection of Korejska and Hradecka street in Brno

Primary subject of this master`s thesis is to manage increasing traffic volumes between streets Korejská and Zborovská. The most appropriate solution seems to be connecting the Korejská and Hradecká street. Next aim of this work is to design terminus station and transfer terminal for passengers between trolleybus (lines n.34 and n.36) and North-south rail diameter (future express train line connecting north and south suburbs with city centre).

Road design of /43 in section Podlesí - Kuřim

This diploma thesis deals with the processing of complex transport solutions in the interest area of the city of Kuřim on the existing road I/43 in the Section Česká – Podlesí – Kuřim. Thus, the proposal for an adjustment of this road takes into account all modes of transport. Transit trafic or target car transport, public transport and also transport in terms of local ties. The design is processed in 4 variants, the difference being mainly in the size of the modifications to the existing road I/43. Subsequently one variant was selected for more detailed completion. This is a design solution in the 2 + 1 arrangement with interchanges of Podlesí and Kuřim – east. The proposed solution does not prevent the planned implementation of the D43 motorway.

Reconstruction of crossing - Brno, Míru square

This master's degree thesis is focused on the traffic problem at Míru square in the city of Brno where the intersection with tram terminus turning loop is located. Thesis is based on local engineering survey with the outcome of six different design variants. The variant with the tram turning loop displaced north-west was chosen for detailed design with the outcome of full technical documentation and drawings.

Apartment building

The subject of this bachelor thesis is the elaboration of project documentation for the construction of an apartment building. It is a detached basement building, which is located on a rectangular plot with flat terrain. The building is designed with four floors and one underground floor. This new building is equipped with a lift for 8 people. In the underground floor there is a technical background of the building and a storage cellars for individual apartments. There are 12 flats with different disposition and size of floor areas. The load-bearing structural system is diagonal. The lower structure is made of a waterproof concrete, the so-called "white tub" which consists of a reinforced concrete foundation slab and reinforced concrete basement walls. The upper structure is built of Porotherm system, which is insulated with ETICS facade contact system with basalt wool insulation boards. Ceiling structures are designed as monolithic reinforced concrete slabs and the staircase is built as a reinforced concrete monolithic three-armed. The façade is shaped according to the partially opened loggias. The last floor recedes from the face of the building and creates spacious terraces. Holes are filled with wooden windows. The roofing of the apartment building is solved by a single-layer flat roof with a classical layer arrangement and a hydro isolation layer made of asphalt strips.

Public Transport Hub Brno-Komárov

The subject of this work is to design a public transport hub in the area of Brno-Komarov in connection with a tram loop, aiming to increase comfort of transfer between transport lines and the possibility to create a bus terminus for more bus lines. This study involves designing of a new tram and bus loop and redesign of pedestrian routes for comfortable transition between public transport lines and for easy movement in the area. Attention is paid mainly to local relations in the area concerned, which are disconnected by the intersection of Svatopetrská/Hněvkovského/Černovická. The work also contemplates the planned extension of the tram line to the south of the city. The content of the work are the results of the traffic surveys carried out for these purposes and the variants of the possible layouts of the new transport hub, from which one variant was selected on the basis of a multiple-criteria evaluation and this variant drafted in the drawing documentation.
