Metabolically syndrome - threat of civilisation diseases

The aim of the bachelor's thesis is the metabolic syndrome as a threat of civilization diseases. The thesis is divided into a theoretical and an empirical part. The theoretical part is divided into two chapters . The first chapter deals with very general civilization diseases, the causes, and the prevention of various diseases. The second chapter is devoted to comprehensive metabolic syndrome, its characteristics, etiology, occurrence, diagnosis, treatment and prevention. The empirical part, implemented quantitative research, based on a questionnaire survey of 102 randomly selected respondents, patients of internal departments and private ambulance Hospital Brandys nad Labem with the objectives to determine how patients are informed about MS who provided them the information and whether they are interested in a greater awareness of the issues. It also determines whether observance of the regime measures for the prevention of MS. The survey shows that respondents are informed about MS the most from the medical staff and the media. There was less than half of them who expressed interest in additional information about MS. From the results it is also clear that respondents fully respected preventive measures regime. Targeted analysis of branched-chain faty acid esters by LC/MS

Lipidomics as a part of metabolomics is a fast-growing area of research due to the improvement in analytical techniques. This master thesis is focused on lipid extraction techniques optimization, using liquid liquid extraction and solid phase extraction as pre-separation methods and ultra performance liquid chromatography coupled with mass spectrometry for extraction and subsequent identification of branched-chain fatty acid esters (FAHFA - branched-chain Fatty Acid esters of Hydroxy Fatty Acids). This newly discovered class of lipid molecules is associated with insulin secretion, which could improve whole body and local glucose metabolism, providing potential for diabetes 2 type treatment. Solid phase extraction of biological samples was optimized on columns regarding to sorbent composition using reversed phase columns with modified styrene divinylbenzene polymer or octadecyl-bonded polymer and normal phase columns packed with silica gel. Column Strata SI-1 Silica was the most effective for FAHFA separation from biological samples. Chromatographic separation of FAHFA was performed on UPLC Ultimate 3000 RSLC equipped with Kinetex C18 1,7 µm, 2,1 x 150 mm column using gradient program. Analysis of novel members of FAHFA lipid class derived from omega 3-PUFA by LC-MS/MS and LC-MS/MS/MS

White adipose tissue (WAT) is a complex organ with both metabolic and endocrine functions. Dysregulation of all of these functions of WAT, together with low-grade inflammation of the tissue in obesity, contributes to the development of insulin resistance and type 2 diabetes. Omega-3 polyunsaturated fatty acids (PUFA) of marine origin play an important role in resolution of inflammation and exert beneficial metabolic effects. Using LC-MS/MS method, we elucidated the structures of novel members of fatty acid esters of hydroxy fatty acids (FAHFA), which were present in murine and human serum and WAT after omega-3 PUFA supplementation in diet. These compounds contained DHA esterified to 9- and 13-hydroxyoctadecadienoic acid (HLA) termed 9-DHAHLA and 13-DHAHLA, and were synthesized in WAT. Our results document the existence of novel lipid mediators, which are involved in the beneficial anti-inflammatory effects attributed toomega-3 PUFA, in both miceandhumans.

The bachelor{\crq}s thesis is divided into theoretical and practical part. The theoretical part deals with the terms: creativity, creative person, adolescence, free time education and pedagogy of experience. It introduces the circumstances of the pedagogical process. It analyzes the term ``art{\crqq} and its role in human{\crq}s life. It introduces the role and the function of creative, esthetic and ecological education during the development of the adolescent{\crq}s creativity. The theoretical part of the thesis is based on the professional literature and on theoretical knowledge of this theme. In the practical part, the knowledge is applied to work with a concrete group of adolescents. It shows the use of theoretical base and experiences gained during preparation and carrying out creative activities that should lead to the development of social abilities and to reestablishing the relationship to nature.
