Work with children with postponement of school entrance in current practice of pre-school education

In my bachelor's thesis I am dealing with issues related to the postponement of compulsory school attendance. I have researched the topic in accordance to the new conditions - the introduction of the compulsory pre-school education of five-year-old childern. The thesis is divided into two main parts − a theoretical part and a practical one. The purpose of the theoretical part is to introduce the topic. The development of the view on the concepts of school maturity, readiness and competence, and the legislative legalisation of the postponement of beginning of the schol attendance is presented here. It also outlines the development of postponements of school attendance from approximately, the middle od the 20th century to the present, and a brief comparison of the Czech Republic and some other European countries concerning the issue of school attendance. Since the rate of postponements of compulsory school attendance in the Czech Republic is relatively high, the countries where the share of posponements is lower, we used for the comaprison. Survivin, a novel target of the Hedgehog/GLI signaling pathway in human tumor cells

Survivin, an important antiapoptotic protein, is expressed in tumors, whereas in normal tissues the expression of this protein is extremely low, defining a role for survivin as a cancer gene. Survivin exhibits multifunctional activity in tumor cells. However, why survivin expression is sharply and invariably restricted to tumor tissue remains unclear. Here, we identified 11 putative consensus binding sites for GLI transcription factors in the survivin promoter and characterized the promoter activity. Inhibitors of the Hedgehog/GLI pathway, cyclopamine and GANT61, decreased the promoter activity in reporter assays. ΔNGLI2 (which lacks the repressor domain) was the most potent vector in activating the survivin promoter-reporter. Moreover, GANT61, a GLI1/2 inhibitor, repressed endogenous survivin protein and mRNA expression in most cells across a large panel of tumor cell lines. Chromatin immunoprecipitation showed GLI2 binding to the survivin promoter. The ectopic GLI2-evoked expression of endogenous survivin was observed in normal human fibroblasts. Molecular mechanisms of the sensitivity of tumor cells to an inbibitor of protein kinase Src dasatinib

Signal transmission is an efficient mechanism that allows fast and specific response of eukaryotic cells to small changes in the extracellular and intracellular information. Many cascade of protein kinases in signaling pathways are deregulated in human diseases and represent a valuable target for their therapy. This work has been devoted to inhibit Src kinase activity after application of dasatinib on melanoma cells in cell culture. Dasatinib is multikinase inhibitor of protein tyrosine kinases, and with high efficiency inhibits Src and other members of this family of kinases. Dasatinib is already in use as anticancer drug in some tumors, while another is in late stage clinical trials. Results showed mainly the effect of Src inhibition on two main signaling pathways important for survival and proliferation of melanoma cells, MAPK and AKT. In the literature it is not known many links MAPK and Src at melamomu, but recently has been reported that the Src inhibiting translocation of ERK1 / 2 in the core, thereby accelerating the proliferation of melanoma cells.
Optimisation of HPLC clotrimazole determination

High-performance liquid chromatographic technique for the determination of clotrimazole and its degradation products (2-chlorophenyl)-diphenyl methanol and imidazole in the formulation Clotrimazol spray 1 % was developed. The presented thesis was focused to the selection of optimal column, composition of mobile phase, its pH and internal standard for the analysis of pharmaceutical formulation based on topically applied antifungal agent clotrimazole. Seven columns were tested in detail. The tested columns were: Discovery HS F5 (5 μm, 150 x 4.6 mm)Chromolith® Performance, RP-18e (100 x 3 mm)ZIC® HILIC (3.5 μm, 50 x 2.1 mm)XTerra® RP 18 (5 μm, 100 x 3 mm)Discovery RP Amide C 16 (5 μm, 250 x 3 mm)Zorbax Extend-C18 (3.5 μm, 75 x 4.6 mm)Discovery ZR-PBD (5 μm, 150 x 4.6 mm). HPLC was carried out using Discovery ZR- PBD column 5 μm, 150 x 4.6 mm. This column is based on polybutadiene-coated zirconium oxide. A zirconium oxide is stable in the whole pH range with the high pressure and temperature up to 200řC. Spectrophotometric detection at 210 nm was performed. The optimal mobile phase was a mixture of acetonitrile and water (pH 9.7) in the ratio of 50:50. The pH of the water component was finally adjusted to 9.7 with NH4OH 25 %. Breathing when playing the brass instruments

In my diploma thesis I deal with breathing when playing the brass instruments. In the first chapter called Process of Breathing I describe the respiratory system, its breathing mechanism and a separate breathing process with the main emphasis on the types of breathing, control of breathing and a lung capacity. Subsequently I occupy with the breathing process when playing, it means with breathing in cooperation with creation of tones, embouchure, activity of tongue and with the finger technique. In the second chapter I mention some important brass players and teachers of the breathing in brass instrumental music; the view of the East Cultures on ordinary breathing is also supplemented to this overview. In the third chapter I quote some methods of breathing development. Firstly I focus on the evolution and usage of various types of breathing, secondly on the methods of breathing development. This chapter includes many breathing exercises done with or without an instrument, which should lead to liberty and speed of inhalation and smoothness and holding of exhalation. The exercises help to the complete fastening of the muscles of respiration.
The Elements of playing the trombon in Primary music school

My work consists of researching today's current possibilities of education in Trombone at Basic artistic schools. In chapter one my primary focus of study is on the musical education development of playing the Trombone. In the second chapter I describe educational methods along with educational philosophy and the importance of teacher personality. In chapter three I briefly go over and list current literature in respect to the Trombone in Bohemia (Czech Republic). My work concentrates on the progress of musical pedagogy of education and training on the Trombone.

Práce charakterizuje regionální a strukturální politiku Evropské unie a srovnává programová období 2007-2013 a 2000-2006. Hlavní část je věnována fungování regionální a strukturální politiky EU v České republice, je provedeno zhodnocení dosavadních výsledků a zkušeností s čerpáním. Konkrétní příklady využití prostředků ze strukturálních fondů v ČR jsou uvedeny v případových studiích, které také ilustrují některé změny regionální a strukturální politiky v období 2007-2013.
