The goal of this masters thesis is focused on segmentation of the organic food market in the Czech Republic based on analysis of similarities or differences among consumers in Czech organic food market and also find out who is a typical organic food consumer. The thesis is divided into three parts, the theoretical, the methodological and the practical part. Process of the market segmentation, methods and approaches of the segmentation, the basic concepts and legislation on the matter are described in the theoretical part. Furthermore, mapping the situation of the contemporary state of the Czech organic food market. Marketing research is introduced in methodological part of the thesis. The practical part includes market segmentation by using secondary data MML - TGI (with Data Analyzer software) and primary data from quantitative research (with SPSS software). Variables were reduced to the four factors by using factor analysis. Then consumers were put into five clusters based on cluster analysis. Segments were characterized in detail using a general analysis, contingency tables, MCART analysis and multivariate statistical methods. Finally, there were elaborated appropriate marketing recommendations for the individual segments to effective marketing communication with them.

Current Transformers and Current Sensors Confrontation
Popek, Jiří ; Bok, Jaromír (referee) ; Topolánek, David (advisor)
Current transformers and current sensors are devices which are mostly used for current measurement in high voltage systems. This devices transform measured currents to range which are workable for measurement and protection devices. Current transformers have been using for current measurement since electric system was built. Current sensors are using for measurement after applying digital devices in their secondary circuits. Theoretical part includes fourth and fifth chapter. These chapters contain construction analysis and characteristic points for both devices. Practical part is in chapter six. This chapter consists results from laboratory measurement. Results are used for verification with theoretical part of the thesis. Accuracy class in nominal range and progress for measurement error in overcurrent range was verified for tested devices. Conclusion of this thesis comparing properties and summarises results from laboratory measurement.

Heterologous Expression of ABC Transporter in Catharanthus Roseus (L.) G. Don
Pomahačová, Barbora ; Dušková, Jiřina (advisor) ; Tůmová, Lenka (referee) ; Grančai, Daniel (referee)
In this thesis, the regulation of transport is investigated as a limiting factor in biosynthesis of plant secondary metabolites. CjMDR1 in Catharanthus roseus (L.) G. Don. has been selected as a model system of active transporters. CjMDR1 is a member of ABC (ATP-Binding Cassette) protein superfamily. Especially, in previous reports it was found to transport preferably berberine in its producing plant, Coptis japonica. In order to look into the role of the ABC transporter berberine was exogenously fed to Catharanthus roseus cell line and evaluate the transport of the compound. Based on a series of experiments in this study it can be concluded as follows. 1. Transport of berberine in wild Catharanthus roseus was not clearly associated with ATP-dependency. Using diverse inhibitors it was proved that berberine was not an exclusive substrate of native ABC transporters in Catharanthus. Alternatively berberine might be transported via H+-antiport or efflux transporters from MATE (multidrug and toxic compound extrusion) group which was previously found in other heterologous plants (Yazaki, 2005). In reality in Catharanthus some effect of pH on plasmatic membrane on berberine transport has been observed. However, the exact mechanism of berberine transport on vacuolar and cell level was not fully uncovered...

Analysis and modification of thin layers using ion beams
Jonner, Jakub ; Lörinčík, Jan (referee) ; Bábor, Petr (advisor)
This diploma thesis deals with analysis and modification of thin layers using ion beams. The first part of this diploma thesis deals with phenomena accompanying ion beam bombardment of solid matter. The second part of this diploma thesis is concerned with Secondary Ion Mass Spectroscopy (SIMS) and Low Energy Ion Scattering (LAIS). This work convey some basic information about these two techniques and it also deals with some benefits result in their connection into parallel depth profiling mode (such as better depth resolution of the LEIS profile, quantification of the SIMS). These benefits are demonstrated on MoSi film measurement. Within the framework of this thesis a new UHV manipulator was designed. This new UHV manipulator is equipped with precise stepper UHV motor and since the proportions are smaller, the manipulation with a sample in a space limited UHV chamber is much more comfortable and more precise. The third part of this diploma thesis deals with ion-beam induced transformation of epitaxially grown Fe films with thickness of 22 monolayer (ML) and 44 ML on Cu(100) single crystal at room temperature. Metastable Fe films of 22 ML thickness were prepared in CO pressure and 44 ML Fe films were prepared by co-evaporation of Fe with Fe64Ni36 (invar). Structural changes are analyzed by scanning tunneling microscopy and low-energy electron diffraction. The aim of this thesis is to discuss the influence of the sputtering parameters such as ion dose and ion energy on the nucleation of bcc nanocrystals, their growth, final shape and size. The influence of different Ni concentration on stability of 44 ML thick Fe films is also discussed.
